    Notizen: Summary The development of granular sludge in laboratory-scale upflow anaerobic sludge-blanket reactors was studied. Acetate was supplied as sole carbon source in order to select the acetotrophs Methanosarcina and Methanothrix. These microorganisms are dominant in methanogenic ecosystems and their ratio seems to control the speed of granulation. Changing the ratio of the above species was followed on the basis of their different F 420-coenzyme content. Five reactors were operated at the same hydraulic retention time but at different feed substrate concentrations. We found that granulation takes place only in acetate-fed systems but this process was slower and the resultant granules looser and less stable than those developed on sugar-starch substrate. In the range of feed acetate levels examined (0.5−0.3 g/1) higher concentrations of feed caused faster granulation of the sludge bed and, presumably, of the microbial population, and resulted in larger granules containing sludge that settled more readily. We found no evidence for selection pressure at substrate concentrations below 0.5 g/1 acetate in the reactor.

    Notizen: Two-phase anaerobic digestion of liquid swine manure has been developed with options for single-cell protein (SCP) or methane production. In the acidogenic phase at two to four days retention time, and 2.5-7.0% dry matter (DM) concentration, 8-46% of the volatile solids was solubilized. Maximum reactor capacity was 3.86 g/L at 7.0% DM concentration, but optimal operation was achieved at 4.5% DM concentration at four days retention time. The second methanogenic phase was operated continuously and had a maximum specific methane production rate of 0.70 L/L day at 12 days retention time. With recirculation, the rate was 1.16 L/L day at 8.5 days retention time with 52.7% conversion of organic matter. Maximum digestibility was 66% of the lignin free organic matter.

    Notizen: Pressed and wilted samples of sweet sorghum [Sorghum bicolor (L.) Moench var. Rio] were ensiled for periods up to 155 days. A kinetic study of the biochemical changes which occurred during ensiling showed that in wilted sorghum ensilage invert sugars and mannitol levels collectively were maintained at 65% of the original ferment able sugar content of the sorghum. The acidic environment produced by ensiling also served as a pretreatment that resulted in enhanced yields of reducing sugar when the sorghum was contacted with cellulolytic enzymes. The quantity of sugar obtained from enzymatic hydrolysis more than compensated for carbohydrate used by organisms during the ensiling process. Both Saccharomyces uvarum and Clostridium acetobutylicum were able to ferment a medium constituted from pressed sorghum juice and the solution resulting from enzymatic hydrolysis of sweet sorghum ensilage.
