    Notizen: There has been substantial interest in mindfulness as an approach to reduce cognitive vulnerability to stress and emotional distress in recent years. However, thus far mindfulness has not been defined operationally. This paper describes the results of recent meetings held to establish a consensus on mindfulness and to develop conjointly a testable operational definition. We propose a two-component model of mindfulness and specify each component in terms of specific behaviors, experiential manifestations, and implicated psychological processes. We then address issues regarding temporal stability and situational specificity and speculate on the conceptual and operational distinctiveness of mindfulness. We conclude this paper by discussing implications for instrument development and briefly describing our own approach to measurement.

    Notizen: The 10 GHz ECR-I ion source at ATLAS is being modified to create a small inner volume within the main plasma chamber to reduce the residual helium background gas load. This effort is part of an experiment to measure the residual concentration of 3He in highly isotopically enriched 4He samples. The modification consists of a new extractor electrode on which is integrally mounted a 2.5-cm-diam quartz tube of approximately 8 cm length. Energizing just the extraction coil produces an ECR only inside the quartz volume. The quartz tube is pumped only through the 1-mm-diam extraction hole. rf power is provided by propagation through the main plasma tank acting as a multimode waveguide. The source must operate in the pressure regime of 10–200 mTorr in order to achieve sufficient sensitivity to measure a 3He/4He ratio of approximately 1×10−15. Results of these tests and the operation of the source in this highly unusual mode will be presented. © 2002 American Institute of Physics.

    Notizen: Two techniques for the discrete injection of material into an Electron Cyclotron Resonance ion source (ECRIS) have been developed for the purpose of measuring the ionization and confinement times of ion species. Previously only solid materials in conjunction with a pulsed laser were used in these studies due to the discrete material introduction produced by this configuration. The first method replaces the pulsed laser with a fast high voltage pulse applied to a sputter sample. The high voltage pulse has a rise time of 100 ns, fall time of 80.0 μs, and variable pulse duration. The second method utilizes a fast-pulsed gas valve capable of producing a gas pulse 160 μs in width. These pulse widths are well below the ionization times of the lower charge states and thus allows for time measurements to be made of all charge states. Both of these techniques can be employed to study the effects of rf power, coil configuration, biased disk, and gas mixing on ionization and confinement times. Rise times for neon, argon, and gold will be presented. © 2002 American Institute of Physics.

    Notizen: Rotationally resolved S1←S0 fluorescence excitation spectra of 2-chloronaphthalene (2ClN) are measured using a newly constructed ultraviolet (UV) laser/molecular beam spectrometer. More than 1000 well-resolved rotational lines are recorded at a resolution of ∼3 MHz for the two overlapping band origins of the 35Cl and 37Cl isotopomers (Δνorigin∼3 GHz) over a 3.5 cm−1 spectral region at a rotational temperature of 10 K. Both spectra are hybrid bands, having 18% a-/82% b-type character in accordance with 1La←1A1 excitations observed for other 2-substituted naphthalenes. Additionally, Cl nuclear quadrupole splittings are observed as broadened asymmetric line shapes with linewidths from 5 to 20 MHz. Least-squares fits of these line shapes coupled with high-precision Fourier-transform microwave measurements of the ground state have provided accurate S1 quadrupole coupling constants. Specified relative to the principal quadrupole tensor orientation of S0, the S0 and S1 values of eQqzz are −71.198(5) MHz and −67.8(30) MHz, and the asymmetry parameters, η=(eQqyy−eQqxx)/eQqzz, are 0.076 38(5) and 0.13(5), respectively. The experimental line shapes are fit to Voigt functions, yielding 4.9(3) MHz homogeneous (lifetime) and 3.0(2) MHz inhomogeneous (Doppler) contributions. All of the observed structure are well reproduced by more than 20 000 transitions predicted for the nuclear-quadrupole-split rotational spectra of the two hybrid-band origins. Furthermore, the homogeneous linewidths show no dependence on either the upper state energy or angular momentum quantum numbers. The absence of additional level structure at the resolution of 1 MHz or more, from heavy-atom-enhanced intersystem crossing mechanisms, is discussed within the framework of radiationless transition theory. The quadrupole tensor components are used in simple theoretical models to investigate the orbital character of the S1 state and its radiationless decay mechanisms. © 2001 American Institute of Physics.

