    Notes: Abstract: Synaptosomes from rat forebrain can easily be isolated by combining centrifugation with partition in an aqueous two-phase system composed of dextran T500 and polyethylene glycol 4000 in which synaptosomes have an extreme affinity for the upper phase. The fraction thus obtained has been characterized by electron microscopy and biochemical markers for synaptosomes and some other cell components. The contamination by microsomes, free mitochondria, and myelin was 4.4, 3.2, and 0.1%, respectively. The morphometric analysis of the electron micrographs shows that 〉60% of the structures are synaptosomes. This preparation of the isolation procedure is remarkably short (〈1 h), formance as assayed by their respiratory activities and ATP level in the absence and presence of depolarizing agents. Synaptosomes prepared by phase partition release the neurotransmitter glutamate in a Ca2+-dependent manner. The duration of the isolation procedure is remarkably short (〈1 h), no ultracentrifuge is required, and the method can be applied for small- or large-scale preparations.

    Notes: This paper is concerned with the microstructure observed in colloidal dispersions of charged polymeric particles and their interaction potential at very low ionic strength. Both aspects are probed with the aid of new experimental data for nonaqueous media. The structure factor of ordered dispersions (with methanol–water and ethanol–water solvent mixtures) was determined using static light scattering. A different behavior (in the studied range of molar fraction) depending on the alcohol type is found. These results are analyzed assuming a Yukawa potential and applying integral equation theories (the Ornstein–Zernike equation and the HNC closure). The obtained effective charge is almost constant for methanol–water mixtures, whereas a decrease with the alcohol molar fraction is observed for ethanol–water. In order to account for these effects, a charge renormalization procedure is applied and discussed. The surface charge turns out to be an unsuitable input parameter for such an approach. © 1999 American Institute of Physics.

    Notes: It is of interest to determine the characteristic length scale that determines giant magnetoresistance (GMR). In order to understand this behavior, GMR multilayers of Co/Ru and Co/Cu have been studied at a temperature of 4.2 K. The total thickness of Co/Ru multilayers has been varied from 96 to 1654 Å and Co/Cu from 77 to 2712 Å by increasing the number of bilayers (N). It has been observed that GMR increases with the number of bilayers and more than 20 bilayers for Co/Ru and 50 for Co/Cu are needed to reach the saturation value. © 2001 American Institute of Physics.

    Notes: The knowledge of the potential between interacting colloidal particles is essential to describe and control the aggregation processes in which they are involved. However, interaction forces also bring out spatial ordering phenomena, which has been used to obtain helpful information on them in three-dimensional dispersions. In this paper we look into this possibility for colloidal monolayers spread at the water–air interface. The radial distribution function has been determined for an assembly of negatively charged polystyrene particles at low concentration and without an additional electrolyte. A simple inversion scheme based on the HNC closure has been proved to give reliable information on u(r), providing that particles are not strongly correlated. Apart from the expected repulsive forces, the application of the inversion procedure suggests the possibility of a long-ranged attractive interaction. © 2001 American Institute of Physics.

    Notes: Abstract: The recent finding that several point mutations in the gene encoding for the microtubule-binding protein tau correlate with neurological disorders has heightened interest in the mechanisms of destabilization of this protein. In this study the functional consequences of the tau mutation R406W on the interaction of the protein with microtubules have been analyzed. Mutated tau is less phosphorylated than its normal counterpart at serines 396 and 404. Furthermore, the phosphorylated mutant protein is unable to bind to microtubules, and, as a consequence, microtubules assembled after transient nocodazole treatment in the presence of this tau variant contain only unmodified tau and appear to form more and longer bundles than those assembled in the presence of wild-type tau. We propose that phosphorylated tau, unbound to microtubules, could accumulate in the cytoplasm.

    Notes: Summary Analytical results are given for whey powders prepared on a commercial or semi-commercial scale by three companies. Altogether, five preparations enriched in β-lactoglobulin, four whey protein isolates and a fraction enriched in α-lactalbumin were analyzed for protein composition, including %β-lactoglobulin, α-lactalbumin, bovine serum albumin, casein (glyco) macropeptide and the main triglycerides. Protein composition was determined by high pressure gel permeation and reversed phase liquid chromatography and by capillary zone electrophoresis. The extent of modification of the native β-lactoglobulin structure was also measured through the degree of lactosylation and the fraction of accessible free sulphydryl groups. One significant finding was that the calculated recovery of protein following quantitation of the chromatogram or electropherogram was seldom above 90% and occasionally below 60% of that loaded onto the column or capillary, raising doubts as to the reliability of the analytical results. Extrapolation by linear regression to 100% recovery allowed estimates to be made of the true β-lactoglobulin composition of the samples. The nine samples could be placed into three distinct groups with estimated true β-lactoglobulin weight % of 70.9 ± 1.1, 62.0 ± 3.4 and 39.5 ± 4.9. Physico-chemical properties of the group of samples are reported elsewhere (Holt et al., 1999).

    Notes: Summary Acquired cutis laxa (ACL) is an uncommon elastolytic disorder of unknown aetiology. In rare instances. ACL has been reported in association with autoimmune diseases and dermal deposit of immunoglobulins, suggesting that destruction of elastic tissue may be immunologically mediated. We report a 35-year-old man with generalized acquired cutis laxa (GACL) associated with a persistent papular erythematous eruption that histopathologically showed some resemblance to dermatitis herpetiformis. A marked reduction and degeneration of dermal elastic fibres was noted in biopsies from loose-hanging skin. Direct immunofluorescence from non-inflammatory loose skin revealed granular immunoglobulin A (IgA) deposits at the basement membrane zone and fibrillar IgA deposits in the dermal papillae. IgA deposits were also observed on the elastic fibres of the reticular dermis. Electron microscopy of skin from the submammary fold revealed fragmented elastic fibres, partial absence of peripheral microfibrils and abundant neutrophils, some of which were degranulated and adjacent to elastic fibres. Immunoelectron microscopy of an erythematous papule revealed IgA deposits around dermal elastic fibres. Antigliadin, antireticulin and antiendomysium antibodies were present. Jejunal biopsies showed a gluten-sensitive enteropathy. A possible IgA-mediated immune mechanism for the development of GACL in our patient is suggested.

    Notes: Abstract: A clinical trial of the pyrethroid permethrin in 1% lotion was performed on 20 children. Ten were treated (or 10 minutes and 10 for 2 hours. All lice washed off the heads of children treated for 2 hours were dead immediately after treatment. Twenty-four percent of the lice removed from one child treated for 10 minutes were still alive 2 hours later. The ovicidal effect was similar in both groups, with egg mortality 88% and 90% following 10-minute and 2-hour treatment, respectively. The mortality of untreated control eggs was 14% and 11%, respectively.
