Synthese eines Carbaanalogen des S-Palmitoyl-coenzyms A, Heptadecan-2-onyldesthio-CoA und von S-Heptadecyl-CoA, zwei wirksamen Inhibitoren der Citratsynthase und der Carnitin-PalmitoyltransferaseDie Synthesen der langkettigen Alkylderivate1 und2 des Coenzyms A werden beschrieben. Heptadecan-2-onlydesthio-CoA(1), ein Carbaanaloges von S-Palmitoyl-CoA, wird zusammen mit seinem isomeren 2′-Phosphat11 durch Kondensation von Adenosin-2′,3′-cyclophosphat-5′-phosphomorpholidat(10) mit dcm in situ aus8b erzeugten Palmitoyldesthio-carba-pantethein-phosphat(8a) erhalten. Der cyclische Phosphodiester, der dabei entsteht, ergibt bei der Hydrolyse ein Gemisch von 3′- und 2′-Phosphat1 bzw.11. S-Heptadecyl-CoA(2) wird durch Alkylierung von CoA mit Heptadecyliodid hergestellt. Sowohl das Isomerengemisch (1 +11) als auch Verbindung2 zeigen Merkmale einer nicht kompetkiven Hemmung der Citratsynthase (K1 = 6.0-7.5 · 10-6 mol/l bzw. 1.0-1.5 · 10-6 mol/l) und einer kompetitiven Hemmung der Carnitin-Palmitoyltransferase (K1 = 1.35 · 10-6 mol/l bzw. 3.34 · 10-6 mol/l).

The syntheses of long chain alkyl-coenzyme A derivatives1 and2 are reported. Heptadecan-2-onyldethio-CoA(1) a carba analog of S-palmitoyl-CoA is obtained along with its 2′-phosphate isomer11 by the condensation of adenosine 2′,3′-cyclic phosphate-5′-phosphoromorpholidate(10) with the in situ generated palmitoyldethio-carbapantetheine phosphate(8a). The cyclic phosphodiester produced upon hydrolysis yields a mixture of 3′-phosphate1 and 2′-phosphate 11. S-Heptadecyl-CoA(2) is obtained from the alkylation of CoA with heptadecyl iodide. Both the isomeric mixture (1 +11) and compound2 exhibit characteristics of non-competitive inhibition with citrate synthase (K1 = 6.0-7.5 · 10-6 mol/I and 1.0-1.5 · 10-6 mol/l, respectively) and characteristics of competitive inhibition with carnitine palmitoyltransferase (K1 = 1.35 · 10-6mol/l and 3.34 · 10-6 mol/l, respectively).
