Notizen: Two static x-ray imagers (SXI) will be used to monitor beam pointing on all target shots in the National Ignition Facility. These pinhole-based instruments will provide time integrated two-dimensional images of target x-ray emissions in the energy range between 2 and 3 keV. These instruments are not DIM based and will view along dedicated lines of sight from near the top and bottom ports of the target chamber. Beams that miss or clip the hohlraum laser-entrance holes will produce x-ray emission on the ends of the hohlraum, indicating improper beam pointing and/or target positioning. The SXIs will also be used to quantify beam focusing and pointing by producing x-ray images of dedicated test targets irradiated by focused beams at precalculated positions. A proposed design is presented, along with supporting data from NOVA target experiments. © 2001 American Institute of Physics.

Notizen: Summary. The capacity of the human placenta to degrade 125I-labelled arginine vasopressin (125I-AVP) was studied in vitro using a dual circuit perfused lobule preparation. Seven placentas were perfused with the perfusate on the maternal side of the lobule containing 125I-AVP at the upper limit of the physiological range. On average, over a 30-min period, 48% of the 125I-AVP appeared to have been metabolized. With one exception, a patient whose labour was augmented with intravenous oxytocin, no 125T-AVP apparently crossed the placental lobule to the fetal circulation. These data indicate that the human placenta has a considerable capacity to degrade AVP.

Notizen: Plasma levels of diphenylhydantoin (DPH) and the 24 hour urinary excretion of both DPH and its major metabolite 5-(p-hydroxyphenyl)-phenylhydantoin (HPPH) were measured serially during pregnancy in a group of five unselected epileptic patients. Plasma DPH levels fell and on average were some 40 per cent lower at term than they had been in early pregnancy. With one exception, the patients were well controlled during pregnancy but two patients who regularly had plasma DPH levels below 10 μ/ml both suffered seizures in the puerperium. There was no convincing change in the excretion of either DPH or HPPH which together accounted for less than half the administered dose. Gestational length exceeded 41 weeks in 4 of the 5 patients studied but all were delivered of normal healthy infants, of average birthweight, with no congenital defects or clotting disorders.

Notizen: A cross-sectional study of γ-glutamyl transferase (γ-GT) excretion in 316 healthy pregnant women showed the enzyme excretion (mU per mmol creatinine) was significantly higher than that of a non-pregnant control group. The increase was apparent before the end of the first trimester and continued thereafter. These changes may be indicative of hypertrophy and/or hyperplasia of the nephron and be part of the overall renal adaptation to normal pregnancy. Even higher levels were recorded in three obstetric patients in whom renal complications were to be expected and this may indicate a specific type of renal damage.

Notizen: The diagnostic instrument manipulator (DIM) provides a diagnostic platform to insert and retract a variety of instruments into and out of the National Ignition Facility target chamber. The DIM is a two-stage telescoping system, designed to fit on any of the DIM designated diagnostic ports on the target chamber, and will provide precision radial positioning, pointing, and alignment-to-target capability. The DIM provides a standard set of utilities, and cables to support the operation of instruments that require insertion into the target chamber. The DIM provides for positioning of diagnostic packages, and enables exchange of manipulator diagnostics between fusion laboratories. Principal design requirements for the DIM are presented. A half-length prototype of the DIM was designed and fabricated by Atomic Weapons Establishment in England and is being tested at Lawrence Livermore National Laboratory. The results of this testing are presented. © 2001 American Institute of Physics.

Notizen: Summary. A technique is presented for the preparation of cultures of replicating human trophoblast cells from term placentas. The adherent cells obtained were very slow growing (doubling time 12.5 days) as measured b y the rate of increase in cell protein, [14C]-leucine uptake and cell number. Cells from individual placentas have been maintained in continuous culture for up t o 1 year (10–12 passages) and have been successfully recultured after storage in liquid nitrogen. Cultured cells showed positive immunofluorescent staining for human placental lactogen, human chorionic gonadotrophin, transferrin and type IV collagen. The adenine nucleotide content indicated that energetically the cells were in balance even after prolonged culture.

Notizen: The rise in the plasma level of folate following an oral load of 15 μg. of folic acid per kg. body weight was examined in a serial study of 12 healthy pregnant women. The average rise in pregnancy was similar to that postpartum and in two male control subjects, but there was a suggestion of a more sluggish rise in early compared to late pregnancy. Individual variation was considerable and greatly exceeded any possible effect of pregnancy. These data provide no evidence that pregnant women suffer an impairment of folate absorption.

Notizen: The accumulation of tritium-labelled folate in the product of conception was examined in eight women undergoing hysterotomy for termination of pregnancy. Folic acid containing a small quantity of tracer material (50n Ci/μg.) was injected intravenously 20 minutes before operation in 4 patients but 24 hours beforehand in the remainder. After operation, maternal blood, placenta, fetal blood, and fetal liver were examined for radioactivity as was the daily urine output during the patient's postoperative hospital stay. The results indicated that only 1/2 percent of the injected material accumulated in the product of conception—uptake was primarily by the fetal liver and no large placental pool of folate was involved.Approximately half of the injected load was excreted in the urine—the majority in the first 24 hours—and the remainder was presumed to have been retained by maternal tissues. Bioassay with L. casei and chromatography on TEAE cellulose suggested that extensive metabolism of the injected material had taken place and that fetal uptake might be more efficient than indicated by counting radioactivity alone.
