    Notizen: Abstract A transient plasmolysis/deplasmolysis (plasmolytic cycle) of onion epidermal cells has been shown to induce the formation of fluid-phase endocytic vesicles. Plasmolysis in the presence of the membrane-impermeant fluorescent probes Lucifer Yellow CH (LYCH) and Cascade Blue hydrazide resulted in the uptake of these probes by fluid-phase endocytosis. Following deplasmolysis, many of the dye-containing vesicles left their parietal positions within the cell and underwent vigorous streaming in the cytoplasm. Vesicles were observed to move within transvacuolar strands and their movements were recorded over several hours by video-microscopy. Within 2 h of deplasmolysis several of the larger endocytic vesicles had clustered around the nuclear membrane, apparently lodged in the narrow zone of cytoplams surrounding the nucleus. In further experiments LYCH was endocytically loaded into the cells during the first plasmolytic cycle and Cascade Blue subsequently loaded during a second plasmolytic cycle. This resulted in the introduction of two populations of endocytic vesicles into the cells, each containing a different probe. Both sets of vesicles underwent cytoplasmic streaming. The data are discussed in the light of previous observations of fluid-phase endocytosis in plant cells.

    Notizen: Summary Changes in the fine structure of flax (Linum usitatissimum) cotyledon leaf cells treated with the herbicide paraquat (1,1′-dimethyl-4,4′-bipyridylium ion.) were investigated. After 6 h treatment under constant illumination, tonoplast breakdown was evident. This was followed by a rapid and progressive deterioration of cell contents including the disruption of mitochondria, and the breakdown of chloroplast thylakoid membrane structure with the accumulation of osmiophilic plastoglobuli. There was no apparent deterioration after 18 h paraquat treatment in darkness but by 30 h there was a limited breakdown of chloroplast membranes.

Results showed an inhibition of photosynthetic CO2 uptake and electron flow of isolated chloroplasts. An increase in membrane permeability, changes in the level of the major lipid components and of malondialdehyde. These are correlated with ultrastructural changes and the discussion includes a proposed mode of action for the herbicides.

    Notizen: A computer program, written in Whetstone Algol, is described for handling the arithmetical operations involved in a precision method of determining lattice spacings from Kossel diffraction patterns. The interpretation treats the Kossel pattern as a three-dimensional array of intersecting cones with a common vertex and depends on the fact that when three or more positions of a cone generator are established, its axis and semivertex angle are defined. The theory and experimental requirements of the method are outlined and the functions of the steps in the computer program are described. The full program, together with data sheets and print-out of results for a typical calculation is available on request. A subsidiary program is also available to provide a graphical display of relevant parts of the results.

    Notizen: A single exposure method is described for the calculation of lattice spacings and crystal orientation, from Kossel diffraction patterns. This method is extremely simple from an experimental viewpoint since the only data required are the coordinates of five or more points on each Kossel diffraction curve in an arbitrary orthogonal system and the wavelength of the characteristic X-ray radiation forming the pattern. The method requires the analysis of at least two diffraction curves which can be either elliptical or hyperbolic. The equation of each curve is first derived in the arbitrary system. It is then transformed to a coordinate system in which the coefficients may be equated with those of an equation of the same curve derived in terms of the lattice spacing, the orientation of the crystal planes responsible for the diffraction cone and the X-ray wavelength. The lattice spacings and the directions of the crystal plane normals are calculated. A computer program has been written to handle the lengthy arithmetical operations required for the analysis, and application of the method to a Kossel pattern from a single grain of Swedish iron is described.

    Notizen: Summary Hydrocephalus in the H-Tx rat first develops in late gestation and causes death at 4–7 weeks. The effect of hydrocephalus on overall cortical dimensions and on five specific regions (frontal, sensory-motor, parietal, auditory and visual) has been studied by quantitative light microscopy at 10 and 30 days after birth. The lateral ventricle volumes in hydrocephalic rats were about 40x larger than controls and increased fourfold between 10 and 30 days. Cortical volume was reduced by a small amount at 10 days but was larger in hydrocephalics at 30 days. Thinning of the cortical mantle was severe with disruption of the laminar structure, particularly in the auditory and visual regions, where it was already present at 10 days. The density of cortical cells (neurones and glia) was not altered in hydrocephalics at 10 days but was reduced in all regions at 30 days. Estimates of total cell number suggest that the lower density was not associated with an overall loss of cells. Capillary numerical density was not affected by the hydrocephalus at 10 days after birth but by 30 days it was significantly lower, particularly in the worst-affected posterior regions. The results show that the cerebral cortex is severely distorted and that in advanced hydrocephalus, although overall cell number is not affected, both cell density and capillary density are lower by up to 30%.

    Notizen: Summary Severe microvascular disease exists at the stage of clinical diabetic neuropathy. A non-invasive test that will identify those diabetic subjects who will eventually develop neuropathy is essential for early intervention. Sural sensory conduction velocity was recorded (x 3) in 12 non-neuropathic diabetic subjects, 15 diabetic subjects with established neuropathy and 16 age-matched normal control subjects, before and after exercise to 80% age/sex predicted maximum heart rate. Fixed sural electrodes were used. Subcutaneous temperature was recorded by a needle thermocouple placed near the sural nerve. Sural sensory conduction velocity increased significantly after exercise in normal subjects (p〈0.01, mean increase 5.07 m/s) and non-neuropathic diabetic subjects (p〈0.02, mean increase 3.99 m/s) but not in neuropathic subjects (mean increase 0.99 m/s). Subcutaneous temperature rose significantly in normal subjects (p〈0.01, mean increase 2.07°C) and non-neuropathic diabetic subjects (p〈0.001, mean increase 2.52 °C) but not in neuropathic subjects (mean increase 0.15 °C). However, sural sensory conduction velocity increased by 1.2 m · s−1. °C−1 following direct warming of the limb in six neuropathic subjects which was comparable to that of normal and non-neuropathic subjects (1.49 and 1.48 m · s−1. °C−1). The impairment of exercise conduction increment in diabetic neuropathy suggests impaired nerve blood flow in diabetic neuropathy.

    Notizen: Summary A mathematical model is described that demonstrated the properties of cerebral vascular resistance and compliance expressed as a function of cerebral perfusion pressure (CPP) and arterial CO2 partial pressure (PaCO2). The hypercapnic induced shift of the lower limit of autoregulation to a higher range of CPP, as shown by this model, is a useful characteristic that facilitates the differentiation between normal and impaired autoregulation described previously in experimental studies. Dynamic properties of cerebrovascular circulation derived from the relationship between pulse wave of CBF waveform and CPP have been analysed at different levels of PaCO2-phenomenon, being often described as dependence of blood flow velocity pulsatility index on the autoregulatory reserve. The model was also used to interpret interhemispheric asymmetry of CBF reactivity to changes in arterial concentration of CO2 in patients with carotid artery stenosis.

    Notizen: Abstract The mineralogy, petrology and Rb-Sr isotope compositions of muscovite granite sills and associated pegmatites from the Barousse Massif, Central Pyrenees, are described. The petrogenesis of the granites is discussed with respect to their structure, mineral assemblages, mineral compositions and initial 87Sr/86Sr ratio. It is concluded that the muscovite granite sills have been intruded as magma, formed from the partial melting of continental crust at 276 million years. Progressive crystallisation of the siliceous magma has concentrated the aqueous phase in the remaining melt, and the associated pegmatites are the final expression of the crystallising intrusion.
