    Notes: An algorithm is given which is suitable for solving molecular structures when the shape of the whole molecular skeleton, or at least the main part of it, can be assumed. The molecular parameters, i.e. three rotations and three translations in the general case, are randomly generated by a Monte-–Carlo method. The packing thus obtained is group-refined using only a few selected strong reflexions, then kept or rejected according to several tests on the weighted reliability index; the best structures are furtherly group-refined using an increasing number of reflexions up to a given limit. The whole program has been arranged to be as fast as possible and fully automatic. Such a method is helpful when the experimental data are poor or restricted to some extent (high-pressure single-crystal data, unstable crystals .... ) or in cases of static or dynamic disorder (plastic crystals). Even when accurate three-dimensional data are available, it may be of value and possibly faster than other methods, if one or several molecular parameters are a priori fixed from symmetry considerations or one atom located (heavy atom). Several typical examples of structures which have been solved or redetermined are given.

    Notes: Respiration as an index of oxidative energy production was investigated in a L-cell suspension culture system previously shown to exhibit density-dependent inhibition of growth. It was found that as cultures progressed from exponential growth to high density nongrowing populations (6-10 × 106 cells/ml) over a 2-week period, the respiratory rate determined from the total amount of oxygen consumed during the daily medium renewal cycle, declined from 5.4 to 1.8 fmoles O2/cell/min. There are two components in this decrement. The first consists of a daily recurrent decline of oxygen uptake resulting from decreased availability of medium oxygen and glutamine and is readily reversed by medium supplementation. The second component which is refractory to medium supplementation and accounts for approximately 50% of the total respiratory decline, is considered to indicate an adaptive change of the respiratory capacity of the cells. This change is reversed during the lag period which precedes resumption of exponential growth upon subculture to low cell densities. The significance of these results is discussed in relation to recent reports indicating a marked depression of respiratory activity in nongrowing dense attached cultures as well.

    Notes: High density L cell suspension cultures were previously shown to remain viable for indefinite periods of time and to exhibit marked inhibition of DNA synthesis and mitosis while the fraction of total protein synthesis represented by collagen is increased. The present study demonstrates that regulation in this system extends to the activity of acetylcholinesterase found to be approximately 100-fold greater in the high density populations than in low density exponentially growing cultures. Kinetic studies of the increase of the activity, its fluctuation over an extended period of time and its decrease upon resumption of exponential growth after dilution of the cultures were performed. The data obtained indicate that the enzyme does not accumulate in high density populations merely as a result of the absence of net protein synthesis and cell division but that changes of its rates of synthesis and possibly degradation are involved. The expression of regulated acetylcholinesterase activity in a cell line of connective tissue origin is considered in relation to phenotype reprogramming and to cell membrane associated growth control mechanisms.

    Notes: Suspension cultures of L-929 fibroblasts grown to densities of 6 to 10 × 106 cells/ml through daily centrifugation and resuspension in fresh media, have been maintained for periods up to five months without change in viability or cell size. DNA synthesis and mitosis in these cultures is limited to 5% of the cells per day, a fraction very nearly equal to the fraction of cells rendered nonviable, most likely during the manipulations associated with medium renewal. The kinetics of the flow of cells into the S and M periods following (a) renewal of the medium and (b) dilution of the high density cultures, suggest that the large majority of the cells are in a G0 or early G1 phase, resuming growth readily in response to decreased cell density. This is further indicated by the sequence of the marked shifts occurring in the cell volume distribution spectrum of the high density cultures after dilution. Long term, steady state regulation of growth with retention of intact viability was thus demonstrated in the case of a long established aneuploid cell line. The fact that this occurs in suspension but not in attached cultures, supports the concept that impairment of growth control in such cells affects predominantly regulatory mechanisms located at the cell surface rather than those concerned with intracellular synthesis and metabolism.
