    Notes: Abstract We have recently shown that the angiotensin I converting enzyme (ACE) gene is linked to NaCl-loaded blood pressure in the stroke-prone spontaneously hypertensive rat (SHRSP), and that high-NaCl loading selectively stimulates ACE in the aorta of SHRSP but not in normotensive Wistar-Kyoto (WKY) rats. We therefore investigated the relationship between cardiac ACE and the development of hypertension and left ventricular hypertrophy in response to normal- and high-NaCl diet in these rats. ACE mRNA and ACE activity were measured in left ventricular tissue after completion of hemodynamic characterization of the animals. While SHRSP rats increased blood pressure (P〈0.0001) and heart rate (P〈0.005) in response to high NaCl, blood pressure remained unchanged in WKY. Similarly, relative left ventricular weight increased only in SHRSP after high NaCl (P〈0.002). A significant two- to threefold increase of cardiac ACE mRNA and fourfold stimulation of ACE enzyme activity in response to high NaCl was found in both WKY and SHRSP rats (P〈0.005). The induction of ACE gene expression was significantly more pronounced in SHRSP compared to WKY (P〈0.02), whereas no significant strain differences in left ventricular ACE activity were found after either normal- or high-NaCl diet. Thus, arterial blood pressure and left ventricular weight remained unchanged in the WKY rats despite the activation of left ventricular ACE activity after high-NaCl exposure. These results demonstrate that left ventricular ACE activity is equally upregulated in response to high-NaCl in the normotensive and hypertensive strain, independently from the development of hypertension. We conclude that the pretranslational induction of left ventricular ACE with high-NaCl loading may be important both for the regulation of cardiac angiotensins and kinins and for local therapeutic ACE inhibition in the heart during high-salt status.

    Notes: Abstract MAG1 and DDI1 are two divergently transcribed DNA damage-inducible genes from Saccharomyces cerevisiae. Previous studies have shown that MAG1 induction requires an upstream activating site (UAS) located between nucleotides −376 and −330. Here we show that a 24-bp oligonucleotide from within the UAS MAG1 region forms a sequence-specific DNA-protein complex with partially purified proteins from S. cerevisiae. Point mutations introduced into the 24-bp oligonucleotide inhibited the formation of the DNA-protein complex and decreased the level of induction of MAG1-lacZ. By determining the transcription and translation start points of both MAG1 and DDI1, an interesting, indeed unprecedented feature of genome organization in eukaryotes was revealed: UAS MAG1 actually lies within the protein-coding region of DDI1. Although tightly linked to each other, and co-induced upon treatment with DNA-damaging agents, DDI1 does not share the UAS MAG1 required for DNA damage induction of MAG1. Furthermore, MAG1 and DDI1 respond differently in the presence of the protein synthesis inhibitor cycloheximide, suggesting that these two genes are regulated by different mechanisms in the absence of de novo protein synthesis.

    Notes: A major cost consideration in the use of anaerobic digestion to convert biomass and waste to utility-grade gas is the expense of separating CO2 from the product gas. Anaerobic digestion has a number of inherent properties that can be exploited to increase the methane content of the gas directly produced by the digester, the most important of which is the high solubility of CO2(40-60 times that of methane) in water under digestion conditions. The methane enrichment concept examined in this study involved the recirculation of a liquid stream from the digester through a CO2 desorption process and the return of the liquid stream back to the digester for absorption of additional CO2 produced by the conversion of organic materials. A steady-state equilibrium model predicted that a digester gas methane content exceeding 94% could be achieved with this scheme using modest recirculation rates provided a desorption process could be designed to achieve a 60+% CO2 removal efficiency in the degassing of the liquid recycle stream. Using fixed-film laboratory digesters operated on synthetic feedstocks, the technique of methane enrichment was tested under pressurized and unpressurized conditions. A 93 + 2% methane gas stream was produced from a volatile-acid-fed bench-scale digester simulating the methanogenic stage of two-phase digestion under conditions of (1) a pH swing achieved without caustic addition that allowed digestion at pH 7. 5 and air stripping at pH 6. 5-7. 0, (2) digester pressurization to 30 psig, and (3) a recycle rate of 0. 33 L/L reactor/day. Significant but lower levels of methane enrichment were achieved with the single-stage digester at the low experimental recycle rate. However, the narrow range among all experiments of CO2 desorption efficiencies achieved in air stripping the recycle stream (35-60% CO2 removal) suggests that comparable methane enrichment-may be achieved with unpressurized single-stage digestion using greater recycle rates. A materials balance analysis of data from an unpressurized, single-stage digester employing no chemical addition and using laboratory degassing efficiencies indicated that 94% methane could be produced at recycle rates of less than 1. 4 L/L reactor/day with a methane loss of less than 2%.

    Notes: A physicochemical model for vapor-liquid equilibrium in multicomponent formaldehyde-containing mixtures (Maurer, 1986; Hasse et al., 1990; Hasse and Maurer, 1991a). is extended to describe enthalpy changes upon vaporization. Predicted enthalpy changes are compared with new experimental results. The measurements were carried out with a thin-film evaporator flow-calorimeter (Liu and Maurer, 1991). More than 80 experimental data points for mixtures of formaldehyde + water and formaldehyde + methanol + (small amounts of water) are reported. The data cover the temperature range from 323 to 363 K (formaldehyde + water) and 312 to 347 K (formaldehyde + methanol) at liquid phase formaldehyde mole fractions up to about 0.3 (formaldehyde + water) and 0.6 (formaldehyde + methanol). Comparisons between predicted and experimental results for the enthalpy change reveal deviations of only a few percent. Further improvements are achieved by fitting some model parameters, which originally were estimated from noncalorimetric data.

    Notes: High-gradient magnetic separation (HGMS) is an existing commercial technology which promises to be an effective means for the large-scale separation of both weakly magnetic and practically nonmagnetic, micron-sized particles. It utilizes the strong magnetic forces created typically by placing filamentary ferromagnetic packing materials inside a separator matrix magnetized by a uniform background magnetic field. In this paper, a practical mathematical model for HGMS is developed for quantitatively predicting the grade and recovery of the separated product and the capacity (concentration breakthrough) of the separator. Computer implementation and experimental verification of the new model for HGMS applied to pilot-scale coal beneficiation are described in Part III.
