    Notizen: Abstract Maize root mitochondrial have been subfractionated after osmotic rupture. A calcium-calmodulin-dependent NAD-kinase activity has been shown to be present in both inner and outer membrane fractions. Cytochrome c-reductase activities are also associated with outer and inner membrane fractions but whereas the former is entirely insensitive to 50 μmol·1-1 antimycin A the latter is reduced by 60% in its presence. This residual antimycin A-insensitive cytochrome c-reductase activity cosediments with the major portion of NAD-kinase activity and equilibrates in sucrose gradients at densities around 1.146 g·cm-3. Rate zonal centrifugation with renografin allows an excellent separation of both cytochrome c-reductase and NAD-kinase activities. We have no evidence for allocating NAD-kinase activity to endo- or plasma membranes.

    Notizen: Abstract Cell wall and thus cellulose microfibril formation in the presence of Congo red or Calcofluor white by Oocystis solitaria autospores was investigated ultrastructurally and chemically. The prevention of microfibril formation by both substances is accompanied by drastic changes of microfibril synthesis and orientation as well as the morphology of plasma-membrane-associated E-face terminal complexes. Removal of Congo red and Calcofluor white from the culture medium results in the recovery of microfibril formation, of a normal patterned microfibril arrangement, and of terminal complexes with extending microfibril imprints.

    Notizen: Summary Polyclonal antibodies raised against the tonoplast and plasma membrane H+-ATPases of corn were used to probe Western blots lof clathrin-coated vesicles purified from zucchini hypocotyls and bovine brain. Antibodies to two tonoplast H+-ATPase subunits recognized their respective polypeptides in purified zucchini coated vesicle preparations. Antibody raised against the plasma membrane H+-ATPase failed to cross-react with zucchini coated vesicles, even though it recognized a 100 kDa polypeptide in purified zucchini plasma membrane preparations. These results suggest that zucchini coated vesicles contain a vacuolar-type rather than a plasma membrane-type H+-ATPase. Antibody to the corn 70 kDa subunit also cross-reacted with a 70 kDa band in bovine brain coated vesicle preparations. Surprisingly, the corn plasma membrane H+-ATPase antibody recognized a 100 kDa polypeptide in the bovine brain coated vesicle preparation. However, at least half of the 100 kDa antigen could be dissociated from the membrane by treatment with 2.0 M urea, indicating that it may represent one of the 100–110 kDa coat assembly proteins. A comparable protein appears to be absent from zucchini coated vesicles.
