    Notizen: Abstract: Metallothionein (MT) protein and mRNA levels were monitored following exposure of rat neonatal primary astrocyte cultures to methylmercury (MeHg). MT-I and MT-II mRNAs were probed on northern blots with an [α-32P]dCTP-labeled synthetic cDNA probe specific for rat MT mRNA. MT-I and MT-II mRNAs were detected in untreated cells, suggesting constitutive MT expression in these cells. The probes hybridize to a single mRNA with a size appropriate for MT, ∼550 and 350 bp for MT-I and MT-II, respectively. Expression of MT-I and MT-II mRNA in astrocyte monolayers exposed to 2 × 10−6M MeHg for 6 h was increased over MT-I and MT-II mRNA levels in controls. Western blot analysis revealed a time-dependent increase in MT protein synthesis through 96 h of exposure to MeHg. Consistent with the constitutive expression of MTs at both the mRNA level and the protein level, we have also demonstrated a time-dependent increase in MT immunoreactivity in astrocytes exposed to MeHg. The cytotoxic effects of MeHg were measured by the rate of astrocytic d-[3H]aspartate uptake. Preexposure of astrocytes to CdCl2, a potent inducer of MTs, completely reversed the inhibitory effect of MeHg on d-[3H]aspartate uptake that occurs in MeHg-treated astrocytes with constitutive MT levels. Associated with CdCl2 treatment was a time-dependent increase in astrocytic MT levels. In summary, astrocytes constitutively express MTs; treatment with MeHg increases astrocytic MT expression, and increased MT levels (by means of CdCl2 pretreatment) attenuate MeHg-induced toxicity. Increased MT expression may represent a generalized response to heavy metal exposure, thus protecting astrocytes and perhaps also, indirectly, juxtaposed neurons from the neurotoxic effects of heavy metals.

    Notizen: Abstract: Exposure of primary astrocyte cultures to isosmo-tic ethanol from 10-100 mA/ led to both swelling of the cells and release of [3H]taurine and r[3H]aspartate. Exposure to hyperosmotic ethanol, in the same concentration range. caused neither swelling nor release. Release was inhibited by the anion transport blocker L-644,711, already shown to inhibit amino acid release evoked by hyposmotic or high-potassium medium, conditions that also cause astrocytic swelling. Ethanol-induced release generally showed a decline in response to successive exposures to ethanol, and release was not dependent on extracellular calcium. Thus, the characteristics of swelling-induced release of amino acids by isosmotic ethanol seem to correspond to those of swelling-induced release from astrocytes due to exposure to hypotonic or high-K+ media. We discuss whether such effects may contribute to CNS damage after head injury and stroke.

    Notizen: Abstract The uptake of 3H-labelled 5-hydroxytryptamine (5-HT, serotonin) norepinephrine ([3H]NE), and 3,4-dihy-droxyphenylethylamine ([3H]dopamine, [3H]DA) was studied in primary astrocyte cultures prepared from the cerebral cortex, corpus striatum, and hippocampal regions of neonatal rat brain. Na+-dependent uptake showed marked regional differences. For [3H]5-HT the magnitude of uptake was corpus striatum ≥ cerebral cortex ≥ hippocampus, whereas for [3H]NE the order was hippocampus ≥ corpus striatum ≥ cerebral cortex. For [3H]DA, only the hippocampal cultures showed significant Na+-dependent uptake. [3H]5-HT uptake was specifically inhibited by 10-7M flu-oxetine whereas [3H]NE uptake was preferentially inhibited by 10-7M desipramine. These results may reflect regional brain specialization and/or different developmental patterns of high affinity uptake of serotonin and catecholamines by astrocytes in situ.

    Notizen: Abstract: Glial fibrillary acid protein (GFAP)-positive astrocytes isolated from the cerebral cortices of 3–10-day-old rats frequently showed increased intracellular Ca2+ concentration responses to l-glutamate and glutamate analogues. However, few of the acutely isolated cells responded to ATP, and no such cells responded to serotonin [5-hydroxytryptamine (5-HT)]. The same cell that failed to respond to ATP or 5-HT often responded to glutamate. Culturing acutely isolated cells in media containing horse serum decreased Ca2+ responses to glutamate but increased the responses to ATP and induced responses to 5-HT. In primary cultures prepared from the cerebral cortices of 1-day-old rats and cultured in horse serum, fewer of the cells responded to glutamate, but almost all cells responded to ATP and 5-HT. The lack of, or limited response to, 5-HT or ATP in the acutely isolated cells seems unlikely to be due to selective damage to the respective receptors because acutely isolated GFAP-negative cells showed responses to ATP, several different proteases and mechanical dissociation yielded cells that also responded to glutamate but not to ATP, and exposure of primary cultures to papain did not abolish Ca2+ responses to several transmitters. The responses of the acutely isolated cells to glutamate but limited or lack of responses to ATP and 5-HT also correspond to what has been seen so far for astrocytes in situ. Thus, the present studies provide direct evidence that some of the receptors seen in primary astrocyte cultures may reflect a response to culture conditions and that, in the context of the relevant information so far available, acutely isolated astrocytes seem to reflect better the in vivo state.

