    Notes: The treatment of male factor-related infertility has been approached with the advent of several methods for microsurgical fertilization, such as the partial dissection of the zona pellucida (PZD) and the injection of sperm into the perivitelline space (PVSI) of oocytes. These techniques are designed to increase sperm-oolemma interaction by circumventing passage of the sperm through the zona pellucida. The present study was performed to evaluate the influence of PZD and PVSI on the in vitro development of mouse embryos by assessing the rate of sister chromatid exchange (SCE). SCE is considered to be a sensitive indirect indicator of DNA lesions due to various conditions. Oocytes were cultured in vitro after PZD or PVSI and then examined for SCE. There was no significant difference in SCE between control and treatment groups of embryos and the values were similar to those reported by Saito et al. (Fertil Steril 41:460-464, 1984). The rate of SCE was low during the first two mitotic cycles, then increased from cycle two to three before declining slightly between the 3rd and 4th cycles of cell division. These data demonstrate that the direct interaction of sperm and oocyte by PZD or PVSI did not have an adverse effect on the development of mouse embryos as assessed by the rate of SCE. © 1994 Wiley-Liss, Inc.

    Notes: Rhodamine phalloidin (Rph) staining was used to examine the microfilament organization of the Xenopus laevis egg cortex during the early stages of fertilization. Unactivated eggs possessed a cytochalasin B (CR)-insensitive Rph-stained matrix that was reorganized upon egg activation and diminished in the presence of CB. Xenopus laevis sperm caused a temporary local increase in Rph staining on the Xenopus cortex. In CB-treated eggs, the local increases of cortical Rph staining later changed to a Rph-free area. These temporary local increases of cortical Rph staining were also observed when Notophthalmus viridescens sperm fertilized Xenopus and Rana pipiens eggs, and were followed by the appearance of concentric rings of stained and unstained areas. Our data suggest that Xenopus and Notophthalmus sperm have activities that can both organize and disrupt the cortical filamentous actin of the Xenopus egg. © 1993 Wiley-Liss, Inc.

    Notes: The thrombospondins are a family of related glycoproteins found in the embryonic extracellular matrix. To date, five members of this family have been identified. Thrombospondin-1 and thrombospondin-2 have similar primary structure, but are expressed in different tissues at different times during development. Thrombospondins-3, -4, and cartilage oligomeric protein belong to a second thrombospondin subgroup in which the carboxyl-half of each molecule is most similar to thrombospondin-1 and -2. Here, we report the cloning and sequencing of a novel probe to avian thrombospondin-4. We have used this probe to determine the origins of thrombospondin-4 in the chick embryo by in situ hybridization. Thrombospondin-4 transcripts first appear in the mesenchyme surrounding bone anlage coinciding with the initial stages of osteogenesis. The expression in osteogenic tissues is transient: thrombospondin-4 mRNAs are not seen in the osteoblasts of bone collars in developing long bones. This pattern is distinct from avian thrombospondin-2, which is expressed in perichondrium and embryonic fibrous connective tissues. Our observations indicate that connective tissues are the principal site of thrombospondin-4 expression in the chick. The diverse origins of different thrombospondin gene family members imply distinctive roles for these proteins related to the growth and differentiation of cartilage, tendons, and bone. ©1995 Wiley-Liss, Inc.

    Notes: Cast linear polyethylene films subjected to dry and solvent annealing display markedly different sorption and difusion barrier properties than do untreated films. The subsequent sorption of liquid o- and p-xylene and cis- and trans-acetylene dichloride per unit volume of amorphous polymer increases as the annealing temperature and/or treating solvent concentration increases. Integral diffusivities calculated from sorption and steady-state permeation rates show a monotonic increase with sorption per unit volume of amorphous polymer. The concentration dependence, however, is less marked than observed in similar systems at low permeant activity. Apparently the above treatment reduces the effective crosslinking imposed by the crystallites on the amorphous polymer chains through disentangling and incorporating some of these chains into crystallites. Thus the polymer is capable of a greater degree of swelling when brought into contact with a compatible liquid in a spite of a higher degree of crystallinity. The low concentration dependence of the diffusivities is probably due to heterogeneous distribution of excess permeant in a treated film. If the excess permeant were preferentially sorbed in regions of low polymer concentration then the above observations could be explained. Long-duration, osmotic stress-induced swelling and recrystallization have been cited to account for time-dependent permeation rates in treated and untreated films.

