    Notes: Conidia of glucose oxidase active Aspergillus niger G 13 strain were subjected to the fourth-stage mutagenization using different combinations of mutagens in each stage (stage I - UV radiation + nitrosomethylurea, stage II - UV radiation + ethyleneimine, Stage III - acryflavine, stage IV - UV radiation + N-methyl-N′-nitrosoguanidine). In all, 400 strains which showed higher glucose oxidase activity with our own diffusion data method (were isolated out of 5208 colonies after mutagenization). Nine most active mutants were tested in the submerged culture determining the activity of glucose oxidase in the post-culture liquid which increased from 10.7 to nearly 30%.

    Notes: The production of extracellular glucose oxidase in a submerged culture by a number of auxotrophic, 2-deoxy-D-glucose resistant and protease-less mutants of Aspergillus niger was evaluated. Among the auxotrophic strains, no evident dependence was found between the kind of the nutritional requirements and the level of the glucose oxidase activity. However, the majority of auxotrophs, requiring serine or niacin, showed a higher enzyme activity (from 16 to 680%) than the parent strain. The dynamics of the glucose oxidase synthesis by the free and immobilized mycelium of the most active niacin- mutant of A. niger was also investigated.

    Notes: Controlled porosity glasses (CPG) are materials very widely employed as supports for chromatography and biotechnology. In this paper CPG with epoxy groups have been applied as carrier of FAD, glucose and 2-deoxy-D-glucose. Glucose oxidase (GOD) was highly purified on these sorbents. The purified fraction of GOD was also immobilized on the activated CPG. The presented results suggest the higher stability of immobilized form of this enzyme in relation to the native enzyme.

    Notes: Spores of Aspergillus niger were exposed to a pulsed electric field. After treatment by the electric field, the viability of the conidia of A. niger varied depending on the field strength, pulse width and frequency. In all cases, these parameters reduced the viability rate of the conidia from 2.0 × 107 to a range from 6.2 × 106 to 8.5 × 106 spores/ml (3.1 to 42.6%). After pulse treatment, the conidia were used as the inoculum for citric acid fermentation in shake flasks. The highest increase in citric acid yield (about 1.4-fold) was reached at a field strength of 2.85 kV/cm, a frequency of 1 Hz and a pulse width of 1 ms. When the parameters of the electric field increased there were important changes in the respiration rate of the Aspergillus niger mycelium (48-h-old) after electric shock treatment. The highest consumption of dissolved oxygen (22.9%) in the medium by Aspergillus niger mycelium was observed at an electric field strength of 2.85 kV/cm, a 1 Hz frequency, a pulse width of 1 ms and a 1-min exposure period. It seems that an electric-field stimulation of the conidia prior to inoculation may offer an important method of improving the efficiency of citric acid. The treatment of the conidia is both simple from the technical point of view and extremely rapid.

    Notes: Optimum conditions for citric acid production by Aspergillus niger in submerged culture were established. Some correlation was found between the ability of the fungus to produce citric acid and its capability to accumulate extracellular acid phosphatase. Mutagenization and passage on sodium citrate (10-25%) medium gave rise to eleven A. niger strains able to grow when the concentration was 25%. Two mutants showed an increase in citric acid production (8.8-25.7%), accompanied by the highest activity of acid phosphatase (43.1-46.6%).

    Notes: A total of 65 yeast strains were screened for their ability to grow and ferment lactose in a standard DURHAM tube test at 30 °C. Based on the kinetic parameters for lactose and whey lactose fermentations in shake flask cultures, the strain Candida psedotropicalis 65 was chosen for further studies.Some of the cultural parameters affecting ethanolic fermentations on lactose were standardized. At an initial lactose concentration of 100-120 g/l in the medium containing concentrated whey or lactose, at 40 °C and within 48 h, the selected strain reached an ethanol concentration of 41-59 g/l, an ethanol productivity of 1.3-3.0 g/l/h, a lactose consumption of 99%, an ethanol yield 0.4-0.49 g/g and a biomass yield of 0.027 g/g.
