    Notes: A huge amount of data is collected by computer monitoring systems in the chemical process industry. Such tools as principal component analysis and partial least squares have been shown to be very effective in compressing this large volume of noisy correlated data into a subspace of much lower dimension than the original data set. Because most of what is eliminated is the collinearity of the original variables and the noise, the bulk of the information contained in the original data set is retained. The resulting low dimensional representation of the data set has been shown to be of great utility for process analysis and monitoring, as well as in selecting variables for control. These types of models can also be used directly in control system design. One way of approaching this is to use the loading matrices as compensators on the plant. Some advantages of using this approach as part of the overall control system design include automatic decoupling and efficient loop pairing, as well as natural handling of nonsquare systems and poorly conditioned systems.

    Notes: Oligonucleotide-directed mutagenesis has been used to replace α-helical glycines in the N-terminal domain of λ repressor with alanines. Since alanine is a significantly better helix-forming residue than glycine, these changes were predicted to have a stabilizing effect. We show that the Gly46→Ala substitution, the Gly48→Ala substitution, and the double substitution increase the melting temperature of the N-terminal domain by 3-6°.

    Notes: The synthesis of a variety of potent macrocyclic growth hormone secretagogues, i.e. 5, 9, 12, and 20-22, based on the known lead structure L-692,429 (1) is described. These conformationally constrained growth hormone secretagogues were prepared by joining the two essential pharmacophores, the amino-acid side chain at the 1H-1-benzazepine moiety and the 1,1′-biphenyl moiety with a variety of linkers. The most potent analog was found to be L-744,080 (21), a derivative in which a 2′-carboxamide moiety at 1,1-biphenyl is N,O-joined to the OH group of the (2-hydroxypropyl)amino-acid side chain by a C4 ester linker. This potent analog may be useful in determining the bound conformation of the benzolactam class of growth hormone secretagogues at the newly identified GHS receptor, L-744,080 (21) with an ED50 of 20 nM was up to fifty times more potent than the seco-acid precursor and 3-fold more potent than the parent 2′-tetrazole compound L-692, 429 (1).

    Notes: Fragmentation of methyl 3-O-benzyl-6-bromo-2-O-(tert-butyldimethylsilyl)-6-deoxy-4-O-(4-methoxybenzyl)-α-D-mannopyranoside (2) and in situ reductive amination give (3R,4S,5R)-6-benzylamino-4-benzyloxy-5-(tert-butyldimethylsilyloxy)-3-(4-methoxybenzyloxy)hexene (3) which is converted by intramolecular aminomercuration into an epimeric mixture of the bromomercuriopiperidine derivatives 4 and 5. The minor D-manno epimer 4 is transformed into mesylate 10 which, upon reaction with LiN3 in DMF, suffers a piperidine-pyrrolidine ring contraction to give azidomethylpyrrolidine 11. Reductive acetylation by means of thioacetic acid affords the fully protected pyrrolidine aza sugar derivative 12. The major L-gulo epimer 5 is transformed into the mesylate 15 which, again, upon reaction with LiN3 in DMF undergoes piperidine-pyrrolidine ring contraction to give the 1-azido-3-O-benzyl-2,5-benzylimino-6-bromomercurio-1,2,5,6-tetradeoxy-4-O-(4-methoxybenzyl)-L-iditol (16). Eliminative dehalomercuration by means of Zn powder is accompanied by in situ reduction of the azido group to give amino alkene 17. Intramolecular aminomercuration of 17 followed by N-acetylation yields the protected 1,2,5,6-tetradeoxy-2,5-imino-D-glucitol derivative 20 as the major epimer. Reductive oxygenation of 20 leads to pyrrolidine 22 which is deprotected to give title compound 1. Selective cleavage of the PMB group of 22 leads to 24. Benzylation of 22 affords again 12 from which the OH group at C-4 is liberated by selective cleavage of the PMB ether to give pyrrolidine 13. Compound 1 inhibits NAGase from bovine kidney.

    Notes: A study of properdin factor B (Bf) in 22 North and Central American populations demonstrated gene clusters delineating Old World Caucasian, New World and Black Ancestoral groups. Result of gene frequencies were comparable to data in the literature of European Caucasian and African Blacks.     Notes: Quantification of low levels of psychotropic drugs (10-7 to 10-9 g ml-1) in small volumes of plasma requires sensitive and accurate methods. Validation of these methods is best achieved by comparing results obtained using several techniques. In this study, amphetamine levels in plasma were measured using gas chromatography mass spectrometry and radioimmunoassay. Correlation of the results obtained by the two methods was found to be positive and high (R = 0.9822). The average coefficient of variation between assays for gas chromatography mass spectrometry was 5.8% and for radioimmunoassay was 12.3%, while the average coefficient of variation within assays for gas chromatography mass spectrometry was 4.9% and for radioimmunoassay 6.9%. Although gas chromatography mass spectrometry was 1.9 times more sensitive than radioimmunoassay, for most purposes, the convenience of the radioimmunoassay method outweighs the technical superiority of gas chromatography mass spectrometry.

    Notes: Micro-polyacrylamide gel electrophoresis (micro-PAGE) has been shown to be useful for molecular size separation of urinary proteins. In addition to the sodium dodecyl sulfate micro-gradient gel electrophoresis in 10 μL capillaries, which is used routinely for examination of proteinuria in our laboratory, we tried micro-gradient slab gel electrophoresis (micro-slab-PAGE) for molecular weight determination of urinary proteins. It was also employed for monitoring patients with proteinuric diseases. For detailed studied of urinary proteins, micro-two-dimensional-electrophoresis is recommended.

    Notes: Two polyacrylamide gel electrophoretic systems for the separation of small peptides are described. In both systems the samples are concentrated by a buffer discontinuity, and the peptides separated according to size and charge in 40-50 % polyacrylamide resolving gels. The acidic gel has an operative pH between 3.0 and 3.1 while the alkaline gel has an operative pH between 9.1 and 9.2. The lower limit of resolution is about 100 daltons for a peptide with one positive charge in the acidic system. The radioactive peptides are detected by autoradiography. Peptides with a free amino group can be fixed in these gels with formaldehyde, so that fluorographic procedures can also be used for increased sensitivity. These techniques enable precise comparison of non-derivatized peptides from complete protein fingerprints and offer a simple one-dimensional analytical method for protein comparison, as well as a general method for separating and analyzing small peptides.
