    Notes: The aim of this study was to develop rapid and accurate procedures to identify microorganisms contaminating cosmetic products, based on the identity of the nucleotide sequences of the internal transcribed spacer (ITS) region of the ribosomal RNA coding DNA (rDNA). Five types of microorganisms were isolated from the inner portion of lotion bottle caps, skin care lotions, and cleansing gels. The rDNA ITS region of microorganisms was amplified through the use of colony-direct PCR or ordinal PCR using DNA extracts as templates. The nucleotide sequences of the amplified DNA were determined and subjected to homology search of a publicly available DNA database. Thereby, we obtained DNA sequences possessing high similarity with the query sequences from the databases of all the five organisms analyzed. The traditional identification procedure requires expert skills, and a time period of approximately 1 month to identify the microorganisms. On the contrary, 3–7 days were sufficient to complete all the procedures employed in the current method, including isolation and cultivation of organisms, DNA sequencing, and the database homology search. Moreover, it was possible to develop the skills necessary to perform the molecular techniques required for the identification procedures within 1 week. Consequently, the current method is useful for rapid and accurate identification of microorganisms, contaminating cosmetics.

    Notes: Background  Dapsone (4,4′-diaminodiphenyl sulphone) is a powerful therapeutic tool in many skin diseases including neutrophilic dermatoses. The drug has an outstanding therapeutic efficacy against many skin diseases characterized by neutrophil-rich infiltrates; however, mechanisms of its action are poorly understood.Objectives  We investigated the effects of dapsone on respiratory and secretory functions of human neutrophils triggered by the chemotactic peptide N-formyl-L-methionyl-L-leucyl-L-phenylalanine (fMLP), the physiological agonist C5a, and phorbol myristate acetate (PMA).Methods  Human neutrophils were isolated from venous blood obtained from healthy donors. We detected extracellular production of superoxide (O2–) by cytochrome C reduction assay, and intracellular production of O2– by flow cytometry. Neutrophil elastase release was measured by the cleavage of the specific elastase substrate N-methoxysuccinyl-Ala-Ala-Pro-Val-p-nitroanilide. Measurement of cytosolic free calcium concentration was performed using the calcium-reactive fluorescence probe, Fluo-3.Results  Dapsone suppressed intra- and extracellular production of O2– and elastase release triggered by fMLP and C5a, but not by PMA. Both fMLP and C5a signalled the above pathways by inducing calcium influx, but PMA functions bypassed calcium influx. Dapsone was capable of antagonizing the induction of calcium influx.Conclusions  These findings suggest that one mechanism of the anti-inflammatory action of dapsone is inhibition of calcium-dependent functions of neutrophils including release of tissue-damaging oxidants and proteases in the affected skin.

    Notes: High density and isotropic poreless structure of Glass-like-carbon makes it practicallyused for fuel cells of space-shuttles and hard disk substrate etc. Glass-like-carbon is one of the newmaterials which attract attention, but it is so hard and brittle that precise machining of this materialis difficult. Surface grinding and slicing were performed to collect the fundamental data of precisemachining. The main results obtained in this study are as follows. (1) The transition from brittlemode to ductile mode in surface grinding occurs at a grain mesh size between #5000 and #3000.(2) The maximum grain depth of cut ‘dg’ necessary for ductile mode grinding is 0.021-0.006 mm orless. (3) When it is being sliced, the kerf width of the #1000 grinding wheel becomes wider thanthat of the #2000 grinding wheel because of the fact that the material removal capability of the#1000 grinding wheel is bigger than that of the #2000 grinding wheel. (4) It is extremely effectiveto use the fine grinding wheel, since the Young's modulus of glass-like-carbon is considerably lowand eventually causes compression transformation

    Notes: Summary The developmental changes in the seminal colliculus of male mice were studied by light and scanning electron microscopy. The epithelium of the mouse seminal colliculus is transformed to the mature epithelium between 20 and 25 days of age, while the transformation of the remaining urethral wall occurred from 5 to 10 days of age. During the process of this epithelial transformation at the seminal colliculus unusual structures, including prominent epithelial proliferations and protrusions, were observed in the area adjacent to the ejaculatory duct openings and the ventral area of the upper medial region of the seminal colliculus in 15-day-old mice. In these unusual structures, three types of cells were recognized by their surface characteristics: cells which had distinct borders and ruffled cell surfaces with microridges; cells which had distinct borders and flat or slightly ruffled cell surfaces with many short microvilli, and small cells which had rounded cell surfaces with many long microvilli. In the remaining area of the seminal colliculus, two types of cells were distinguished: those which had flat or slightly rounded cell surfaces with many short microvilli and those cells with many long microvilli similar to the ones in the previously described area. In 25-day-old mice, the unusual protruding structures were not observed, but some cells with ruffled surfaces were still seen.

    Notes: Summary Pregnant ICR/JCL mice were treated with 6 mg of cyproterone acetate (CA) from days 14 to 20 of pregnancy to feminize male offspring. Feminized males delivered on day 20 of pregnancy by cesarean section were castrated the same day, injected with estradiol-17β(E2) or sesame oil from the day of delivery (=day 1) to day 10 and sacrificed on day 60. In oil-injected feminized males, the vaginal epithelium was atrophic and did not show cornification. In feminized males given 20 μg E2 neonatally, the vaginal epithelium exhibited well-differentiated stratified squamous organization, but was not cornified in seven out of the nine mice of this group. In the mice treated with 50 μg E2, persistent cornification was recognized most frequently in the posterior two-thirds of the vaginal epithelium which is considered to originate from the which may contain the epithelial cells of müllerian duct was low. These results provide supporting evidence for the possible participation of epithelial cells which come from the urogenital sinus in the development of estrogen-independent persistent vaginal cornification in neonatally estrogenized mice.

    Notes: Summary Lafora-like bodies in an 8-year-old cat were studied light and electron microscopically and histochemically. In addition to Lafora-like bodies composed of branching filaments, glycogen granules and electron-dense materials, abnormal accumulations of glycogen granules attracted attention. The most remarkable features were the developmental processes of the branching filaments originating directly from glycogen granules. Lafora-like bodies in the present study showed ultrastructural, histochemical, and enzymatic similarities to those described in the previous reports in Lafora's disease, glycogenosis and other cases. From these results, a certain disturbance of the glycogen metabolism is considered to be probably related to the productive mechanism of Lafora-like bodies.
