    Description / Table of Contents: Zusammenfassung Mit Fraktion H ließ sich bei Kaninchen ein wirksames Antiserum (AFHS) erzeugen, das bereits in einer Menge von 0,05 ml 1000 μg Fraktion H vollständig absorbierte und die lipolytische Wirkung vollkommen aufhob. Der Antikörper (AK) im AFHS, der sich als IgG isolieren ließ, hatte in einer Konzentration von 0,08 mg den gleichen Effekt. Zwischen AFHS und den lipolytisch wirksamen Polypeptidhormonen ACTH, STH und α-MSH ließen sich keine Präcipitationslinien nachweisen. Während Fraktion H mit AFHS drei Präcipitationslinien ergab, zeigte sich bei der weiter gereinigten Fraktion J nur noch eine Präcipitationslinie mit APHS, die mit einer der drei Linien von Fraktion H das Bild der Vollidentität bot.
    Notes: Summary 0.05 ml of a potent antiserum (AFHS), we gained by immunisation of rabbits with fraction H from pig pituitaries, totally absorbed 1000 μg fraction H and completely blocked their lipolytic effect in vivo. The same could be achieved by the isolated antibody (IgG) in a concentration of 0.08 mg. There were no precipitation lines between AFHS and the polypeptides ACTH, STH and α-MSH. Instead of three lines between fraction H and APHS the purified fraction J gave only one precipitation line with APHS, which was identical with one of the lines of fraction H.

    Notes: Summary To establish whether additional therapy with 3-hydroxy-3-methylglutaryl (HMG) coenzyme A (CoA) reductase inhibitors enhances the low-density lipoprotein (LDL) cholesterol lowering effect of LDL apheresis with immunoadsorption in the treatment of patients with familial heterozygous hypercholesterolemia and coronary artery disease we studied eight patients initially on immunoadsorption therapy alone for 3 years. The adding of HMG CoA reductase inhibitors decreased pretreatment LDL cholesterol from 6.76±0.98 to 4.97±0.98 mmol/l and posttreatment LDL cholesterol from 2.33±0.80 to 1.94±0.67 mmol/l and increased pre- and posttreatment high-density lipoprotein (HDL) cholesterol by 0.08 and 0.13 mmol/l respectively. The LDL/HDL ratio was reduced from 4.0 to 2.8 (prior to any therapy the ratio was 13.4). The increase in LDL cholesterol between weekly treatments was less steep under the combined therapy. At the same time the treated plasma volume during LDL apheresis could be decreased from 5070±960 to 4370±1200 1200 ml. We conclude that in patients with severe familial heterozygous hypercholesterolemia LDL apheresis should be combined with HMG CoA reductase inhibitors.

    Description / Table of Contents: Zusammenfassung An Hanford-Miniaturschweinen wurde die In vivo-Wirkung des von uns aus Schweinehypophysen gewonnenen lipidmobilisierenden Peptid A und seiner weniger gereinigten Vorstufe Fraktion J in einer Dosierung von 100 µg/kg i.v. untersucht: Peptid A (FFS-Anstieg um 2,13 ± 0,2 mval/l) war stärker wirksam als Fraktion J (1,25 ± 0,23 mval/l). Die lipolytische Wirkung war bereits 5 min nach Injektion nachweisbar. Beide Substanzen führten zwar zu einem signifikanten Blutzuckeranstieg; nach Peptid A-Gabe war jedoch 60 min nach Injektion ein zwischenzeitlicher Glucoseabfall unter Ausgangswerte festzustellen, der mit dem Maximum der FFS zusammenfiel.
    Notes: Summary The effect of pig pituitary lipidmobilizing Peptide A and its precursor fraction J was studied in Hanford minipigs. Within 5 min after the intravenous injection of 100 µ/kg of the polypeptides free fatty acids and free glycerol were elevated; the maxima were obtained 60 respectively 30 min p.i. Peptide A was more active than fraction J (2.13 ± 0.21 respectively 1.25 ± 0.23 mval/l FFS increment compared to the initial values). Glucose was significantly elevated 210 min after the injection of either peptide, but peptide A produced a decrease of glucose 60 min p.i. when FFS were maximal.
