    Notes: The Ti–Si system is chosen as a model system to study the mechanical alloying of elemental powder blends in which one of the components is brittle. In addition, the stability of intermetallic compounds upon milling is investigated, in order to interpret the metastable phase formation during the milling process. Detailed x-ray investigations show that during mechanical alloying, a solution of Si in Ti preceeds the formation of amorphous or intermetallic phases. The selection of phases formed, as well as the final state, are influenced by the overall composition of the powder blends, and by the milling procedure applied. However, predictions can be made by considering the free energy curves of the stable and metastable phases in the Ti–Si system, which have been calculated by the CALPHAD method. Milling of the intermetallic compounds generally leads to the same final state as the mechanical alloying of elemental powder blends with corresponding compositions. These results demonstrate that the intermetallic compounds are energetically destabilized upon milling, allowing for phase transformations into metastable phases, such as amorphous or metastable crystalline alloys. The energetic destabilization of intermetallic compounds mainly originates from the chemical disorder caused by the milling process.

    Notes: The solid-state reaction in Ti/Cr multilayered diffusion couples at temperatures between 450 and 650 °C was investigated by x-ray diffraction. It was found that an amorphous phase is formed at a concentration around 55 at. % Cr. The results are in excellent agreement with those obtained from mechanically alloyed powders, and with the corresponding metastable phase diagram calculated by the calculation of phase diagrams method. The observations strongly suggest that inverse melting is possible in the Ti-Cr system at around 55 at. % Cr.

    Notes: An endoreversible three-heat-source refrigerator only affected by thermal resistance, like a reversible three-heat-source refrigerator, may be treated as a combined cycle of a two-heat-source engine driving a two-heat-source refrigerator. The theory of finite time thermodynamics in two-heat-source cycles is then used to analyze it and derive its basic optimum relation. Thus, the fundamental effect of thermal resistance on the optimal performance of a three-heat-source refrigerator is expounded. The conclusions obtained here are more realistic than those of classical thermodynamics. They provide some new theoretical bases for further exploitation of the three-heat-source refrigeration apparatus that applies to "cheap'' heat sources, such as solar energy, geothermal energy, waste heat, and so on.

    Notes: The luminescence from a planar metal–alumina–metal structure was investigated under ac voltage application in vacuum. Two kinds of electrode contacts, i.e., with/without sputtered gold film, showed quite different optical phenomena. For the nonsputtered alumina samples, no luminescence was detected until extremely irregular light pulses resulted from partial discharges under higher voltage. While for the sputtered samples, from far lower applied voltage, there was faint but stable light emission due to electroluminescence (EL) observed. Based on the band theory of solids, we proposed a model to explain the interesting phenomena qualitatively. Taking account of the sputtering process, a marked reduction of the potential barrier between electrodes and alumina surface would result. Thus, charge carriers can be easily injected from the electrodes into the surface layer of alumina, and EL will emit due to the radiative recombination of electrons and holes. It is considered that EL has a significant influence on the developing process of surface flashover. © 2001 American Institute of Physics.

    Notes: Cytotoxic T lymphocyte-associated molecule-4 (CTLA-4) is a receptor present on T cells that plays a critical role in the downregulation of antigen-activated immune responses. CTLA-4 interacts with the ligands CD80 and CD86 on antigen-presenting cells (APC), and also directs the assembly of inhibitory signalling complexes that lead to quiescence or anergy. In this study, we show that human monocytes constitutively express CTLA-4. About 3% of monocytes expressed CTLA-4 on the cell surface, whereas the intracellular expression was higher and present in about 20% of the monocytes. The sequences of the cDNAs from human monocytes were identical to the sequences of CTLA-4 from T cells. Expression of CTLA-4 was also confirmed in the activated myelomonocytic cell lines U937 and THP-1. Monocytes, but not T cells, activated by interferon (IFN)-γ also secreted soluble CTLA-4 in vitro. The CTLA-4 expression was upregulated upon treatment with phorbol 12-myristate 13-acetate (PMA) and IFN-γ. This increased expression could be partially abolished by staurosporine, an inhibitor of protein kinase C (PKC). Ligation of CTLA-4 in the monocyte-like cell-line U937 with antibodies against CTLA-4 partially inhibited the proliferation of cells and the upregulation of cell-surface markers CD86, CD54, HLA-DR and HLA-DQ induced by IFN-γ and Staphylococcus aureus, Cowan I strain (SAC). Ligation of CTLA-4 suppressed the PMA-stimulated activation of transcription activator protein 1 (AP-1) and nuclear factor (NF)-κB in the U937 cell line, indicating the involvement of an inhibitory signal transduction. These data provide the first evidence that CTLA-4 is constitutively expressed by monocytes and thus might be important for the regulation of immune mechanisms associated with monocytes.

    Notes: Dendritic cells (DCs) have been identified as effective antigen-presenting cells (APCs). We demonstrate that extracellular matrix (ECM), hyaluronic acid (HA) and chondroitin sulphate A (CSA), in the presence of granulocyte-macrophage colony-stimulating factor (GM-CSF), can rapidly promote the differentiation of monocyte-derived immature DCs, as characterized by the remarkable upregulation of human leucocyte antigen (HLA-DR), CD40, CD54, CD80 and CD86 expression to levels higher than those in the DCs generated by culturing with GM-CSF and interleukin (IL)-4 for 7 days and aggregation of the cells within 48 h. The upregulation of expression of HLA-DR, CD40, CD54, CD80 and CD86 was dose-dependent. Further studies showed that HA and CSA were able to augment nuclear factor (NF)-κB activity, as determined by gel mobility shift assay and promote protein phosphorylation. Inhibition of NF-κB by pyrolidine dithiocarbamate and sodium salicylate, and serine-threonine and tyrosine kinase by starosporine as well as phosphatidylinositide-3-kinase (PI-3-K) by wortmannin could prevent the effects of HA and CSA on the expression of HLA-DR, CD40, CD80 and CD86 in various degrees. Thus, our data demonstrate that HA or CSA can effectively and rapidly promote the differentiation of immature DC, suggesting that HA and CSA may possess a potential capacity in regulating immune responses.