    Notizen: Past reports suggest that reduced P300 amplitude is associated with risk for alcoholism. We examined whether visual P300 amplitude could identify familial risk for alcohol disorders in individuals not known to be at risk at the time P300 was recorded. These individuals were twins from pairs where neither twin had an alcohol disorder at age 17 but familial risk was established at age 20 when one twin developed an alcohol disorder whereas the other did not. Of special interest was the P300 of the unaffected twin recorded at age 17 when both twins were alcoholism free. We found reduced P300 in the unaffected twin compared to pairs where both members were continuously disorder free. Hence, P300 was reduced in alcohol disorder-free individuals whose twin siblings subsequently developed alcoholism, further supporting reduced P300 amplitude as an endophenotype indexing familial risk for alcoholism.

    Notizen: . For the dinoflagellates, an important group of single-cell protists, some nuclear rDNA phylogenetic studies have reported the discovery of rDNA pseudogenes. However, it is unknown if these aberrant molecules are confined to free-living taxa or occur in other members of the group. We have cultured a strain of symbiotic dinoflagellate, belonging to the genus Symbiodinium, which produces three distinct amplicons following PCR for nuclear small subunit (18S) rDNA genes. These amplicons contribute to a unique restriction fragment length polymorphism pattern diagnostic for this particular strain. Sequence analyses revealed that the largest amplicon was the expected region of 18S-rDNA, while the two smaller amplicons are Symbiodinium nuclear 18S-rDNA genes that contain single long tracts of nucleotide deletions. Reverse transcription (RT)-PCR experiments did not detect RNA transcripts of these latter genes, suggesting that these molecules represent the first report of nuclear 18S-rDNA pseudogenes from the genome of Symbiodinium. As in the free-living dinoflagellates, nuclear rDNA pseudogenes are effective indicators of unique Symbiodinium strains. Furthermore, the evolutionary pattern of dinoflagellate nuclear rDNA pseudogenes appears to be unique among organisms studied to date, and future studies of these unusual molecules will provide insight on the cellular biology and genomic evolution of these protists.

    Notizen: The low-temperature cation-ordered superstructure of anhydrous sodium sodalite, a zeolite with composition Na6[AlSiO4]6, has been determined through the use of both density functional theory (DFT) and classical force-field lattice energy minimizations. The charge-balancing Na+ cations are assumed to occupy their characteristic locations within the cubic alumino–silicate framework near the centers of the 6-ring windows. Within the constraints of the volume-doubled pseudotetragonal supercell reported in a previous x-ray diffraction study [B. Campbell, S. R. Shannon, H. Metiu, and N. P. Blake (submitted)], all possible arrangements of cations and vacancies amongst the 6-ring window sites were considered. Force-field calculations employing the ab initio based potential energy function derived by Blake, Weakliem, and Metiu [J. Phys. Chem. B 102, 67 (1998)] and the empirical shell-model potential of Catlow et al. [J. Chem. Soc. Commun. 1984, 1271; Mol. Simul. 1, 207 (1988)], were used to perform full lattice-energy minimizations of each configuration, and to assess their relative stabilities both before and after minimization. The most stable configurations were then examined in more detail via ab initio density functional calculations in the generalized gradient approximation. The lowest-energy supercell ordering proved more stable than the lowest-energy parent cell ordering, and also yielded a pseudotetragonal distortion (space group Pnc2) and a calculated diffraction pattern that qualitatively match experimental results. The structural influences that contribute to the low energy of the correct vacancy ordering are described in detail. © 2000 American Institute of Physics.

