    Notes: Zusammenfassung Die Bestimmung der HWZ von Fettpartikeln im Blut mit Hilfe intravenös injizierter radioaktiv markierter Fettemulsionen wird beschrieben. Es wird diskutiert, wieweit ein darauf aufbauender Test Einblick in den Chylomikronenstoffwechsel geben kann. Die Brauchbarkeit des Testes wird an größeren Personengruppen geprüft. Bei normolipämischen Personen beträgt die HWZ 3,74±0,69 min. Bei Hyperlipämien lassen sich zwei Formen trennen. Mögliche Gründe für die Verlängerung der HWZ bei Hyperlipämien werden diskutiert.

    Notes: Zusammenfassung Corpus luteum und Corpus rubrum von Schweineovarien wurden auf proteolytische Enzyme, saure und alkalische Phosphatase untersucht. Die Herstellung von Zellfraktionen erfolgte nach der Methode vonLang undSiebert. Bei pH 3,5 findet sich eine außerordentlich hohe proteolytische Aktivität im Corpus luteum. Diese beträgt für 1 ml Homogenat 21450γ Tyrosin/Std, was einer Aktivität von 64350γ Tyrosin/g Frischgewebe/Std entspricht. Die proteolytische Aktivität des Corpus rubrum ist nur 1/10 so hoch wie die des Corpus luteum. Die saure Phosphatase bei pH 5,0 ist über zweimal so hoch wie die Aktivität der alkalischen Phosphatase bei pH 9,0. Es wird diskutiert, ob zwischen der hohen proteolytischen Aktivität des Corpus luteum und den auf das Corpus luteum wirksamen Proteohormonen eine Beziehung bestehen könnte.

    Notes: Abstract The effects of the peptide polycations salmon protamine (M r = 4332,z = + 21) and poly-l-lysine (M r ≅ 100,00,z ≅ + 775) on ion channels formed by synthetic alamethicin Alm-F30 (one negative charge), natural Alm-F50 (neutral) and phosphorylated Alm-F50 (two negative charges) reconstituted in planar lipid bilayers have been studied at the single channel level. It was observed that both polycations in micromolar concentrations transiently block ion permeation through the channels formed by each alamethicin analogue, although in case of the neutral Alm-F50 to a significantly lesser extent. Poly-l-lysine showed to be more effective than protamine in blocking these channels. If either polycation is present in the cis-compartment, blockade occurs only at cis positive membrane voltages. At constant polycation concentration, dwell times in the blocked state increase when salt concentration is lowered, and decrease at acidic pH with an apparent pK of 4.8. Mean lifetime of blockade events shortens when membrane voltage is increased, which suggests that both polycations may permeate through the oligomeric alamethicin channels if conductance levels are 〉 2. We suggest that blockade is caused by electrostatic binding of a single polycation molecule to the C-terminal channel mouth; in case of Alm-F30, Glu18 has to be considered as the putative binding site. Our results provide further evidence for the barrel-stave model and a parallel orientation of dipole monomers in the channel aggregate, the C-termini facing the membrane side with the more positive membrane potential.

    Notes: Abstract This paper describes the chemical characterization of staphyloferrin B, a new complexone type siderophore isolated from low iron cultures of Staphylococcus hyicus DSM 20459. Purification of the very hydrophilic metabolite was achieved by anion exchange high performance liquid chromatography HPLC. Mass spectrometry showed a molecular mass of 448 amu. Hydrolysis with 8 mHCl revealed the presence of l-2,3-diaminopropionic acid, citrate, ethylenediamine and succinic semialdehyde. The connections between the four building blocks were determined by two-dimensional nuclear magnetic resonance measurements. UV/Vis and circular dichroism spectra are consistent with the proposed structure, which could also be confirmed by precursor feeding. The siderophore activity of staphyloferrin B was demonstrated by iron transport measurements.

    Notes: Abstract Bacterial cell wall components such as lipopolysaccharide, a variety of membrane proteins, murein, and lipoprotein can act as immunoadjuvants for bacterial vaccines, thus enhancing protection from bacterial infections. Synthetically prepared N-terminal parts of the lipoprotein from Enterobacteria carrying three fatty acid residues or lipopeptide analogs containing one to four aminoacids bound to S-glycerylcysteine act as potent immunoadjuvants in vivo in combination with or covalently linked to antiges. Here we demonstrate that the supplementation ofSalmonella vaccines with these synthetic lipopeptides significantly enhances their vaccine efficiency in mice. Variations in the native lipopeptide structure regarding chain length and amino acid sequence of the peptide moiety, as well as modifications of the lipoamino acid, lead to reduction or even complete loss of the adjuvant activity. The immunoadjuvant properties of the lipopeptides as described here are mediated by an enhancement of the humoral immune response.

    Notes: Summary Differentiated mesophyll cells ofTriticum aestivum (cv. Star) exhibit a lobed outline resembling tube-shaped balloons with almost regularly spaced constrictions. It was shown that these constrictions are probably the result of hoops of wall reinforcements laid down during early stages of cell expansion. It appears that these hoops prevent expansion in the corresponding regions and thus give rise to the peculiar cell shape. The comparatively thin cell walls of the bulges are uniformly reinforced after the lobed shape is established. By using immunofluorescence techniques a change in the pattern of cortical microtubule arrangement was observed which corresponded to the pattern of cell wall deposition. Discrete bands of microtubules were found beneath the sites of hoop reinforcement. These bands disintegrated during late stages of cell expansion with microtubules fanning out into the almost empty regions of the bulges.

    Notes: Zusammenfassung Es wird über Erfahrungen mit dem131J-Öltoleranztest zur Prüfung der Resorptionsleistung bei Krankheiten der Verdauungsorgane (37 Patienten) berichtet, die dem Verhalten stoffwechsel- und verdauungsgesunder Personen (15 Personen) gegenübergestellt werden. Die Untersuchungen führten zu folgenden Ergebnissen: Die Normalwerte zeigten eine beträchtliche Schwankung aller Meßwerte. Die maximale Resorptionszeit (MR) lag zwischen der 3. und 6. Std (Mittel 4,4 Std), die Gipfelaktivität für die Gesamtaktivität zwischen 3 und 9,9% (Mittel 5,6%) und für die Lipidaktivität zwischen 1,0 und 5,0% (Mittel 2,9%) der verabfolgten Dosis pro 1000 ml Plasma. Der prozentuale Anteil der Lipidaktivität an der Gesamtaktivität schwankte im Gipfelpunkt zwischen 67 und 20% (Mittelwert 48%). Faßt man die Gipfelpunkte zu einer Null-Ordinate zusammen, beginnt der Anstieg der Resorptionsphase zwischen der 2. und 3. Std vor Erreichen des Gipfelpunktes. Die mittlere Lipidaktivität betrug 1 Std davor 1,17±0,19% (Standardabweichung), im Gipfelpunkt 2,93±1,56% und 2 Std danach 1,65±0,97% der applizierten Dosis. Die Blutaktivitätskurven bei Krankheiten der Verdauungsorgane lagen zum Teil im normalen Streubereich, zum Teil (acht Patienten) darüber, zum Teil darunter (neun Patienten). Krankheitsspezifische Kurventypen ergeben sich jedoch nicht. Das wird besonders am Beispiel nach Billroth II Magenresezierter demonstriert. Aus den Untersuchungen wird der Schluß gezogen, daß die enterale Resorptionsleistung nur einer von mehreren Faktoren ist, die den Kurventyp bestimmen. Das schränkt die diagnostische Brauchbarkeit des131J-Öltoleranztestes zur Prüfung der Resorptionsleistung erheblich ein.