    Notes: The configurational entropy of the polyethylene chain at the melting points calculated in two ways. In both calculations, tetrahedral angles and discrete trans and gauche arrangements of all bonds are assumed, and trans bonds are assumed more stable than gauche by energy U1. First, calculations are made on chains of up to N = 18 bonds, disallowing all configurations having overlapping atoms, and the result is extrapolated to large N. Second, a calculation is made directly for long chains, with overlaps excluded only over every short chain segment. The results are in almost exact agreement, suggesting that the second method can be safely used with other molecules. The calculated configurational entropy is in line with that suggested by the entropy of fusion, assuming the chains to acquire a configurational freedom in the melt which approaches that of independent chains.

    Notes: A vacuome (‘Golgi apparatus’) consisting of small globular inclusions has been demonstrated in Chlamydomonas sp. These inclusions may be seen in the living, unstained organism; they are stainable vitally with neutral red; they have been stained vitally with neutral red and then blackened with osmic vapor under direct observation, and they have been impregnated by osmic and silver methods without previous treatment with neutral red.The reaction of these inclusions to the iodin test for starch suggests that they may play some rǒle, possibly one of storage, in the cycle of starch metabolism.

    Notes: The parotid and the principal and accessory submandibular glands of the little brown bat, Myotis lucifugus (Vespertilionidae), were examined using light microscopy and staining methods for mucosubstances. The parotid gland is a compound tubuloacinar seromucous gland. Parotid gland secretory cells contain both neutral and nonsulfated acidic mucosubstances. The principal and accessory submandibular glands are compound tubuloacinar mucus-secreting glands. They contain somewhat atypical mucus-secreting demilunar cells that often appear to be interspersed between mucous tubule cells. The mucous tubule cells in both the principal and accessory submandibular glands contain sulfomucins. Demilunar cells of the principal submandibular gland contain moderate amounts of nonsulfated acidic mucosubstances, but the corresponding cells of the accessory submandibular gland contain considerable neutral mucosubstance with very little acid mucosubstance. Intercalated ducts composed of cuboidal or low columnar epithelial cells are present in all three glands. Striated ducts in all glands are composed of columnar cells whose apices bulge into the ductal lumina. Excretory ducts are composed of simple columnar epithelium, with occasional basal cells that suggest a possible pseudostratified nature. The cells of the excretory ducts also have bulging apices. All duct types contain apical cytoplasmic secretory material that is a periodic acid-Schiff positive, neutral mucosubstance. Ductal apical secretory material is more evident in intercalated and striated ducts than in excretory ducts.

    Notes: The Puerto Rican tree frog, Eleutherodactylus coqui, has internal fertilization and direct development on land. In light of these reproductive adaptations, the events of fertilization and early development were studied. Cytological examination of just-fertilized eggs showed that sperm entry is restricted to about 10% of the surface of these large, yolky eggs, and all nuclear events of the first cell cycle occur near the animal pole. Although the oocytes have cortical granules, a number of polyspermic fertilizations were found. One clutch consisted of eggs with a high frequency of polyspermy and of normal development. This raises the possibility that normal development can occur despite multiple sperm entry, a situation not found in other anuran amphibians. With respect to saline requirements, the sperm and the embryo are similar to those in amphibians with external fertilization and aqueous development. Sperm motility was high in low-tonicity conditions, and the normally terrestrial embryo could develop completely from a fertilized egg to a froglet in a low-tonicity aqueous solution.

    Notes: It is shown that the Hirai-Eyring model for the liquid state is capable of accurately describing the p, V, T behavior of liquid polymers in the temperature range over which measurements are now made, and below. Once the parameter choices necessary to accomplish the fit are made for a particular polymer, the excess thermodynamic functions (differences in properties, liquid less solid) are determined by the same parameters. Above the glass transition temperature Tg the volume, excess enthalpy, and square of the excess entropy are predicted by the model to be essentially linear with temperature, in agreement with experiment. Below Tg, these functions do not remain linear (as is usually assumed in extrapolating the equilibrium behavior to low temperatures), but instead they rapidly approach zero in a continuous way as the temperature is lowered. These remarks apply to glass-forming materials composed of small molecules, as well as to polymers. The “paradox” raised by Kauzmann is thus resolved, and the Gibbs-DiMarzio second-order transition appears to be unnecessary.

    Notes: A study of the effect of catalyst (base) concentration and N-acylcaprolactam cocatalyst size and substitution on the fast anionic polymerization of caprolactam indicated that a steric effect due to cocatalyst size exists, and perhaps an electronic effect due to cocatalyst substitution was noted. The rate of polymerization, degree of polymerization, and yield of polymer are related to these effects. It was also noted that at high base concentrations, the rate and degree of polymerization along with the product yields all decrease. These latter observations suggest that reinterpretation of some of the reaction mechanism data may be important if polymer degradation is not an appreciable factor during the reaction.
