    Notizen: Abstract— Neurofilaments and neurotubules are the principal fibers of the mature normal neuron. In this study the protein subunits of these neurofibrils were isolated from human autopsy tissue, and compared by sodium dodecyl sulfate-polyacrylamide gel electrophoresis, and by two dimensional peptide maps of the tryptic digest of these proteins labelled with 125I. The α and the β monomers of neurotubule are related but distinct in their peptide maps, while the major neurofilament protein subunit (molecular weight, 50,000) is remarkably similar to β tubulin. The neurofilament fraction binds colchicine, but the specificity is not yet determined. Neurotubule and neurofilament are also similar in having minor proteins which coelectrophorese on the gels. These results suggest that neurofilament and neurotubule may share one or more protein subunits.

    Notizen: In the rat phenylacetate readily penetrates into such tissues as brain, eyes, heart, kidneys, and liver. One hour after the subcutaneous injection of sodium phenylacetate into rats ranging in age from 1 to 65 days, the ratios of the concentrations (μmolig) in blood/tissue were approx 1:1 in all of these tissues with the exception of the brain. The blood-brain ratio changed from 1:1 in the 1–7 day old rat to the adult ratio of 2:1 at 21 days of age. The free access of phenylacetate into the immature brain is especially relevant to phenylketonuria, a metabolic disease in which excessive amounts of phenylacetate are produced in peripheral tissues.

    Notizen: Phenylacetate is not excreted to any significant extent as the free acid in rat urine, but must be metabolized in the liver and kidney, first to phenylacetyl-CoA, then to phenylacetylglycine. One hour after [14C]phenylacetate loading, the radioactivity in the liver and kidneys of the young rat could all be accounted for as unchanged phenylacetate (50-5573, phenylacetylglycine (35–40%), and phenylacetyl-CoA (5–8%). In the brain, the radioactivity was present mainly as phenylacetate (82–90%); only 10–18% was found as phenylacetyl-CoA. The formation of phenylacetyl-CoA appeared to be the rate limiting step in the clearance of phenylacetate.In the urine at least 95% of the radioactivity was present as phenylacetylglycine, less than 1% as phenylacetate, and 3–4% as phenylacetyl-CoA. The concentration of phenylacetylglycine in the urine was therefore used as a measure of the in vivo rate of phenylacetatc clearance. This detoxication process was found to develop postnatally. The formation of phenylacetylglycine was barely detecrabie in the newborn rat and remained relatively slow for about 2 weeks. During the third week a large increase in enzymatic activity, approx 40% occurred. Adult level of activity was reached in the 40 day old rat.The extremely slow rate of detoxication in the newborn animal was reflected in the persistence of high concentrations of phenylacetate in the tissues. The relevance of our findings to human phenyl-ketonuria is discussed

    Notizen: Abstract— Oligodendroglia isolated from frozen human brain induced EAE in the guinea pig when injected with Freund's complete adjuvant. An acid extract of the isolated cells was highly encephalitogenic. The disease was clinically and histologically similar to EAE induced with CNS white matter or myelin basic protein(MBP). The isolated cells were essentially myelin-free but contained a large amount of myelin basic protein as determined by polyacrylamide gel electrophoresis. It was shown that the cells can absorb 125I-MBP during isolation and the possibility that contamination from extra cellular MBP is responsible for the encephalitogenic activity is considered.

    Notizen: Summary Experimental allergic encephalomyelitis was induced by sensitization with guinea pig spinal cord in complete Freund's adjuvant with increased content of myobacterium in Hartley guinea pigs between the first and 28th day after birth. The animals either died during acute EAE or showed chronic progressive, chronic relapsing or chronic EAE with delayed onset. In animals immunized between the 1 and 13 day after birth, predominatly ordinary and chronic progressive EAE was noted, whereas animals sensitized between the 14th and 23rd day frequently suffered from hyperacute or chronic relapsing EAE. Immunization on the 24th day led to very high mortality in acute EAE. The relation of the pathohistologic alterations to different types of human inflammatory demyelinative disease is discussed.

    Notizen: Summary The inflammatory reaction in chronic relapsing EAE was studied with special reference to the alterations found in the early stages of the disease (first attack, first remission, and first relapse). The inflammatory reaction during the early stages of the first attack was dominated by polymorphonuclear leukocytes, together with edema and sometimes hemorrhage, resembling the alterations found in hyperacute or neutrophilic EAE. The tissue reaction consisted of myelin, as well as axonal damage, and involved predominately the root entry zones and the subpial white matter in the lower thoracic and lumbo-sacral spinal cord, with less severe damage in grey and white matter in other regions of the brain. During the first remission, a variable amount of perivascular inflammatory cuffs of lymphocytes and mononuclear cells were found, but cellular infiltration into the neural parenchyma was confined to a few small perivascular areas. In the first relapse, a massive invasion of the neural parenchyma by mononuclear cells was noted, leading to large plaque-like zones of demyelination in the white matter. In this stage of the disease axonal damage was rare. The lesions were predominantly confined to the lower thoracic and lumbo-sacral spinal cord, but with increasing duration of the disease, higher portions of the spinal cord, as well as brain stem, cerebellar white matter, centrum semiovale and fornix were involved, too. Further relapses showed a similar pattern as described in the second attack, although in these animals lesions of different ages were always found. Parallel with the clinical observations, the inflammatory reaction in the central nervous system also diminished with the duration of the disease. A comparison between Strain 13 and Hartley guinea pigs revealed a more severe reaction during the first attack in the latter, possibly responsible for the high mortality of these animals during this stage of the disease.

    Notizen: Summary A recurrent type of primary demyelination was found in the anterior roots and dorsal root ganglia of strain 13 guinea-pigs with relapsing experimental allergic encephalomyelitis. Single nerve fibre studies revealed a predominant nodal—paranodal distribution of the destruction of myelin in these nerves. Damage of the Schwann cell—axolemmal junction was found in the majority of the abnormal nodes of Ranvier. This was accompanied by the formation of supernumerary adaxonal Schwann cell processes which further altered the normal myelinating cell—axon relationship. It is concluded that variations in antigenic composition may play a role in the selective involvement of the anterior roots. The pathogenesis of the observed nodal changes is discussed.
