Notizen: A microelectrode technique has been developed to enable the study of stress-generated potentials (SGP) in bone to a spatial resolution of 5 μm. The technique has been used to measure the electrical potentials as a function of bone micromorphology in four-point bending. Electric fields ranging from 30 to 103 times greater than is measured by conventional macroscopic methods have been discovered at the Haversian canals for human and bovine cortical bone. The amplitude and direction of the electric field in the osteons depend specifically upon the amplitude and the sign (i.e., compression or tension) of the stress. The implications of this finding with regard to the origin of SGP and their possible physiological significance are considered.

Notizen: The effect of collagen modification on the magnitude of stress-generated potentials has been studied. A whole bone model has been developed for determining the effects of collagen crosslinking on the electrical signals. Increased crosslinking results in larger electrical signals, and such increases result whether in vivo or in vitro changes in the collagen are made.

Notizen: By use of a previously developed microelectrode technique, the effect of nonuniform stresses on the stress-generated potentials (SGP) in bone were studied to a resolution of 5 μm. Comparison was made between uniformly and nonuniformly applied compression of human cortical bone. It was found that the radial electric fields for osteons in a specimen under uniform compression were equivalent, and such specimens possessed no macroscopic SGP; for nonuniform compression, the electric fields of osteons differed, and a macroscopic SGP was measured. The magnitude of the macroscopic SGP thus appears to be dependent upon local stress differences and, hence, on the SGP of local regions.

Notizen: The osteoconductive nature of calcium phosphate ceramics (CPC) follows from several proven effects, such as a direct bone attachment and enhanced bone tissue formation. Mechanisms leading to these phenomena are still largely undiscovered. Specifically, little is known about the CPC surface and cell-driven reactions. These atomic and molecular level events are involved in tissue attachment and enhanced tissue formation. It is hypothesized that the zeta potential of these ceramics is directly related to the surface reactivity governing osteoconductivity. As a first step in our analysis, the zeta potential of stoichiometric and Ca-deficient hydroxyapatite was determined as a function of immersion time. It is concluded that, under the conditions of the experiment, the observations support the hypothesis in a dual way. First, the absolute values of the zeta potential which were measured are related to electrokinetic potentials known to cause substantial effect on the cellular activities and bone tissue formation when applied exogenously. Second, the magnitude and duration of the changes in zeta potential are related to an ion exchange between the hydrated layer around the ceramic and the ceramic surface, and a net precipitation of new material. If these findings could be confirmed in other solutions, i.e., solutions with a substantially equivalent composition as the fluids in developing bone tissue, a basis would be provided to explain the bridging of the ceramic surface with the surrounding developing tissue.

Notizen: The preparation of high molecular weight linear polymethylene is described at low temperatures. The onset of a polymorphic transformation (triclinic to orthorhombic) is observed on warming this low crystallinity (∼50%) polymer to about -25°C. Relative rates of transformation above this temperature are also given. No significant increase in crystallinity occurs on warming the polymer up to room temperature, where some triclinic phase still persists. Annealing polymethylene at 100°C. leaves only the orthorhombic material which is approximately 80% crystalline. A glass transition temperature around -30°C., is reported for the low crystallinity polymer.
