    Notizen: Summary A study of 6-phosphogluconate dehydrogenase and malate dehydrogenase isozyme expression in Triticum turgidum conv. durum /Hordeum chilense monosomic addition lines has revealed the location of two structural genes, 6-pgd-H ch 2 and Mdh-H ch 1, on chromosome 1Hch of H. chilense. The homoeology between 1Hch and other chromosome of Triticeae related species is discussed on the basis of isozyme gene analysis.

    Notizen: Summary Polyacrylamide and starch gel electrophoresis of esterase (EST), glutamate oxaloacetate transaminase (GOT) and phosphoglucomutase (PGM) isozymes in Hordeum chilense, Triticum turgidum conv. durum, the amphiploid H. chilense X T. turgidum (Tritordeum), and the durum wheat/H. chilense monosomic addition lines revealed the chromosomal location of one EST locus, two GOT loci and one PGM locus. Loci Est-H ch1 and Got-H ch2 were found on chromosome 6Hch,Got-H ch3 on chromosome 3Hch, and Pgm-H ch1 on chromosome 4Hch. These results lend evidence for the assumed homoeology relationships between chromosomes of Triticeae species.

    Notizen: The process of T-lymphocyte differentiation within the thymus involves a series of molecular interactions. In this work we have carried out an analysis of the chick thymus microenvironment in order to evaluate its heterogeneity during development. We have produced 11 monoclonal antibodies whose staining patterns detected by the immunoperoxidase technique allowed us to divide them into five groups. A first group (E19-E2, P0-E5, and P15-T1) binds to thymic medullary stroma showing a reticular pattern on medullary epithelial cells and whose significance would be related to thymic stromal secretion. The second group of monoclonal antibodies (P15-T3) stains thymic corpuscles of 10- and 15-day chicks. The third group of antibodies includes P0-E1, P0-E3, P5-A6, and P15-T2 whose staining pattern is both medullary and cortical. The fourth group (P10-HB1 and P10-HB2) binds to thymic stromal and cortical thymocytes, and the fifth group (P5-A1) is characterized by the staining of medullary vessels of 5-day chicks. These five groups of monoclonal antibodies corroborate the existence of an antigenic diversity of the chick thymus microenvironment. Their possible relationships with T-cell differentiation and stromal-thymocyte interactions are discussed. © 1993 Wiley-Liss, Inc.

    Notizen: We have analyzed the effects of colchicine on the cell shapes in chick neuroepithelium. Cell shapes were ascertained by the position of the nucleus in plastic serial sections. We tested three colchicine doses (5 × 10-5 M, 5 × 10-6 M, and 5 × 10-7 M) by two experimental treatments (in ovo and in vitro). Colchicine treatment in vitro is always effective in depolymerizing microtubules of neuroepithelial cells and reduces the percentages of wedge-shaped cells in the median area of neuroepithelium. The same effect can be observed when the embryos are treated with 5 × 10-5 M or 5 × 10-6 M colchicine in ovo. A concentration of colchicine of 5 × 10-7 M in ovo cannot disrupt microtubules in stage 8 and stage 10 embryos, and the percentage of wedge-shaped cells is the same as that of the untreated cells. In stage 6 embryos this colchicine dose effects the microtubules and the percentages of wedge-shaped cells. These facts are interpreted in respect to variations in microtubular resistance to microtubular-disrupting agents that are shown by the neuroepithelial cells from different developmental stages.

    Notizen: Background: Preimplantation development of mammalia is characterized by cell surface changes functioning in intercellular communication and adhesion. The glycoconjugate role in cellular interactions has been analysed for several groups but not in sheep embryos. The binding patterns of eleven lectins during sheep preimplanatation development were investigated and the role of glycoconjugates in early development was discussed.Methods: Ultrathin sections from preimplantation ovine embryos (3-7 days) were incubated with different colloidal gold conjugated lectins and the frequency of gold particles on the cell membrane, some organelles, and the zona pellucida was evaluated.Results and Conclusions: We observed a higher staining of WGA, DBA, and SBA lectins in the intercellular contact zone with respect to the free cell surface of blastomeres during cleavage. This indicates that the N-acetyl galactosamine and N-acetyl glucosamine residues may be involved in sheep morula compaction. In contrast, the trophoblast cell displays an increase of staining of some lectins previously identified during cleavage (LcH, WGA, SBA, MPA, and PNA) on the free membrane, and a lack of sugar residues in the intercellular surface. This polarization of the trophoblast cell surface is not observed in the inner cell mass and could provide a mechanism for differentiation within the blastocyst. Intracytoplasmic vesicles show a cytochemical identity with lysosomes in the blastocyst (abundant GlcNAc and Man/Glc residues) that may reflect a functional relationship between both organelles in an intracellular cycle. The zona pellucida presents abundant GalNAc, GlcNAc, and Gal residues during preimplantation ovine development. © 1994 Wiley-Liss, Inc.
