    Notizen: Abstract It has been suggested that distribution of lateral interspace resistance in association with a highly conductive junction can significantly affect the measurement of outer membrane(o)/epithelial(t) voltage divider ratios (F o=δV o/δV t), thereby leading to erroneous inferences regarding the outer membrane fractional resistance [fR o=R o/R c=R o/(R o+R i)], whereR o andR i are the outer and inner cell membrane resistance respectively andR c is the total cell membrane resistance. We present here experimental evidence for this point of view. During seasons when frog skins were highly permeable to Cl, transepithelial conductanceg t often exceeded 2 mS/cm2. High concentrations of external amiloride rapidly blocked cellular transport, butg t initially remained high andF o remained appreciably less than 1.0. These values ofF o were found here to result from low junctional resistanceR j: increase ofR j, either gradually following the administration of amiloride, or abruptly with external replacement of Cl by other anions, was associated with increase ofF o to near unity, without effect on the membrane potential or significant change in the short-circuit current. Experimental results following amiloride validated a simple equivalent circuit model predicting near-linear increase inF o with progressive decrease ing t and led to plausible values ofR j and lateral space resistanceR l. The possible influence of the paracellular resistance pattern on the evaluation of cell membrane resistances from voltage divider ratios is discussed.

    Notizen: Abstract Wash out analysis of tracer Na from frog skins after labelling from the epithelial or corial side was performed under control conditions and after transport inhibition by amiloride or ouabain. Wash out from the transport pool, i. e. after labelling from the epithelial side, into the corial bathing solution could be resolved into two individual components under all conditions. The fast one with half times of wash out of about 1.8 min correlates with the rate of transepithelial Na transport. It cannot unequivocally be associated with intra- or extracellular tissue spaces. The slow component with half times of about 18 min is essentially independent of the transport rate. The amount of Na washed out as this component increased considerably after transport inhibition with ouabain, suggesting intracellular location of at least a fraction of the transport pool, which appears to be made up by the entire epithelial cell layers. Wash out of tracer Na from the transport pool to the epithelial side is small compared to wash out to the corial side. Tracer wash out characteristics after labelling from the corial side were similar for wash out to the epithelial and the corial side. This suggests that considerable paracellular fluxes contribute to the Na flux from the corial to the epithelial side. The total intracellular Na present in the epithelium after ouabain can be labelled from the corial side. Carrier mediated Na−Na exchange across the basolateral membranes is proposed to account for this.

    Notizen: Summary The potential profile of the frog skin epithelium incubated in Cl-Ringer was reinvestigated with improved technique. Under open circuit conditions (PDtr up to 120 mV) the potential profile was demonstrated to be trough like in contrast to the stair-step like potential profile reported by previous investigators which has probably been recorded after injuring of the punctured cells. In 67 successful impalements the potential difference across the basolateral membranes was 108±2 mV. The potential across the outer, epithelial-facing membrane was inversely related to the transepithelial PD, but was found to be negative (with respect to the epithelial bathing solution) in all punctures. Electrogenic sodium transport might be responsible for part of the potential difference across the basolateral membranes which correlated directly with the short circuit current. The intracellular potential in the short circuited state was −73±2 mV and decreased with increasing short circuit current. It is suggested that these changes result from variations of the outer membrane sodium conductance.

    Notizen: Summary The wash out kinetics of Na-24 out of the transport pool of the frog skin were followed simultaneously to the epithelial and corium side in control experiments and after inhibition of the Na transport with ouabain. A perfusion system was used which allowed changes of the perfusion chambers within 5 sec between labelling and tracer wash out. In the control, the Na of the transport pool is washed out with a half time of 2.9±0.3 min and 3.2±0.7 min to the corium and the epithelial side, respectively. After ouabain, the half times increased to 8.9±0.9 min and 8.6±1.1 min for the corium and the epithelial wash out. From this observation it is concluded that the transport Na is distributed in an intracellular compartment. The ratio of the two tracer effluxes from the transport pool to the corium and the epithelial sidej 23 * /j 21 * is 30±8 in the control and 4.9±2.0 after ouabain. The wash out to the epithelial side shows an additional fast component (half times 0.70±0.08 min in the control and 0.68±0.13 after ouabain) which is interpreted as wash out of an extracellular compartment. This view is supported by measurements of the wash out of Inulin C-14.

    Notizen: Summary The effect of ADH upon the intracellular potential and the resistance of inner and outer borders of the transport pathway was investigated on isolated skins ofRana temporaria. Within 40 min after ADH (100-300 mU/ml), the intracellular potential under short-circuit conditions decreased to about 40% of the control value (−79±4 mV), concomitant with an increase in the short-circuit current to about 160% of the control value. Amiloride, applied when steady values under ADH had been reached, caused an immediate rise of the intracellular potential to values typical for control conditions. This confirms (i) the intracellular location of the microelectrode and the absence of impalement artifacts, and (ii) the ineffectiveness of ADH upon the electromotive forces of the inner border. ADH had no effect upon the intracellular potential after blockage of the Na entry by Amiloride. The equilibrium potential of the outer border was estimated to be about +20 mV under the influence of ADH. As this value is considerably less positive than might be expected for the chemical potential of Na, a significant contribution of ions other than Na to the outer border conductance and equilibrium potential is implicated. The resistance of the outer border was more significantly decreased than that of the active transcellular pathway after ADH due to an increase in the inner border resistance, which exceeded that of the outer border after ADH. The effect of ADH upon the outer membrane characteristics would be underestimated by a factor of two, if the alterations of the electrical potential difference were not taken into consideration.

    Notizen: Summary The effect of Li upon the intracellular potential of frog skin (Rana esculenta) was investigated. In the range between 1 and 25mm Li in the epithelial bathing solution, a semilogarithmic linear relationship between [Li] and intracellular potential under short circuit conditions was obtained. The intracellular potential at all [Li] is quantitatively sufficient to explain the previously reported accumulation of Li in the intracellular space of the frog skin epithelium (Leblanc, G. 1972.Pfluegers Arch. 337:1) on the basis of a passive entrance step at the outer border. A reduction of the intracellular potential by Li is also observed in the presence of 6mm Na in the epithelial bathing solution. Consequences regarding the mechanism of uptake of Na across the outer border of the frog skin are discussed.

    Notizen: Summary The path of the transepithelial Na transport through the frog skin was investigated by the use of compartment analysis. Therefore tracer wash-out kinetics from the preloaded epithelium to both sides of the skin and time course of tracer uptake at the epithelial side were measured under control conditions and after alteration of transport parameter by amiloride, ouabain and incubation at 0°C. In all experiments the skins were short-circuited throughout and the SCC registered. The results indicate that the major part of the Na involved in transport is already transported and located in an extracellular compartment, possibly the intercellular space between the epithelial cells. The decisive step of the transepithelial transport seems to be located at the outer epithelial border of the cell layer since entrance of Na into the skin is similarly altered by all inhibitors of transport. Two additional compartments which are labelled from the epithelial side are detectable. The Na content of the compartment with the lowest exchange rate is increased by ouabain and 0° C but remains unchanged after amiloride. It might be located intracellularly but is not responsible for transepithelial transport. The actual transport pool must be very small.
