    Notizen: Summary Influence of food on the serum concentration and kinetics ambenonium chloride (AMBC) has been examined in thirteen patients with myasthenia gravis (MG). Mean serum concentrations and Cmax during fasting were higher than those in the non-fasting state. The AUC (0–3 h) was also about four-times larger. The drug effects versus the serum concentration were observed to be anti-clockwise or clockwise. The effective range of the Cmax varied between patients. The unexpected increase in Cmax led to adverse muscarinic actions of AMBC, when the condition was changed from the non-fasting to the fasting state. It is recommended that the dose be changed during non-fasting treatment when adjusting the optimum regimen for patients myasthenia gravis. Patients must be advised to keep to the dosing and dietary schedule in order to avoid unexpected adverse actions to AMBC.

    Notizen: Abstract Objectives: The present study was conducted to compare pharmacokinetic behaviors of nicardipine enantiomers given in different doses with different formulations of racemic nicardipine in healthy volunteers. Methods: One or two 20-mg racemic nicardipine tablets, and a 40-mg sustained-release capsule of nicardipine were administered to eight healthy volunteers in a cross-over fashion and pharmacokinetic parameters were evaluated. Enantiomer concentrations were determined by GC-MS combined with chiral stationary phase HPLC. Results and conclusions: Serum concentration of (+)-nicardipine was approximately 2–3 times higher than that of (−)-nicardipine in 20- and 40-mg doses of conventional formulations and a non-linear increase in bioavailability with dose was demonstrated. The value for AUC of (+)-nicardipine was approximately 2.3–2.8 times greater than that of the (−)-nicardipine (P 〈 0.05) when 20 and 40 mg racemic nicardipine were administered in a conventional preparation. Relative bioavailability of the sustained-release preparation vs the conventional preparation was 28% and 44% for (+)- and (−)-nicardipine, respectively, for the 40-mg dose.

    Notizen: Summary A callus line of white clover capable of forming numerous meristemoids (meristematic cell masses) has been selected and subcultured on agar B5 medium containing 0.5 mg/l 2,4-D and 0.5 mg/l kinetin for three years. The meristematic callus was successfully cryopreserved by vitrification and subsequently regenerated plants. Preculturing callus in liquid B5 medium containing 0.6 M sorbitol at 25°C for 16 hr was essential to the process. Precultured samples (50 mg) were transferred to a 1.8 ml plastic cryotube and then 1 ml of a highly concentrated cryoprotective solution (designated PVS2) was added and mixed. After treatment with PVS2 at 25°C for 7 min or 0°C for 20 min, the sample was directly plunged into LN. After rapid warming, PVS2 was drained from the cryotubes and replaced twice with liquid B5 medium containing 1.2 M sucrose. Samples were transferred onto filter disc over agar B5 medium. Some surviving cells in the cryopreserved meristematic callus proliferated and produced new meristemoids. After 30 days the meristematic callus was transferred onto hormone-free MS agar medium. The meristemoids developed directly into shoots and spontaneously formed roots. Plant regeneration efficiency expressed as a percent of control amounted to about 90%. This vitrification method appears promising as a routine method for cryopreserving meristematic callus of white clover.
