    Notizen: For the precise low background experiments on β decay and material sciences, we are planning to build a new ISOL-type facility to accelerate various β-radioactive nuclei utilizing the existing 5 MV Van de Graaff accelerator and an RFQ linac. Present status of the designing work is reported. In the present work, optical elements of the 41-m-long beam transport line to connect Van de Graaff accelerator and an RFQ linac were designed. Using two dipole magnets, two electric quadrupole doublets and six electric quadrupole triplets, low energy radioactive nuclear beams can be transported with sufficient efficiency. © 2000 American Institute of Physics.

    Notizen: We have developed a time pickup system for monitoring the beam bursts from a cyclotron. The time pickup can measure the phase and the width of the beam bursts by detecting secondary electrons emitted from a foil or a wire target inserted into the beam with time resolutions around 200 ps full width at half maximum in spite of its simple structure. By using the foil and the wire targets appropriately, monitoring the bursts of a variety of beams can be achieved. In addition, by adjusting the position of the target, a high enough detection rate of the beam burst is obtained with a minimum of interference with the beam. The present investigation shows that it is needed to adjust the collection voltage of the time pickup because the optimum collection voltages for high resolution measurement are in the range from −0.5 to −1.0 kV and those for high efficiency measurement are not less than −4.0 kV. © 2002 American Institute of Physics.

    Notizen: We describe the use of endosseous implants in the autogenous particulate cancellous bone and marrow grafted alveoli after orthognathic surgery for dental rehabilitation of patients with cleft lip and palate. This procedure has been applied to two patients and produced good results functionally and aesthetically. The results are encouraging and indicate that implant placement after orthognathic surgery is useful for patients both with congenital missing teeth and retrognathic maxillae.

    Notizen: summary  We report here on a patient with bilateral cleft lip and palate (BCLP) and a missing premaxilla, who underwent dentoalveolar reconstruction of the cleft and premaxillary alveolus using endosteal implants after bone grafting. The patient, whose maxillary incisors and premaxilla were missing, had corticocancellous bone grafting from the iliac crest, followed by excellent bone formation at the anterior alveolus. After the placement of the endosteal implants and the completion of the pre-surgical orthodontic alignment, orthognathic surgery was performed for the restoration of a Class III open bite. After post-operative orthodontic preparation, the final fixed prostheses were completed. This treatment procedure offers an option of dentoalveolar reconstruction for BCLP patients with an excised premaxilla.

    Notizen: Abstract We studied a cystic ganglioglioma (GG) located in the right frontal lobe of the brain. Interestingly, the fibrillary spindle glial cells were often arranged in a fascicular pattern, and the generally uniform, round-to-oval delicate nuclei appeared to resemble those of ependymoma; and the neoplastic neurons often contained neurofibrillary tangles (NFTs). The glial component was positive for glial fibrillary acidic protein and occasionally contained granular or microvesicular structures positive for epithelial membrane antigen. Ultrastructural investigation revealed that the glial cells were ependymal in nature; intracytoplasmic lumina and intercellular microrosettes lined with cilia and microvilli, as well as long zonulae adherentes, were evident. In addition, chromogranin A-positive granular staining, neurosecretory-granule-like structures, and parallel arrays of microtubules were sometimes associated with the blood vessels. We considered the present case to be an unusual example of GG with an ependymoma, more precisely a tanycytic ependymoma, as the glial component; to our knowledge, the existence of ependymoma as the main glial component of this particular tumor has not been described before. The occurrence of NFTs, which has been reported in several cases of GG, was an additional, unusual feature.

