    Notes: Summary Histamine content in the rat hypothalamus was lower at 4°C and higher at 31°C compared to that at 21°C. Pretreatment with α-fluoromethylhistidine, a ‘suicide’ inhibitor of histidine decarboxylase, attenuated both the increased level of hypothalamic histamine and rat adaptive behavior at 31°C. Increase of histamine content in the hypothalamus appears to be an important factor contributing to rat adaptive behavior to high environmental temperature.

    Notes: Summary Using an in vivo intracerebral microdialysis method coupled with an HPLC-fluorometric method, we investigated the extracellular level of endogenous histamine in the anterior hypothalamic area of urethaneanaesthetized rats. The basal rate of release of endogenous histamine in the anterior hypothalamic area measured by this method was 0.09 + 0.01 pmol/20 min. When the anterior hypothalamic area was depolarized by infusion of 100 mM K+ through the dialysis membrane or electrical stimulation at 200 μ A was applied through an electrode implanted into the ipsilateral tuberomammillary nucleus, histamine release increased to 175% and 188%, respectively, of the basal level. These increases were completely suppressed by removal of extracellular Ca2+. The basal release of histamine was also suppressed after infusion of 10−6 M tetrodotoxin or i.p. administration of 100 mg/kg of α-fluoromethylhistidine. On the other hand, 3-fold increase in the basal release was observed after i. p. administration of 5 mg/kg thioperamide. These results clearly indicate that both the basal and evoked release of histamine measured by our method are of neuronal origin.

    Notes: Summary A polymer-based, reversed-phase column (VA-C18), prepared by grafting octadecyl chain onto vinyl alcohol copolymer gel, was investigated for its chromatographic characteristics. n-Alkanes and n-alkyl alcohols were found to be retained only by hydrophobic interaction between the solutes and the octadecyl chain. In the case of aromatic hydrocarbons, in addition to the hydrophobic interaction, π-π interaction between the solutes and the based material was elucidated to contribute to the retention. For aromatic tertiary amines which are known to strongly interact with the residual silanol group of the silica-based reversed-phase columns to produce broadened and skewed peakes, the VA-C18 column also retained these substrates strongly by the combination of hydrophobic, π-π, and ionic interactions. In this case, however, symmetrical peaks were observed. From these results, it was determined that in the case of VA-C18, the base material was found not to produce undesirable effect although the solutes interact with the base. Further conclusion obtained was that in reversed phase liquid chromatography, chromatographic properties of base matrix is highly responsible for the overall retention.

    Notes: Summary Separation mechanisms for single-stranded oligodeoxyribo-or oligoribonucleic acid fragments were explored on an Asahipak polyvinyl alcohol gel column (GS-320) by use of sequential isomers of such molecules. Substrates having different base numbers were found to be separated by size-exclusion chromatography while those having the same numbers with different base sequences were isolated by use of the reversed-phase mode. By using those dual modes, a limit for the separation of the samples was found to arise because one mode shifted the peaks of the substrates in the sense opposite to the shift resulting from the other mode and it was found that when substrates had less than nine bases, the solutes eluted separately.

    Notes: Summary Histamine N-methyltransferase (S-adenosylmethionine: histamine N-methyltransferase, E.C. 2.1.1.8) was purified to homogeneity from rat kidney, and antibody was raised against it in guinea pigs. The antibody immunoprecipitated histamine N-methyltransferase. Immunofluorescent histochemical studies with anti-histamine N-methyltransferase antibody as the first antibody and goat antiguinea pig IgG conjugated with fluorescein isothiocyanate as the second, showed the presence of immunoreactive structures in the proximal tubules of rat kidney. The brain showed no immunoreaction with the antibody.
