    Notizen: Abstract: In the human neuroblastoma cell line LA-N-2, recombinant rat ciliary neurotrophic factor (CNTF) induced neurite growth and cholinergic differentiation that were both half-maximally saturated at 〈100 pM of the neurokine, but was not required for cell survival in serum-free conditions over a 13-day period. CNTF markedly stimulated choline acetyltransferase activity and acetylcholine synthesis, whereas high-affinity choline transport was only slightly enhanced and acetylcholinesterase activity was unchanged. Leukemia inhibitory factor had effects identical to CNTF on neurite growth and choline acetyltransferase activity, but interleukin 6 had no effect. Radioiodinated CNTF binding and affinity cross-linking studies were consistent with tripartite receptor activation as a mediator of the observed biological effects.

    Notizen: Abstract: We have probed the molecular basis of functional effects of ciliary neurotrophic factor (CNTF) and nerve growth factor (NGF) on aspects of the neuronal differentiation of LA-N-2 neuroblastoma cells. The influence of CNTF on the cholinergic phenotype can be accounted for by transcriptional/translational effects without implicating posttranslational mechanisms. Although both NGF receptors are expressed constitutively by LA-N-2 cells, CNTF has a marked stimulatory effect on trkA mRNA and protein. The NGF receptors are functional in serum-free conditions where they mitigate CNTF effects on cell adhesion but do not support process extension. Following priming by CNTF, NGF and CNTF have synergistic influences on process formation but not on choline acetyltransferase-specific activity.

A number of studies have demonstrated that the nervous system produces multiple molecular species of proteoglycans (PGs), and that these PGs on cell surfaces and in the extracellular matrix of the nervous system have the potential to act both as ligands for and receptors on neurons. PGs are now known to play an important role in cell-cell and cell-substrate interactions. This review focuses on a discussion of the expression, characterization and modulation of individual astrocytic PGs and the role that astrocytic PGs play in both the development and pathophysiology of the nervous system.

    Notizen: Abstract: A rigorous extraction of the deuteron charge form factors from tensor polarization data in elastic electron-deuteron scattering, at given values of the 4-momentum transfer, is presented. Then the world data for elastic electron-deuteron scattering is used to parameterize, in three different ways, the three electromagnetic form factors of the deuteron in the 4-momentum transfer range 0-7 fm−1. This procedure is made possible with the advent of recent polarization measurements. The parameterizations allow a phenomenological characterization of the deuteron electromagnetic structure. They can be used to remove ambiguities in the form factors extraction from future polarization data.

    Notizen: Abstract A QOL instrument was developed to measure the specific concerns of long term cancer survivors. The QOL-CS is based on previous versions of the QOL instrument developed by researchers at the City of Hope National Medical Centre (Grant, Padilla, and Ferrell). This instrument was revised over a one year pilot by Hassey-Dow and Ferrell. The revised instrument included 41 items representing the four domains of quality of life incorporating physical, psychological, social, and spiritual well being. The present study was conducted as a mail survey to the membership (n=1,200) of the National Coalition for Cancer Survivorship with 686 subjects responding to the survey. This survey included a Demographic tool, QOL-CS and the FACT-G tool developed by Cella. Psychometric analysis, performed on 686 respondents, included measures of reliability and validity. Two measures of reliability included test-retest and internal consistency. The overall QOL-CS tool test-retest reliability was 0.89 with subscales of Physicalr=0.88, Psychologicalr=0.88, Socialr=0.81, and Spiritual,r=0.90. The second measure of reliability was computation of internal consistency using Cronbach's α coefficient as a measure of agreement between items and subscales. Analysis revealed an overallr=0.93. Subscale alphas average ranged fromr=0.71 for spiritual well being,r=0.77 for physical,r=0.81 for social, andr=0.89 for psychological. Several measures of validity were used to determine the extent to which the instrument measured the concept of QOL in cancer survivors. The first method of content validity was based on a panel of QOL researchers and nurses with expertise in oncology. The second measure used stepwise multiple regression to determine factors most predictive of overall QOL in cancer survivors. Seventeen variables were found to be statistically significant accounting for 91% of the variance in overall QOL. The fourth measure of validity used Pearson's correlations to estimate the relationships between the subscales of QOL-CS and the subscales of the established FACT-G tool. There was moderate to strong correlation between associated subscales including QOL-CS physical to FACT physical (r=0.74), QOL-CS Psych to FACT Emotional (r=0.65), QOL Social to FACT Social (r=0.44). The overall QOL-CS correlation with the FACT-G was 0.78. Additional measures of validity included correlations of indimeasures of validity included correlations of individual items of the QOL-CS tool, factor analysis, and construct validity discriminating known groups of cancer survivors. Findings demonstrated that the QOL-CS and its subscales adequately measured QOL in this growing population of cancer survivors.

    Notizen: Summary A single neuronal cell assay of neurite growth was utilized to determine types and domains of neuronal proteoglycans involved in neurite growth on laminin. Perturbations of biosynthesis and processing, enzymatic digestion with specific lyases, and competition with glycosaminoglycan side chains produced complementary data consistent with a molecular model implicating glycosaminoglycan (GAG) residues of heparan sulphate proteoglycans (HSPGs) in neurite growth. The observations suggest that HSPGs promote neurite growth on laminin by bridging between binding domains for HSPGs on laminin and on the neuronal cell surface, and that the bridge is tethered at both ends by noncovalent interactions between the binding domains and GAG side chains. Sulphation of the GAGs of HSPGs appears to be critical to the tethering and/or neurite growth-promoting activity of neuronal HSPGs.
