Notizen: Summary Chemical modifications suggested that acidic amino acids such as aspartic and glutamic acids are involved in the active sites ofBacillus cereus sphingomyelinase. Among aspartic acid residues in the conserved regions of this enzyme, Asp-126, Asp-156, Asp-233 and Asp-295 were converted to glycine by site-directed mutagenesis. According to prediction on structural similarity to pancreatic DNase I, His-151 and His-296 were also converted to alanine. The Asp and His mutants, D126G, D156G, D233G, D295G, H151A and H296A, were produced inBacillus brevis 47, a protein-hyperproducing strain. The catalytic activities of D295G, H151A and H296A were completely abolished, and sphingomyelin-hydrolyzing activity of D126G or D156G was reduced by more than 50%. The activity of D126G towardp-NPPC was comparable to that of the wild-type, while D156G catalyzed the hydrolysis of HNP andp-NPPC more efficiently than the wild-type. Hemolytic activities of the mutants were parallel to their sphingomyelin-hydrolyzing activities.

Notizen: In order to clarify the general kinetic behavior of emulsion polymerization initiated by oilsoluble initiators, the emulsion polymerization of styrene initiated by 2,2′-azoisobutyronitrile was as a typical example, investigated thoroughly. The variations of the polymerization rate and the number of polymer particles produced with changes in emulsifier (sodium lauryl sulfate), initiator, and monomer concentrations initially charged and the reaction temperature were determined. It is shown from these experimental results that the kinetic behavior of this emulsion polymerization system is quite similar to that of styrene emulsion polymerization initiated by the water-soluble initiator, potassium persulfate despite the difference in the principal loci of radical production in both systems.

Notizen: Abstract Cattle avoid grazing forage near their feces on pasture for more than a month. The relation of cattle feces odor to the rejection was studied in feeding choice tests using cattle. The feeding deterrent of feces odor was investigated using a trough partitioned to allow passage of feces odor through test food to the cattle. The cattle did not feed from the trough containing feces 0–35 days after excretion. Volatile chemicals isolated from feces 0–35 days after excretion by a cold trap method also inhibited cattle from feeding. These results demonstrated that feces odor is a major cause of the unpalatability of forage around cattle feces. The volatile chemicals were separated into ether and aqueous fractions. The ether fraction inhibited the feeding behavior of cattle. Furthermore, the ether fraction was separated into neutral, acidic, and basic fractions. The neutral fraction of the three was bioactive. These results suggested that specific volatile chemicals emitted from cattle feces were deterrents to the feeding behavior of cattle.

Notizen: The mechanical properties of the hydrogel membrane of poly(methyl methacrylate-N-vinylpyrrolidone) were studied for the purpose of making soft contact lenses for extended wear. The dependence of tensile fracture energy, flexibility, and recovery from deformation on the water content and thickness of the hydrogel membrane was studied. Reducing the thickness of a lens was found to exert a more advantageous effect than increasing its water content on maximizing the tensile fracture energy under the condition of an adequate supply of oxygen to the cornea through the contact lens. As long as its water content is controlled between 63 and 78%, the contact lens made of poly(methyl methacrylate-N-vinylpyrrolidone) has the flexibility on the same order as that of conventional soft contact lenses of poly(2-hydroxyethyl methacrylate). The rate of recovery from deformation increases with water content, and the residual elongation is negligible in the range of water content over about 70%. It is concluded that practical requirements for use of these contact lenses, determined by these three mechanical properties, can be satisfied at the same time if the water content of the contact lens is adjusted at about 70%.