    Notizen: Summary The factors affecting fluorescence fading in cytofluorometry were investigated using different kinds of nuclear staining, mounting media, and procedure of specimen preparation. Acceleration of fluorescence fading was observed in smear specimens treated with RNase, trypsin, or hypotonic solution before pararosaniline Feulgen nuclear staining. Similar effect was found for other DNA-stainings such as “33258 Hoechst” and Feulgen reactions with different Schiff-type dyes, such as acriflavine-SO2 and cresylviolet-SO2, when chemically pure DNA was used. Fluorescence decay was rapid for all fluorochromes examined, when glycerin or buffer solution was used as mounting medium. Marked stabilization of fluorescence emission was induced in specimen mounted in non-fluorescent resin, Entellan (Merck), after post-staining fixation with absolute methanol for all tested fluorochromes. The same treatment induced almost complete fluorescence stabilization of fluorescein isothiocyanate (FITC); no detectable fluorescence fading was observed in a specimen stained with indirect immunofluorescence reaction using anti-UV-DNA antibody, during storage for 2 years at room temperature without special protection against light. These observations suggest that factors which bring about conformational stability of macromoleculedye complexes generally induce fluorescence stabilization.

    Notizen: Summary A technique for isolation of cells from paraffin embedded tissue is indispensable for the performance of Feulgen-DNA cytofluorometry in parallel with the definition of histological characteristics. Background fluorescence due to nonspecific dye-binding by a “pseudo-plasmal reaction” is usually found to be so intense on cells isolated from formalin-fixed tissues, that we are often forced to abandon quantitative DNA determinations. In the present work, we report that fixation of tissues with Carnoy's fixative for 12 h at 5° C not only reduces nonspecific dye-binding but also facilitates the process of cell isolation. Furthermore, we find that pre-treatment of cells isolated from Carnoy-fixed tissues with acidic azocarmin G solution completely blocks nonspecific dye-binding in subsequent acriflavine Feulgen nuclear staining. This combination of techniques for specimen preparation enables us to carry out Feulgen-DNA cytofluorometry on cells isolated from histological sections with satisfactorily low coefficients of variation (less than 8%). The techniques should be widely applicable for parallel DNA determinations and histology.

    Notizen: Summary The progress of polyploidization in the human heart muscle cell was investigated by cytofluorometry, involving selective measurements of heart muscle cell nuclei. Thirty-two tissue samples, taken from the free wall of the left ventricle of each autopsied heart, were fixed in Carnoy's fluid. From thick (100–150 μm) paraffin sections, isolated cells for smears were obtained by enzyme digestion and ultrasonic treatment. The smears were stained with azocarmin G to eliminate background fluorescence and subsequently stained by an acriflavine-Feulgen reaction. Cytofluorometric DNA-determinations were carried out selectively on heart muscle cell nuclei, using the muscle striations revealed by azocarmin G-fluorescence as specific markers. The dynamic process of polyploidization in normal hearts could be divided into four stages. In the first stage (under 1 year of age), almost all heart muscle cell nuclei (94.3±1.8%) were diploid. In the second stage (1 to 9 years of age), the number of tetraploid nuclei increased (13.6±7.1%). In the third stage (9 to 22 years of age), octaploid nuclei first appeared and the number of tetraploid nuclei increased (26.7±3.9%). The DNA pattern in the fourth stage (22 to 75 years of age) was relatively constant, with a ratio of diploid (62.4±8.7%), tetraploid (31.4±6.7%) and octaploid (5.8±3.9%) nuclei. From these results it was concluded that physiological polyploidization progresses in proportion to the increase of heart weight. The frequency of polyploid nuclei in human heart was not so high as resported by previous investigators.

    Notizen: Summary In Feulgen nuclear staining nonspecific dye-binding due to the “pseudo-plasmal reaction” is intensified in isolated cells with intact cytoplasm, and cannot be eliminated by the post-irradiation method. Fluorescence intensity in the cytoplasm sometimes exceeds that of specific nuclear fluorescence, especially in brain and heart muscle cells, and it was almost impossible to perform cytofluorometric DNA quantification on such specimens. Various kinds of aldehyde-blocking agents such as sodium borohydride, 2,4-dinitrophenylhydrazine, aniline, and sodium pyrosulfite were effective in reducing the “pseudo-plasmal reaction”. But the blocking effects were not complete because of additional release of reactive aldehyde groups during subsequent Feulgen hydrolysis. Acidic azocarmin G produces a complete block of all “pseudo-plasmal reaction” in acriflavine-Feulgen nuclear staining, allowing accurate DNA-cytofluorometry to be carried out.
