    Notes: A method combining the techniques of laser blowoff and laser-induced fluorescence has been developed in order to measure the local electron density below 1013 cm−3. Characteristics of a Li0 beam produced by laser blowoff of a thin Li film are investigated using beam–plasma interactions and laser-induced fluorescence. Such a beam has a near-Maxwellian velocity distribution with a temperature around 4.5 eV and a density of the order of 1010 cm−3 at a distance of about 1 m from the film target. The feasibility of measuring electron density with this Li0 beam and a dye laser is demonstrated with an ECR plasma.

    Notes: To measure local electric field in an rf-plugged sheet plasma in the RFC-XX-M open-ended machine, a spectroscopic system using a combined technique of Li-beam probing (laser blow-off method) and laser-induced fluorescence has been developed. A Li atom in the rf field can be populated to 42F by a stepwise laser pumping (22S→22P→42F) because of the Stark mixing between 42F and 42D. A cascade fluorescence of 610.4 nm (32D→22P) subsequent to the direct transition (42F→32D) was observed to estimate the rf field strength (rms) Erms. To correct the effect of electron collisions in the plasma, we observed a blue fluorescence consisting of two lines: a forbidden line of 460.2 nm (42F→22P) and an allowed line of 460.3 nm (42D→22P) induced by electron collisional transfer (42F→42D). From these intensities, we obtained Erms and electron density ne by solving numerically the rate equations, including the relevant radiative and collisional processes under the experimental conditions. Erms and ne were estimated to be 2.4 kV/cm and 3.1×1011 cm−3 near the center of the sheet plasma, respectively. It was found that the obtained Erms was enhanced by about 2.5 times with respect to the vacuum field (1 kV/cm). The obtained ne agreed well with ones by other methods. A spatial distribution of ne was also obtained from that of the Li0 resonance line (22P→22S) due to electron impact excitation. The present spectroscopic system is applicable to plasmas of ne(approximately-less-than)1013 cm−3 and static or quasi-static electric fields of E(approximately-greater-than)400 V/cm.

    Notes: Summary We describe here a further Japanese family with pyrimidine 5′-nucleotidase (P5′N) deficiency diagnosed using a nuclear magnetic resonance (NMR) spectrum, in Kumamoto prefecture where two families having the disease have been reported before. The specific spectra in1H-NMR of P5′N deficient erythrocytes were due to three methyl protons of CDP-choline at 3.22 ppm and to H-2, H-8 and ribose-1′ of pyrimidine nucleotide phosphate(s) in the lower fields (at 5.82 and 8.00 ppm). The other specificities in31P-NMR spectra were due to CDP-choline, CDP-ethanolamine and UDP-glucose. Those spectra were not detected in other types of hemolytic anemia.

    Notes: Summary The light- and electron-microscopic localization of serine:pyruvate aminotransferase (SPT) in rat kidney was studied using immunoenzyme and protein A-gold techniques. Rat kidneys were fixed by perfusion through the abdominal aorta and small tissue slices were embedded in Epon, Lowicryl K4M, or LR Gold. The Epon was removed from the semithin sections, which were then stained using the immunoenzyme technique. Ultrathin sections of Lowicryl K4M- or LR gold-embedded materials were labeled using the protein A-gold technique. At light microscopy, discrete granular reaction deposits were exclusively present in the proximal tubule, all of whose segments were positive for SPT. A weakly positive reaction was observed in the distal tubules. At electron microscopy, gold particles indicating the antigenic sites for SPT were confined to the peroxisomes and mitochondria. The labeling intensity of both organelles was dependent on the embedding resins used. The labeling of Lowicryl K4M-embedded material was weaker than that of LR gold-embedded material; Quantitative analysis confirmed this result. Our results indicate that, in rat kidney, the main intracellular sites for SPT are peroxisomes and mitochondria of the proximal tubule.

    Notes: Summary The localization of serine:pyruvate aminotransferase (SPT) in human liver was investigated by indirect immunoenzyme and protein A-gold techniques. By light microscopy, diaminobenzidine reaction product was present in cytoplasmic granules of the parenchymal cells. By electron microscopy, gold particles indicating the antigenic sites for SPT were exclusively confined to peroxisomes but not to mitochondria. By double labeling technique, both peroxisomal marker enzyme, catalase and SPT were detected in the same peroxisomes. Quantitative analysis of the labeling density showed that SPT is contained only in peroxisomes. The results indicate that in human liver most of SPT is contained in the peroxisomes.

    Notes: Summary Replicating circular molecules of SV 40 DNA were isolated from SV 40 infected VERO cells. Preparations from dilute and undiluted passaged SV 40 virus stocks were shown to give different frequency distributions of contour length of replicating DNA molecules. The mean length of RF-DNA was 1.26±0.21μ and 1.57±0.18μ for the undiluted and dilute passaged SV 40 stocks, respectively. In undiluted passaged virus stocks, small replicating molecules with a contour length less than 1.0μ were noted, which were not detected in dilute passage stocks. The presence of shortened replicating forms of DNA in the undiluted passaged virus stocks would indicate that the DNA of defective SV 40 virus can replicate.

    Notes: The change in x-ray diffraction intensity with time following rapid stretching by various amounts is measured for a low-density polyethylene sample at several temperatures. An appreciable decrease in intensity with time at the meridian of the diffraction from the 110 and 200 planes is observed to occur within the first 5 sec following stretching. The change takes considerably longer times at lower temperatures. By use of a calibration curve, the change in crystal orientation functions for the a, b, and c crystal axes were calculated, and an increase in c-axis orientation was shown. From the results of birerringence measurements on the same sample, the crystalline and amorphous contributions to birefringence were calculated. It was shown that stretching is initially accompanied by a greater amount of amorphous than crystalline orientation, and that relaxation involves an increase in crystalline orientation accompanied by a decrease in amorphous orientation.
