    Notes: Summary In order to elucidate the action mechanism of dapsone on blood cell membranes, we assessed the dose-dependent effect of dapsone on the activities of choline phosphotransferase (which mediates the production of the structural phospholipid, phosphatidylcholine) and methyltransferase (which produces phosphatidylcholine from phosphatidylethanolamine, representing the dynamics of the cells) in the membranes of red cells, lymphocytes, and neutrophils obtained from 16 healthy human subjects. The methyltransferase activity of lymphocyte and neutrophil cell membranes was slightly inhibited by dapsone, although only at a high concentration (1 mM), while that of red cells was not affected. On the other hand, dapsone significantly decreased the choline-phosphotransferase activity of red-cell membranes in a dose-dependent fashion, but did not significantly inhibit that of lymphocytes or neutrophils. The mechanisms of the hemolytic side effect of dapsone on erythrocytes and its anti-inflammatory effect on neutrophils are discussed in connection with its inhibitory effect on the enzymatic activities of membrane phospholipids.

    Notes: Summary Oxygen intermediates (OIs) generated by stimulated polymorphonuclear leukocytes (PMNs) are known to induce auto-oxidative tissue damage at the site of inflammation. PMNs from five patients with severe and chronic cement dermatitis generated markedly increased levels of OIs. However, only a slight increase in OI generation by PMNs was observed in cement workers without cement dermatitis. Dapsone, which has recently been shown to decrease OI levels, was found to be clinically effective in the treatment of cement dermatitis in these five patients. After treatment, a significant decrease in OI generation was observed in all patients studied. In skin tissues from the cement workers without cement dermatitis, enhanced superoxide-dismutase (SOD) activities as well as increased OI generation by PMNs were noted. In spite of the greatly increased OI generation by PMNs, the SOD activities in the patients were comparable to those in healthy controls. These findings suggest that the severe skin manifestations in patients with cement dermatitis can partly be explained by a defective capacity for enhancing SOD activity which removes increased PMN-derived OIs and thus prevents subsequent tissue injury by OIs at the site of inflammation.

    Notes: Summary The levels of plasma testosterone, testosterone-oestradiol binding globulin (TeBG) and total serum acid phosphatase (TSAP) following antiandrogenic hormone therapy were investigated in 17 patients with prostatic carcinoma. The low levels of plasma total and free testosterone induced by castration decreased further after diethylsitlboestrol diphosphate (DES-D) administration. Plasma TeBG binding capacity after castration was 118.9% of the pre-treatment level and increased to 193.9%, 204.0% and 212.7% at 1, 2 and 3 weeks after DES-D dosing. The in vitro binding of 3H-testosterone to TeBG was not influenced in the presence of DES-D or stilboestrol. Clinical response following the DES-D therapy was associated with a decrease in the levels of TSAP. A significantly inversed correlation was found between the decrease in TSAP and increase in TeBG at completion of DES-D therapy. These results suggest that the high binding capacity of TeBG lowers the biologically active fraction of testosterone and thus may produce clinical effects.
