    Description / Table of Contents: Zusammenfassung Die Aufnahme von Substraten in die Zelle erfolgt oft über zwei oder mehr Transportsysteme. Die Charakterisierung einer einzelnen Komponente solcher multipler Transportsysteme erfordert die Aufstellung gewisser Kriterien. Einige experimentelle Daten wurden herangezogen, um die Notwendigkeit solcher Kriterien zu zeigen. Ideale Konstellationen derK- undV- Werte der verschiedenen Transportsysteme, die eine Trennung der verschiedenen Komponenten durch graphische Analyse ermöglichen, werden diskutiert. Es werden drei graphische Auftragungen erörtert. Die Diskussion beruht auf den Verhältnissen der Steigungen und Achsenabschnitte der zwei extremen Tangenten der Kurven in diesen Auftragungsarten.
    Notes: Summary The uptake of substrates often involves two or more transport systems. The characterization of single components of multiple transport systems requires setting up certain criteria for their determination. Some experimental data were used to demonstrate the necessity of such criteria. Ideal constellations ofK andV values of the different transport systems for a single substrate to separate the various components by graphical analysis are discussed. The discussion is based on the slopes and the intercepts of the two extreme tangent equations.

    Description / Table of Contents: Zusammenfassung Es wurden Untersuchungen mit der direkten Immunfluorescenzmethode bei Kranken mit rezidivierenden herpetischen, aphthösen und anderen Mundschleimhautkrankheiten mit Antiherpes- und Antiadenoviruskonjugaten durchgeführt. Sowohl bei den Patienten wie bei den Personen mit intakter Mundschleimhaut wurde eine spezifische Fluorescenz beobachtet. Erhöhte Adenopositivität wurde bei Patienten mit rezidivierenden, besonders mit ständig rezidivierenden Aphthen, erhöhte Herpespositivität bei Kranken mit Erythema multiforme beobachtet. Die mögliche pathologische Rolle der virusantigen-haltenden Lymphocyten wurde angenommen.
    Notes: Summary Circulating lymphocytes of patients with recurrent herpetic, aphthous and other oral mucosal diseases, as well as of persons with intact oral mucosa were examined with direct immunofluorescence method using HVH and adenovirus hyperimmune conjugated sera. Some of the circulating lymphocytes showed a specific fluorescence both in patients and in persons with intact oral mucosa. Increased herpes-positivity was observed only in erythema exsudativum multiforme and increased adeno-positivity in patients with recurrent aphthous stomatitis, especially in cases with constantly recurring aphthae. The possible pathologic role of the circulating virus-antigen-carrier lymphocytes is presumed.

    Notes: Alcohols inhibit the exchange transport of glucose in human erythrocytes. Comparing the inhibition by monohydroxy-alcohols, which have different distribution coefficients between medium and membrane, shows that the degree of inhibition depends mainly upon the concentration of the alcohol in the membrane.1-butanol exerts a mixed-type inhibition; Vmax decreases and Km increases. Since also the Km of the equilibrium transport increases upon the addition of the alcohol, the changes in the Km of exchange transport cannot be attributed solely to the differently affected mobilities of the loaded and free carrier, but the affinity of glucose to the transport system is reduced.The transport system can bind two alcohol molecules. With one alcohol molecule bound the affinity of the transport system for the second alcohol molecule already increases.The nature of the bond of the alcohols to the transport system is discussed and possible explanations for the cooperative effect upon the binding of the second alcohol molecule are offered.

    Notes: In glucose exchange transport into red blood cells the rate of glucose uptake showed two pH dependent maxima, with the larger at approximately pH 7.5 and the smaller one at pH 3.In the studied pH range the relation between the rate of glucose uptake and the substrate concentration followed Michaelis-Menten kinetics. While the maximal velocity (V) reflected the pH changes of the media, the Michaelis constant (Km) remained constant.The dissociation constants of the groups of the free carrier and the carrier-glucose complex were the same. The pK of the acidic group was 5.2 and of the basic group 9.5.Glucose was not bound to groups of the carrier which dissociated protons in the pH range of three to nine.By rearranging the equation for the pH dependence of the relative influx a more definitive graphic determination of the pK values was produced.

    Notes: The temperature dependence of the uptake of glucose by exchange transport is investigated at two pH values (7.5 and 5). The Arrhenius's energy of activation, the heat of activation, the free energy of activation and the entropy of activation are calculated. The parameters are different at pH 7.5 and 5. For both pH levels the heat of activation and the entropy of activation change abruptly at 20°C. This finding leads us to assume that at this temperature a change in the structure of the membrane takes place.

    Notes: Wild-type cultured cells of the frog cell line ICR 2A give rise to 5-bromodeoxyridine (BUdR)-resistant colonies only when the selecting concentration of the drug is 5 × 10-5 M or lower. The progeny of these colonies multiply in 10-4 M BUdR; resistance is correlated with the absence of a thymidine (TdR)-specific transport reaction with a Km in the range of 2-7 × 10-4 M. All of the TdR transport-deficient (TT-) isolates examined (25) had TdR kinase activity (4% to 100% of wild-type). Variants deficient in TdR kinase activity (5% of wild-type) were obtained by exposing TT-cultures to 10-3 M BUdR. The TK - variants multply continuously in 10-3 M BUdR and retain the phenotype after prolonged culture in the absence of the drug. The frequency with which they occur is increased 20 to 50 fold by prior treatment of the culture with ICR 191, an acridine mustard mutagen.In haploid cells, it would be expected that TK- variants would arise in equal numbers from wild-type and TT- cultures if loss TdR kinase occurred independently of loss of the transport reaction. However, wild-type cells give no colonies resistant to 10-3 M BUdR under conditions the give 1 to 50 colonies per million TT- cells. The TT- phenotype seems to be a required intermediate state in the origin of the TK- phenotype. Therefore, the TK- clones described above are unlikely to be products of mutation at a single genetic locus.
