    Notizen: Abstract A proteolytic system was found in all the species of dairy Propionibacterium. Two types of activities could act on [14C] \gb-casein or [14C] \gas1-casein. The first was the highest at the beginning of the exponential growth phase, tightly linked to the cells and hydrolysed preferably \gb-casein. The second was released at the end of growth and acted similarly on both substrates tested. This activity was located in the cell membrane of these cheese-ripening bacteria. The enzyme could be gently extracted from the cell by incubation in Ca2\s+ \t- or Mg Mg2\s+-free buffer.

    Notizen: Abstract The adaptation to osmotic stress in Propionibacterium freudenreichii subsp. shermanii was investigated by using natural-abundance 13C nuclear magnetic resonance spectroscopy. Cells incubated either in a standard laboratory medium or in a medium designed to simulate the physicochemical conditions of Swiss-type cheese were found to accumulate different levels of osmotic-stress-protectant molecules. Proline, betaine, trehalose and glutamate were found simultaneously. Moreover, two types of polysaccharides were in evidence in this strain. Lactate catabolism was not mainly directed towards cell growth requirements and organic acid production but also towards biosynthesis of osmolytes requested for adaptation in a cheese environment. The possible involvement of such type of metabolite accumulation in the main cheese-ripening bacteria in Swiss-type cheeses is discussed.

    Notizen: Summary Dynamic modelling of a continuous lactose fermentation system for control and optimisation of operation has been carried out. The bioreactor used was coupled with an ultrafiltration module and an electrodialysis unit. A unstructured model taking into account cell growth, substrate consumption, and metabolite (lactic acid) production, has been analysed. The metabolite production model was represented by the Luedeking-Piret equation, modified in order to improve the description of lactic acid production at low dilution rate.

    Notizen: Summary An off-line parameter estimation method has been developed to predict the dynamic behaviour of a continuous lactose fermentation system. The model used is an unstructured model taking into account cell growth, substrate consumption, and metabolite production (lactic acid). This method, based on the Hooke-Jeeves non-linear-programming technique, results in a good estimation of the biological parameters of the model, and so gives a better understanding of the different phenomena involved in lactose fermentation.

    Notizen: Summary Erwinia chrysanthemi cells were used to study the possibility of producing bacterial enzymes in a bioreactor coupled with a membrane filtration unit. Continuous fermentations with total cell recycle failed to give good production of pectate lyase (PL). Enzymatic, mechanical and physico-chemical damages were involved in this phenomenon. With a sequential recycle mode, we obtained productivity of 1.5 units·h−1·1−1 with a high PL concentration. Protease accumulation occurred when the bioreactor was coupled to a filtration unit. Moreover we have observed no loss of activity due to high shear stress caused by pumping.
