    Notizen: The fine structural changes accompanying the regeneration of adrenocortical transplants in the rat were studied with the electron microscope. One-half gland autotransplants were made to the dorsal musculature of male Wistar rats, weighing approximately 120 gm. Transplants were recovered after 2, 4, 7, 14, 21 and 60 days of regeneration. During the first week of regeneration there was an increase in the granular endoplasmicreticulum at the expense of the agranular form. The internal structure of the numerous mitochondria was transformed from the normal tubular and vesicular forms of the zonae glomerulosa and fasciculata, respectively, to a lamellar type. The quantities of free ribonucleoprotein particles also were reduced. During this period the viable cortical cells are considered to be more deeply involved in protein synthesis and growth rather than hormone biosynthesis. Following 14 days of regeneration and thereafter, the characteristics of the mal intact adrenal cortex became established. Highly osmiophilic “dark cells,” present in the inner zones of the normal intact adrenal cortex, firstappeared after 14 days of regeneration, and were widely scattered throughout the cortexafter three weeks. After two months of regeneration, the “dark cells” were again concentrated in the inner cortical zones. During this latter period, hormone biosynthesis appears to be the major cell function. Possible structural-functional relationships are discussed.

    Notizen: The lungs of male LAF1 mice were locally irradiated with doses of 5, 9, and 13 Gy. The animals were killed at times corresponding to the appearance of histologically identifiable fibrosis or, for 13 Gy, at the LD50 for these doses and strain of mouse: 63, 36, and 28 weeks postirradiation (PI) respectively. Lungs were excised, incubated in buffer alone, or partially digested with enzymes for determination of relative glycosidase resistance, fixed with ruthenium red/Triton X-100 for demonstration of basal laminar anionic sites, and processed for electron microscopy. Sham-irradiated and untreated control groups (0 Gy, 0 times) were also processed. Tissue was examined ultrastructurally and alterations in both alveolar and capillary basal laminar anionic sites were quantitated. In each of the doses examined the number of anionic sites surpassed normal levels; however, the glycosidase resistance of the regenerated laminae at these late time points was not significantly altered from controls. This contrasts with the marked increase in the glycosidase resistance of laminae regenerating from radiation damage (4-12 weeks PI) reported earlier. The increased numbers of anionic sites were compared to expected values derived from models based on compensatory synthesis and continued accumulation and indicate close correlation with certain aspects of the compensatory synthesis model but not with others. The effects on basal laminar permeability, basal laminar thickening, and fibrotic induction are discussed.

    Notizen: The purpose of this study was to determine the dimensions of capilaries in various tissues, in vivo, in frogs, mice, and pups. Capillaries were measured in the web, strap muscles of the neck, mesentery and bladder of frogs, and in the mesentery of mice and pups. The light source was a fused quartz rod transilumination apparatus. In all cases the exposed tissues were kept moist by a continuous flow of Ringer's solution over their surfaces.The mean values found for the tissues of the frog are web 11.94 μ, skeletal muscle 11.30 μ, mesentery 11.97 μ, and bladder 11.40 μ. The mesenteric capillaries of the mice and pups had a mean value of 4.64 μand 7.36 μ respectively.Difficulties encountered during the course of observation included vibrations of the optical equipment, movement of the tissue under the microscope, fogging of the objective lens, and the determining of which of the microscopic vessels were “capillaries.”A statistical analysis of the data indicates that the variance in the measurements recorded on the capillaries of a single animal is greater than the variance from animal to animal.

