    Notes: We present new measurements of inelastic He atom scattering from the surface phonons of Cu(001) as a function of crystal temperature, incident energy, and parallel momentum transfer. A careful subtraction of the multiphonon intensity and other background contributions from the time-of-flight intensities reveals three distinct surface-localized vibrational modes which are ascribed to the Rayleigh phonon, the longitudinal bulk resonance, and a further acoustic bulk resonance at higher energy transfers. The longitudinal resonance couples very strongly to the scattering He atoms and, for a wide range of incident conditions, gives peaks which are more intense than those due to the Rayleigh mode. The energy and momentum dependence of these peak intensities are analyzed with the aid of a simple distorted wave Born approximation, and the different coupling parameters for the two modes are determined and compared with other available data. The incoherent diffuse elastic peak is shown to decrease as a function of parallel momentum transfer according to the theory of Fraunhofer scattering from a random array of point defects. The multiphonon background is shown to be in agreement with a quick scattering approximation. © 1994 American Institute of Physics.

    Notes: Abstract The time evolution of the electron density has been measured in the outer regions of a fast theta pinch by means of a Mach-Zehnder interferometer. The results show that the electron density outside the central plasma column is relatively high, i.e. between 1 and 5×1014 cm−3. A comparison of these measurements with theoretical calculations leads to the conclusion that the pinch must be unstable during the implosion phase.

    Description / Table of Contents: Zusammenfassung Elektronentemperatur und Elektronendichte wurden im ”Rotating magnetic field pinch”—Experiment mit Hilfe von Laser-Licht Streuung gemessen. Die Resultate werden mit früheren Messungen und mit numerischen Berechnungen verglichen. Für die numerischen Berechnungen wurde ein magnetohydrodynamisches Dreiflüssigkeitsmodell verwendet. Der Vergleich zwischen Theorie und Experiment zeigt, dass die Wände des Entladungsrohres eine Plasmaquelle darstellen, die etwa 0.6*1015 Elektronen und ebensoviele Ionen pro cm2 pro μs erzeugt.
    Notes: Summary Electron temperature and density were measured in the rotating magnetic field pinch by means of laser light scattering. The results are compared with previous measurements and with numerical calculations based on a three-fluid magnetohydrodynamic model. The comparison between theory and experiment shows that the interior surface of the discharge tube acts as a plasma source producing approximately 0.6*1015 electron-ion pairs per cm2 per μs.

    Notes: Abstract Cyclic GMP-dependent protein kinase (cGMP kinase) is involved in the relaxation of smooth muscle. The enzyme has been cloned and expressed in eukaryotic cell lines but so far not in prokaryotic cells. Three vectors were constructed for the expression of Iα cGMP kinase inEscherichia coli. Transformation with the pET3a/cgk vector which uses the T7 RNA polymerase/promotor system resulted in efficient accumulation of cGMP kinase. Most of the protein was in an insoluble and catalytic inactive form. Various solubilization and refolding conditions did not yield an active enzyme. A small fraction of the cGMP kinase was present in the soluble cell extract. This fraction bound cGMP with high affinity but had no cGMP stimulated kinase activity. To prevent aggregation two additional vectors were constructed. (I) A bacterial leader sequence, which directs the export of proteins into the periplasmic space, was fused to the aminoterminus of the cGMP kinase. (II) A gram/gram+ shuttle vector for expression under the control of the tac promotor was used. Both constructs directed the synthesis of an isoluble and inactive cGMP kinase. These results suggest that large amounts of cGMP kinase can be expressed inE. coli, but mainly in an isoluble and inactive form. In contrast to eukaryotic cells, bacteria may lack systems for correct protein folding and/or posttranslational modification that are crucial for the productive folding and/or activation of cGMP kinase.
