Proteins and peptides were separated in the reversed-phase mode on microcolumns packed with nonporous octadecyl-group bonded silica gel with an average particle diameter of 4.5 or 20 μm. Separation columns were prepared from glass-lined stainless steel tubing of 39 or 56-mm × 0.5-mm i.d. An artificial mixture of several proteins could be separated within 1 min by split-flow gradient elution. The system was applied to the separation of a triptic digest of mouse amyloid A. On-column sample enrichment allowed direct injection of 2-5 μL of sample solution, leading to improved detectability. The limits of detection were ≤ 1 μg mL-1.