    Notizen: For individual varieties, tolerance of septoria leaf blotch was quantified by the slope of the relationship between disease and yield. Variation in disease severity and the associated yield responses were provided across two sites and three seasons of field experiments. Slopes were fitted by residual maximum likelihood for two contrasting models: (i) a fixed-effects model, where no prior assumptions were made about the form of the variety slopes; and (ii) a random-effects model, where deviations in individual variety slopes away from the mean variety slope formed a normal random population with unknown variance. The analyses gave broadly similar results, but with some significant differences. The random model was considered more reliable for predicting variety performance. The effects of disease were quantified as symptom area and green canopy duration. Models of the relationship between symptom area and yield were site-specific. When site effects were not taken into account, these models had poor predictive precision. Models based on the canopy green area gave robust predictions of yield and were not site-specific. Differences in disease tolerance were detected in a comparison of 25 commercial winter wheat varieties. Tolerance was not detected directly through symptom measurements, but instead through measurements of canopy green area, which provides a measurement of the effects of disease that accounts for differences in canopy size across sites and seasons. The varieties showing greatest tolerance tended to have lower attainable yield than the intolerant varieties. Presence of the 1BL/1RS chromosome translocation, which has been reported to increase radiation use efficiency, appeared to be associated with intolerance.

    Notizen: Aim : To assess the pharmacokinetics of alosetron, its effect on in vivo enzyme activities, and influence of demographic factors during repeated dosing.Methods : Thirty healthy men and women received 1 mg oral alosetron twice-daily for 29.5 days and a single oral dose of a metabolic probe cocktail before and on the last day of alosetron dosing. Serum alosetron concentrations were measured on days 1, 8, 15, 22 and 29. Probe-substrate and metabolite concentrations were measured after each cocktail dose.Results : Alosetron accumulation in serum was negligible. Exposure to alosetron did not alter probe-metabolite/substrate ratios associated with CYP2C19, 2E1, 2C9, or 3A4 activity, but modestly decreased those associated with CYP1A2 and N-acetyltransferase activity. Systemic exposure to alosetron was higher in women, positively correlated with age and body mass index, and negatively correlated with CYP1A2 activity. Incidence of constipation was higher in women, but not associated with alosetron concentration.Conclusions : Single dose data can reliably predict the pharmacokinetics of alosetron after repeated doses. Alosetron exhibits limited potential for inhibition of cytochrome P450-mediated metabolism. Interindividual differences in alosetron pharmacokinetics associated with demographic factors may be related to strong dependence on metabolism by CYP1A2.

    Notizen: Bovine serum albumin (BSA) enhances nicotinic agonist-induced 86Rb+ efflux from synaptosomal fractions of the mouse thalamus, but how it does so is not understood. The experiments reported here indicated that BSA enhancement of nicotinic acetylcholine receptor function was rapid, reversible, depended on BSA concentration, and occurred at all points of the nicotinic agonist concentration-response curve. We hypothesized that BSA-extractable compounds, such as long-chain fatty acids, were responsible for inhibiting nicotinic responses in the absence of BSA. The hypothesis was tested by applying arachidonic, linolenic, or oleic acids in the absence of BSA after an initial prewash with BSA. All three fatty acids exhibited a rapid, concentration-dependent inhibition of nicotinic-agonist stimulated ion flux. Concentration-response curves produced after 30 s of pre-treatment with arachidonic acid were similar to those seen when BSA was completely absent. The effects of pre-treatment were reversed immediately by the introduction of BSA. Furthermore, no effects of fatty acids were observed when preparations were continuously exposed to BSA or when BSA was continuously absent. These results suggest that the removal of endogenous, inhibitory compounds is largely responsible for the rapid, potentiating action of BSA at nicotinic acetylcholine receptors expressed in the mouse thalamus.
