Zusammenfassung Um Informationen über das Verhalten der Harnblasenwand gegenüber pharmakologisch interessierenden Substanzen zu erhalten, wurde an isolierten Harnblasenstücken von Meerschweinchen die Aufnahme von zwei sehr unterschiedlichen Verbindungen, nämlich von 14C-markiertem Prilocain und 14C-Harnstoff, untersucht. Gemessen wurde die Aufnahme in Abhängigkeit von der Zeit, von der Konzentration und dem pH-Wert im Angebot. Prilocain (pK a =7,7 bei 36° C) dringt als freie Base (pH-Wert des Milieus 9,0) schneller in das Blasengewebe ein als in protonisierter Form (pH 6,4). Prilocain wird im Blasengewebe angereichert. Die relative Anreicherung nimmt mit steigender Konzentration (0,156; 1,95 und 39 mmolar) im Angebot ab; es wird bevorzugt freie Base der Inkubationslösung entzogen. Etwa 3,5 μMol Harnstoff/Gramm Feuchtgewicht werden im Blasengewebe fest gebunden. Sie nehmen an Austauschvorgängen nicht teil. Bei Inkubation in 14C-Harnstofflösungen (0,2; 2,0 und 39 mmolar) erreicht die Gewebskonzentration jeweils etwa 75% der angebotenen Konzentration. Aufnahmegeschwindigkeit und Gewebskonzentration im Gleichgewicht sind vom pH-Wert der Inkubationslösung (6,4; 7,7 und 9,0) unabhängig; Harnstoff (pK a =0,18) liegt bei diesen pH-Werten in nicht-dissoziierter Form vor.

Summary In order to obtain information on the behaviour of the urinary bladder wall towards pharmacologically interesting compounds, the uptake of two considerably different substances (i.e. 14C-labelled prilocaine and 14C-urea) by isolated bladder pieces (guinea pig) has been investigated. The uptake was measured as a function of the incubation time, and of the concentration and pH-value of the bath fluid. The free base (pH-value of the medium 9.0) of prilocaine (pK a =7.7 at 36° C) penetrates more rapidly into the bladder tissue than the protonated form (pH-value of the medium 6.4). Prilocaine is concentrated by bladder tissue. The relative accumulation decreases when the prilocaine concentration of the bath fluid is raised (0.156; 1.95 and 39 mM). The free base is transferred from the medium to the isolated tissue to a relatively larger extent than the protonated form. Approximately 3.5 μmol urea/g tissue (wet weight) are firmly bound by the bladder tissue. The urea thus bound does not participate in exchange processes. Upon incubation in urea solutions (0.2; 2.0 and 39 mM) a tissue medium ratio of about 0.75 is reached at equilibrium. The uptake rate and also the tissue concentration at equilibrium are independent on the pH-value of the bath fluid (6.4; 7.7 and 9.0). At these pH-values urea (pK a =0.18) is present in its nondissociated form.
