    Notes: SUMMARY: The effect of psychrotolerant bacteria on the water-holding capacity as measured by the extract release volume (ERV), pH and protein degradation in chicken skin stored at 5°C was determined using strains of Achromobacter and pigmented and nonpigmented Pseudomonas. All organisms caused a rapid decrease in ERV to about 50% of the original value during the early log phase of growth before development of off-odor. Changes in pH and content of extractable nitrogenous materials occurred during bacterial growth but were not pronounced until after the count was greater than 108 cells/g of skin and a faint off-odor was detectable. The content of extractable materials decreased during the early log phase, corresponding to the period of rapid increase in pH; later, during the late log or stationary phases of growth, it rapidly or gradually increased, depending on the organism.

    Notes: Changes induced by type-cultures of Achromobacter, nonpigmented Pseudomonas, pigmented Pseudomonas and a mixture of all three types in the amino acid content of fresh chicken skin were studied during storage at PC for up to 21 days. The Achromobacter and nonpigmented Pseudomonas cultures reduced the amount of all amino acids to below detectable levels during the early log phase of growth and produced no detectable change subsequently. The pigmented Pseudomonas caused no appreciable change initially but produced a marked increase in the amount of most amino acids during the late log phase, after off-odor had developed; an increase in free and total extractable proline and hydroxyproline indicated that collagenous proteins were attacked. The mixed inoculum gave intermediate results, producing comparatively little change in the total free amino acid content throughout the incubation period. Results indicate that the pigmented pseudomonads are the most proteolytic of the common types of psychrotolerant spoilage bacteria and that they possess collagenase.

    Notes: The addition of as little as 2 ppb of manganese to ferrocyanide-treated beet molasses during citric acid fermentation by Aspergillus niger NRC A-1-233 caused a 10% reduction in acid yield and undesirable change in the morphology of the organism from the normal pelletlike form to the filamentous from. Still smaller additions (0.4-2ppb) caused undesirable pellet clumping, while greater additions (2-100 ppb) gave further decreases in yield. The yield obtained at 100 ppb was less than 25% of that obtained at 1 ppb or less. None of the other metals tested (Al3+, Ca2+, Co2+, Cu2+, Fe2+, Mg2+, Ni2+, Zn2+) visibly changed pellet morphology, and only Al3+, Fe2+, and Zn2+ at relatively higher concentrations (5-25ppm) reduced acid yield. The adverse effect of manganese on growth and acid production was not affected by addition of the other metals.

    Notes: The addition of potassium ferrocyanide to the molasses substrate (mash) used in the citric acid fermentation had little or no effect on the total carbon, nitrogen, or phosphorus content of the substrate but reduced the ash content by 1-4% depending on the type and crop year of molasses. Eighteen of the 21 metals identified in beet molasses by spectrographic analysis were precipitated in part by the treatment. The metals known to interfere with citric acid production, particularly manganese and iron, were those most efficiently precipitated. At mash temperatures of 80°C. and approximately 100°C., precipitation was completed within 15 min. after ferrocyanide addition and the reductions in the soluble mineral content in each case were similar. At 25°C. the precipitation reactions occurred but required more than 50 hr. to complete. Significantly more total mineral and more iron, calcium, and copper were precipitated at pHs 4 and 6 than at pH 8; manganese precipitation, however, was not affected by these pH changes.
