    Notes: Abstract Protease activity was detected in the culture medium of Flavobacterium balustinum P104 grown at 10 °C, which was isolated from salmon (Oncorhynchus keta) intestine. The enzyme, designated as CP-70 protease, was purified to homogeneity from the culture broth by ion exchange and gel filtration chromatographyies. The molecular mass of the protease was 70 kDa, and its isoelectric point was close to 3.5. Maximal activity toward azocasein was observed at 40 °C and from pH 7.0 to 9.0. The activity was strongly inhibited by phenylmethylsulfonyl fluoride, suggesting that the enzyme is a serine protease. The n-terminal amino acid sequence was Asp-Thr-Arg-Gln-Leu-Leu-Asn-Ala-Asn-Ser-Asp-Leu-Leu-Asn-Thr-Thr-Gly-Asn-Val-Thr-Gly-Leu-Thr-Gly-Ala-Phe-Asn-Gly-Glu-Asn. A search through the database for sequence homology yielded no significant match. The initial cleavage sites for oxidized insulin B-chain were found to be the Glu13-Ala14 and Phe24-Phe25 bonds. The result of the cleavage pattern of oxidized insulin B-chain suggests that CP-70 protease has a broader specificity than the other cold-active proteases against the peptide substrate.

    Notes: Summary.  The nucleotide sequence of the region including the RNA-dependent RNA polymerase (RdRp) of the porcine enterovirus 1 (PEV-1) Talfan strain was determined. Amino acid identities of Talfan RdRp with those of other picornaviruses were significantly lower than those seen among the viruses of the same genus of picornaviruses. The phylogenetic analysis of RdRp indicated that picornaviruses were divided into 8 clusters, in which Talfan was genetically distinct from other picornaviruses in contrast to the current taxonomy. In this study, we propose that PEV-1 Talfan be regarded as a new genus for the family Picorna- viridae.

    Notes: Abstract A study was undertaken to evaluate the incidence of nasopharyngeal α-streptococci with inhibitory activity against pathogens, as a defense mechanism of the normal bacterial flora against infection. Cultures were taken from the nasopharynges of 43 children with otitis media with effusion (OME). The detection rates of α-streptococci with inhibitory activity against Haemophilus influenzae, Streptococcus pneumoniae, Staphylococus aureus and group A streptococci were significantly lower in the nasopharynx than those isolated from the tonsils of the same patients. Moreover, the detection rates of α-streptococci with inhibitory activity against all of these pathogens derived from the nasopharynx were lower than those in healthy children, streptococcal strains with activity against H. influenzae and Strep. pneumoniae were also lower than that in patients with tonsillitis. These findings suggest that low nasopharyngeal levels of α-streptococci with inhibitory activity against respiratory pathogens may render children susceptible to OME. Further studies are needed to investigate the relationships between the prevalence of pathogens in the nasopharynx and the inhibitory activities of α-streptococci against them in order to devise and select optimal treatment for patients with OME.

    Notes: Abstract The eighth nerve compound action potential (CAP) in 95 guinea pigs was measured using click stimuli to investigate age-related changes in their neural auditory thresholds. The animals were separated into three groups: group A (n = 43, 86 ears; 2–4 months old); group B (n = 29; 58 ears, 13–15 months old); and group C (n = 23; 46 ears, 23–25 months old). With increasing age, a gradual elevation of CAP thresholds was clearly seen among the three groups. The negative peak (N1) latencies of the CAP were prolonged, and the N1 amplitudes of the CAP decreased. There were significant differences in N1 latencies among the three groups and in N1 amplitudes between groups A and B, and between groups A and C. However, the rate of decline of the thresholds as well as the input-output function curves of the CAP varied in some of the oldest animals, suggesting that there were some individual differences in degenerative aging processes of the auditory system.

    Notes: Abstract The orthostatic test was administered to 134 dizzy patients (37 males and 97 females) who were identified as having orthostatic dysregulation from December 1990 through November 1994. The mean age of the male and female patients was 42.5 and 44.6 years, respectively, and the peak age decade was the 50s. Among the questionnaire items for orthostatic dysregulation, orthostatic dizziness was the most frequent major symptom and occurred in 116 of the patients (86.6%). Fatigue was the most frequent minor symptom and was found in 97 (72.4%). The incidence of positive orthostatic test results in the patients with orthostatic dysregulation (50.7%) was significantly higher (P 〈 0.01) than that in the control subjects (27.8%). There was also a significant difference (P 〈 0.01 or P 〈 0.05) between systolic pressure decrease during the orthostatic test procedure in the patients with orthostatic dysregulation and that in the control subjects. These results suggest that autonomic imbalance as reflected by the orthostatic dysregulation questionnaire and orthostatic test results is closely related to the occurrence of dizziness or vertigo.

    Notes: Abstract Chlorine and oxygen evolving at RuO2/Ti and RuO2–TiO2/Ti anodes have been simultaneously determined at electrode potentials from 1.0 to about 2V (vs Ag/AgCl) by differential electrochemical mass spectroscopy (DEMS). On the RuO2/Ti anodes, the threshold electrode potential for oxygen evolution increased with a decrease in RuO2 loading, while the chlorine evolution potential was unchanged. Low RuO2 loading anodes gave a high chlorine evolution ratio under various constant electrolysis potentials. On the RuO2–TiO2/Ti anodes, the threshold electrode potential for oxygen evolution increased with an increase in the TiO2 content more remarkably than that for chlorine evolution. High TiO2 content anodes gave a high chlorine evolution ratio at various constant electrolysis potentials. The combination of RuO2 and TiO2 exhibits a remarkable effect with respect to the enhancement of chlorine evolution selectivity.

    Notes: Abstract The method explained in this paper for quantitative evaluation of fatigue limit for materials containing defects is based on the experimental evidences that inhomogeneities and micro-notches can be treated like cracks. First, the basic concept of the √area parameter model is explained introducing the various data obtained by the first author's group for over last 15 years. Evidences are shown that small cracks, defects and nonmetallic inclusions having the same value of the square root of projection area, √area, have the identical influence on the fatigue limit regardless of different stress concentration factors. Various applications of these concepts to various defect types and microstructural inhomogeneities are shown. Since the estimation of fatigue strength is related to the estimation of the size of maximum defects occurring in a piece, the methods for searching the defects and the quality control of materials with respect to inclusion or defect rating as well as their statistical implications are discussed.

    Notes: Abstract  For the advancement of Human Genome Project, we have developed an automated DNA sequencing system ‘HUGA-I.’ It is composed of several automated instruments and transfer robots connecting them. In this paper we describe the results of the performance evaluation test of HUGA-I. Although some of the system units showed good performances, the total performance of the HUGA-I was about 1/6 of the designed value. By revealing principal reasons of this poor performance, we would like to contribute to the automation in genome analysis, particularly in human genome analysis.

    Notes: Abstract Fine particles of the amorphous Cr2O3-Fe2O2 solid solutions were prepared by dehydration of coprecipitated hydroxides and their crystallization behavior was studied by differential thermal analysis and X-ray diffraction. The peak temperature for crystallization attained a maximum at a composition near Fe2O3 content of about 60 mol % and the activation energy for crystallization attained a minimum at a composition near Fe2O3 content of about 50 mol % in this quasibinary system. Phase separation occurred in a range of Fe2O3 content from about 35 to 80 mol % in the corundum-type solid solutions heat treated at 600 °C for 2 h. Crystallization behavior was discussed briefly related with phase separation and diffusion in fine particles.