    Notizen: Snell adrenocortical tumor 494 was implanted into male Sprague-Dawley rats and recovered 7, 14, 21, 28 or 35 days following initial detection by palpation (7-10 days following transplantation). Electron microscopic, stereologic and biochemical analyses of the tumor were compared to adrenals of normal animals to serve as a baseline for further studies of the effects of chemotherapeutic agents on tumor cells. Tumor cells possessed oval or elongated mitochondrial profiles with tubular cristae, one or two very large (〉 5μ) lipid droplets, abundant ribosomes and coated vesicles, and sparse rough and smooth endoplasmic reticulum. Stereologic evaluation revealed that tumor lipid volume was 41% and mitochondrial volume 29% that of the normal adrenal controls. Tumor nuclei were 2.5 times larger than adrenocortical nuclei while cellular volumes were similar. On a net weight basis, tumor cholesterol was 55%, cholesterol ester 2.2%, and lipid phosphate 25% of respective mean values for normal adrenal glands. The tumor cholesterol: cholesterol ester ratio progressively decreased with time, but remained 18-fold greater than the normal adrenal. Plasma corticosterone levels in tumor-bearing rats were elevated 3-fold by 14 days after initial detection. The adrenals of the tumor-bearing host exhibited marked involution, the extent of which was directly related to tumor size.

    Notizen: In human mammary carcinoma, positive immunohistochemical staining for p53 protein is not always indicative of mutation in the p53 gene. Although positive staining is seen in excess of 50% of tumours, mutations have been found in only some 20% of cases. In this presentation, positive p53 staining in mammary carcinomas will be related to the presence and absence of mutation and other possible underlying mechanisms.In some positively stained tumours a mutation has been found. In others, no mutation has been demonstrated and apart from possible stabilisation by a protein such as MDM2, there are alternative underlying mechanisms for this discrepancy. Wild type p53 is elevated in response to DNA damage. This effect can be seen in patients given pre-operative chemotherapy and in cell lines irradiated with UV light and with x-rays. Strong positive staining in scattered nuclei has been found in cell lines with activated ras and myc genes. We postulate that this may also be the reason for similar patterns observed in human tumours.Comparable mechanisms may be active in inherited cancers. Although positive p53 staining in some Li-Fraumeni syndrome patients is associated with mutation, in other Li-Fraumeni-like families, no mutation has been found despite positive staining in tumour and normal tissues.Whatever the mechanism underlying the stabilisation of the protein, increased expression of p53 protein in the majority of tumour cells appears to be associated with poor prognosis in breast carcinoma.

    Notizen: In order to characterize the endogenous gene product for rad (ras-related protein associated with diabetes), we prepared antibodies to synthetic peptides that correspond to amino acids (109-121, 178-195, 254-271) within the protein. These antibodies were used to analyze the expression, structure, and function of rad. Western analysis with these antibodies revealed that rad was a 46 kDa protein which was expressed during myotube formation. Further, immunolocalization studies showed that rad localized to thin filamentous regions in skeletal muscle. Interestingly, when muscle biopsies from diabetic and control Pima Indians were compared, no differences in rad protein or mRNA expression were observed. Similarly, no differences were observed in protein expression in diabetic and control Zucker diabetic fatty (ZDF) rats. Functional analysis of muscle rad revealed that its GTP-binding activity was inhibited by the addition of N-ethylmaliemide, GTP, GTPγS, and GDPβS but not ATP or dithiothreitol. Moreover, cytosol-dependent rad-GTPase activity was stimulated by the peptide corresponding to amino acids 109-121. Antibodies corresponding to this epitope inhibited cytosol-dependent rad-GTPase activity. Taken together, the results indicate that 1) rad is a 46 kDa GTP-binding protein localized to thin filaments in muscle and its expression increases during myoblast fusion, 2) expression of rad in Pima Indians and ZDF rats does not correlate with diabetes, and 3) the amino acids (109-121) may be involved in regulating rad-GTPase activity, perhaps by interacting with a cytosolic factor(s) regulating nucleotide exchange and/or hydrolysis.     Notizen: Rats bearing adrenocortical carcinoma 494 were injected daily for 7, 14, or 21 days with aminoglutethimide (AG) or o,p′-DDD. Reversibility of these steroidogenic inhibitors was determined by injecting other animals for either 14 or 21 days and sacrificing them 14 days later. While the drugs had little effect on body or tumor growth, plasma corticosterone levels were reduced a maximum of 88% in normal and 95% in tumor-bearing rats during AG chemotherapy. These levels were unaltered in normal rats by o,p′-DDD and reduced a maximum of 64% in tumor-bearing animals. Relative adrenal weights generally increased during chemotherapy and then returned to control levels. These changes were mainly due to alterations in the lipid and mitochondrial volume fractions. Lipid increased with both drugs while mitochondria increased with o,p′-DDD and decreased with AG. Cholesterol ester levels paralleled the lipid stereology more closely with AG than o,p′-DDD. With both drugs the most notable changes in tumor fine structure was a decrease in mitochondrial internal membranous vesicles and matrical density. Adrenal mitochondria had the irregular, elongated forms characteristic of tumor-bearing animals and were vacuolated (AG) or had internal rings (o,p′-DDD). The large lipid droplets observed during chemotherapy with both drugs were replaced by numerous small droplets in recovery periods.