    Notizen: Abstract: The carbonic anhydrase activity of 3-week-old primary astroglial cultures started from the dissociated cerebral hemispheres of neonatal rats was increased up to twofold after treatment of the cultures with 0.1 mM-norepinephrine or histamine. Stimulation due to addition of norepinephrine was inhibited by propranolol. The carbonic anhydrase activity of primary cultures derived from the cerebellum plus brain stem regions was about fourfold greater than the activity of primary cultures started from cerebral hemispheres, but in contrast was not stimulated by norepinephrine. Treatment of the cerebral cultures with norepinephrine in the presence of 32P resulted in a two- to threefold increased incorporation of 32P into carbonic anhydrase purified from the same cultures, and this increased incorporation was inhibited by propranolol. It is suggested that one of the consequences of the stimulation of 3′,5′-cyclic AMP levels in brain by norepinephrine is activation of astroglial carbonic anhydrase activity due to 3′5′-cyclic AMP-stimulated phosphorylation of the enzyme.

    Notizen: Addition of norepinephrine or isoproterenol to primary cultures started from the brains of 1-3 day old rats caused up to 200-fold increases in cAMP levels, which reached a maximum by 5-10 min and then declined. This effect was studied in detail for norepinephrine. The rise in cAMP levels was followed by morphological changes, in which up to 65% of the cells exhibited an astrocyte-like morphology, and 2-3 fold increases in carbonic anhydrase and (Na+-K+) ATPase activities. However, morphological transformation also occurred after much smaller increases in total cAMP levels. These effects on cell morphology and enzyme activities reached a maximum 1-2 h after addition of norepinephrine and then declined. Carbonic anhydrase activity was found both in the particulate and post 100,000 g supernatant fractions from homogenates of these cultured cells, and in the latter case the activity was activated 3-fold by addition of cAMP. The significance of these obscrvations on the cellular localization of, and functional role for similar increases in cAMP in brain tissue is discussed.

    Notizen: Abstract— Increasing the HCO3− concentration of incubation media containing raised K+ concentrations (18-71 mm) caused increased swelling of monkey cerebral cortex slices. This swelling was mainly associated with increased intracellular levels of Na+ and Cl− ions. It was independent of the type of buffer used and was not a result of the increased Na+ concentration in the media due to added HCO3− or the increased osmolarity. The levels also were unaffected by alteration of the pH in the range of 6·9- 7·8 or pCO2 in the range of 3–81 mm Hg.The anatomical locus of this HCO3− stimulated swelling appeared in electron micrographs to be an expanded glial compartment. The significance of these findings is discussed in terms of the transport processes involved and the role of glial cells in maintaining correct cerebro-cortical ion balances under normal and pathological conditions.

    Notizen: Abstract: Carbonic anhydrase (CA) was studied in primary monolayer cultures from neonatal rat cerebral hemispheres with both immunocytochemical and biochemical techniques. In such cultures, which consist predominantly of astrocytes, immunocytochemical staining for CA using antibody raised against the type II enzyme from rat erythrocytes resulted in positive staining of the flat, glial fibrillary acidic protein-positive, astrocytic monolayer. Smaller, process-bearing, round cells that grew on top of the astrocytes stained intensely for CA. We estimated that these cells represented 1% or less of the total cells in the cultures, and they have been identified by others as oligodendrocytes. The intensity of the staining of astrocytes for CA could be increased to that observed in oligodendrocytes when the astrocytes were made to round up and form processes by treatment with 2′,3′-dibutyryl cyclic AMP. Enzymatic assays showed that CA activity of the cultures after 3 weeks of growth was 2.5-to 5-fold less than that found for cerebral homogenates from perfused 3-week-old rat brains. However, both activities were totally inhibited by acetazolamide with an I50 of 10−8M, confirming that both rat brain and the astrocyte cultures possess the high-activity type II enzyme. CA-II activity was unaffected by treatment of the cultures with a method reported to remove oligodendrocytes. Thus, the immunocytochemical and biochemical studies reported here demonstrate that astroglial cells in primary cultures from neonatal rat brain contain CA-II.

    Notizen: —The ouabain-sensitive K+ uptake and ATPase activities of cultured glioma and neuroblastoma cells were studied. Both cell lines showed ouabain-sensitive K+ uptake which correlated with the level of [Na++ K+]ATPase activity found in the respective total cell homogenate. The glioma cells had a 2.1-fold higher rate of K+ uptake than neuroblastoma cells, and a 2.4-fold higher [Na++ K+]ATPase activity. In the presence of ouabain neuroblastoma cells released K+ and took up Na+ in a 1:1 ratio. These results are compared and contrasted with similar studies on brain tissue and isolated cells. It is suggested that the cultured cell lines may serve as good models for the cation transport properties of their tissue counterparts.
