    Notizen: We have described previously the detection and tissue distribution of free cell surface receptors for ecotropic R-MuLV envelope glycoprotein and the growth factor EGF in vivo [1]. More recently, we have reported the chromosomal map position of the ecotropic viral receptor and its conservation between subspecies of the genus Mus [2]. This work has shown, for the first time, the presence of multiple, independently segregating cell surface receptor genes specific for different classes of ecotropic type C viral envelope glycoprotein. In this report we extend these findings and identify chromosome 2 as coding for the receptor used by M813, an ecotropic MuLV from a feral Asian mouse. This new receptor is probably also used by oncogenic, recombinant (MCF class) MuLV of C3H origin.

    Notizen: Hair proteins have been successfully analysed by high resolution two-dimensional electrophoresis by avoiding S-carboxymethylation which has previously dictated the use of alternative two-dimensional procedures. The high and low-sulfur proteins are simultaneously resolved within the pI range 3-9 and their electrophoretically determined Mr values are consistent with molecular weights obtained by physical methods.

    Notizen: Ampholine pH 4-6 and Pharmalyte pH 4.2-4.9, 4.5-5.4 or 5-6 have been used to improve the resolution of the simplified technique of high resolution two-dimensional electrophoresis. All give excellent resolution but Pharmalyte interferes with protein detection methods to an extent which increases with the pH range of the mixture. The effect is characterised by a progressive increase in the intensity and area of background stain associated with the anode and cathode regions of the two-dimensional gels. It is minimal following Serva Blue R (Coomassie Blue) staining but severe following silver staining.

    Notizen: A methylamine-incorporating silver stain is described for the detection of deoxyribonucleic acid (DNA), protein and lipopolysaccharide in thick (3 mm) polyacrylamide gradient gels (4-20 % w/v). Its sensitivity for DNA and protein is more than 100 times greater than ethidium bromide and Coomassie Brilliant Blue R-250, respectively. It detects 0.01 ng DNA, 0.16 ng of human serum albumin and ng quantities of lipopolysaccharide. DNA gives red bands whereas protein and lipopolysaccharide give brown/black bands. The staining procedure is inexpensive, highly reproducible and overcomes the problem of background stain associated with thick gels. It also gives excellent results with thin (1 mm) gels and can be used to stain a number of gels simultaneously. A simplified staining procedure is also discussed.

    Notizen: One-dimensional sodium dodecyl sulphate-polyacrylamide gel electrophoresis (SDS-PAGE) of protein in the presence of Nonidet P-40 (NP-40) induces vertical streaking which breaks horizontal bands of protein to produce a “multiple spot” effect. The same effect can be induced upon high resolution two-dimensional electrophoresis if the NP-40 concentration of the first-dimension isoelectric focusing (IEF) gels is increased to 4 % w/v and the gels electrophoresed (SDS-PAGE) without SDS-equilibration. The relevance of this phenomenon as regards interpretation of two-dimensional electrophoretic patterns is discussed.

    Notizen: An improved method is described for Nitro Blue Tetrazolium staining of creatine kinase isoenzymes following agarose gel electrophoresis or isoelectric focusing in polyacrylamide gels. The method allows the standard although transient fluorescent stain to be followed by Nitro Blue Tetrazolium staining to give a more permanent positive stain without background interference. It employs. 0.05 M para-chloromercuribenzoate to block the adverse effects of sulfhydryl compounds (included in commercial creatine kinase-reagents) upon Nitro Blue Tetrazolium staining.

    Notizen: The native protein patterns of human serum and unconcentrated cerebrospinal fluid, amniotic fluid, urine, sweat and tears have been detected by silver staining following two-dimensional electrophoresis in the absence of protein denaturants. Over 100 proteins were detected in both urine (80 μl) and sweat (40 μl), thereby demonstrating the high sensitivity of the approach. However, optimal sensitivity was only achieved following pre-equilibration of the gels in sodium dodecyl sulphate. The implications and possible nature of stain enhancement are discussed.

    Notizen: Four two-dimensional electrophoresis methods, each incorporating different protein denaturing/dissociating conditions, have been used to detect changes in rat serum proteins following exposure to dimethylformamide. The methods were evaluated on the basis of (i) their ability to detect changes, (ii) the resolution achieved and (iii) the response of resolution to the use of narrow Ampholine pH ranges in the first dimensional isoelectric focusing gels. All methods reveal minor but reproducible protein changes in response to dimethylformamide. However, high resolution two-dimensional electrophoresis of the completely dissociated polypeptides reveals most changes and the resolution of this method is most improved by the use of Ampholines of narrow pH range. In general, the number of serum components detected and the resolution achieved is related to the severity of the protein denaturing/dissociating conditions employed. The significance of this observation in relation to others factors likely to influence the choice of two-dimensional method is discussed.

    Notizen: The proteins of cheese are rapidly solubilised by heating to 95 °C in buffered 2 % sodium dodecyl sulfate, 5 % 2-mercaptoethanol. Electrophoretic analysis of the solubilised proteins by either one-dimensional sodium dodecyl sulphate-polyacrylamide gel electrophoresis or high resolution two-dimensional electrophoresis yields reproducible patterns characteristic of an individual cheese and its extent of ripening. The patterns reveal (i) the residual amounts of milk casein and whey proteins, and (ii) the appearance of casein degradation products, including pink-violet components as detected by Coomassie Blue staining.

    Notizen: A simplified technique of high resolution two-dimensional electrophoresis has been used to investigate heterogeneity of human serum myeloma proteins. Three types of microheterogeneity have been detected. The heavy chains of IgA, IgM and IgD myeloma proteins demonstrate charge heterogeneity which is far less extensive following neuraminidase treatment and is therefore largely due to sialic acid. The underlying charge heterogeneity, which is apparently resistant to neuraminidase, resembles the charge heterogeneity demonstrated by the heavy chains of IgG myeloma proteins which show no response to neuraminidase treatment. The third type of microheterogeneity is most clearly demonstrated by the heavy chains of IgD myeloma proteins which exhibit heterogeneity with respect to small variations (∼3000) in molecular weight. This type of heterogeneity has also been observed, to a lesser extent, in the heavy chain of IgM and IgA myeloma proteins.