    Notizen: Postcastrational adrenocortical carcinomas in the CE/Ki inbred strain of mice and the adrenals of noncastrated CE/Ki mice were studied using light and electron microscopic techniques. Most of the tumors appeared as large nodules of cells separated by septae comprised of collagen and blood sinusoids. The majority of tumor cells (Type 1) showed few or no lipid droplets (sudanophobic), polymorphic hyperchromatic nuclei, lack of SER, abundant RER and free ribosomes, prominent Golgi complexes, and few mitochondria with scant internal membranes. Clusters of Type 1 cells were surrounded by a basal lamina. In contrast, Type 2 cells revealed abundant and dilated tubules of SER, large number of lipid droplets and mitochondria with tubulovesicular cristae. These results suggest that Type 2 cells were probably active in steroid hormone synthesis and secretion while Type 1 cells were highly anaplastic and apparently non-steroid-secreting cells.

    Notizen: During cardiac valve morphogenesis, a series of interactions between the mesodermal-derived myocardium and the overlying endothelium lead to condensed leaflet structure formation. At the atrioventricular (AV) canal, endocardial cells are transformed by specialized underlying myocardial cells into endocardial cushions, and then remodeled into mitral and tricuspid valves. Aortic and pulmonary valves develop by a similar mechanism in the primitive outflow tract. Few genes exhibit restricted spatiotemporal expression in these critical embryonic structures, thus limiting the clues to the sequence of molecular events necessary for valvulogenesis. Apolipoprotein J (ApoJ), a secreted glycoprotein expressed in a variety of cell types at tissue interfaces, exhibits a highly restricted and dynamic expression pattern in the developing heart. ApoJ transcripts were detected in mice at day 9.0 of gestation in the wall of the developing truncus arteriosus. By day 10, intense signal occurred in a thin layer of myocardial cells adjacent to developing endocardial cushions of both atrioventricular canal and truncus arteriosus. No apoJ mRNA was present in the overlying endocardial cushions until day 13.5 when prevalvular condensation begins. Intense expression occurred in the stromal connective tissue throughout leaflet formation. The highly restricted spatiotemporal expression pattern of apoJ in the developing heart implicates its role in the morphogenesis of the AV canal and outflow tract into cardiac valves. © 1994 Wiley-Liss, Inc.

    Notizen: Complete understanding of the role of the mammalian main olfactory bulb in sensory processing has remained elusive despite many detailed studies on its anatomy and physiology. Several lines of recent evidence viewed in the context of earlier knowledge have provided new insights into the bulbar mechanisms of olfactory coding. The output cells of the olfactory bulb receive a localized olfactory nerve input and interneuronal input via dendrodendritic synapses on distinct sets of dendrites. The spatial arrangement of granule cell contacts on output cell basal dendrites suggests that lateral inhibitory interactions may occur between neighboring output cells. The input from olfactory receptor cell axons to the bulb also has spatial order, but does not represent a precise map of the receptor surface. Recent studies with antibodies and lectins suggest that different groups of axons from chemically similar receptor cells collect into certain glomeruli, even if the axons originate from cells that are not contiguous in the mucosa. Electrophysiological studies have begun to explore the participation of spatially organized circuits in olfactory processing. The degree to which neighboring output cells respond similarly to odor stimulation, for example, depends on the distance between the cells, with those further apart showing complementary responses. Also, a single output cell can show 2 or more different temporal response patterns when different odors are presented. Intracellular recordings indicate that these responses are shaped by IPSPs. Electrical stimulation during such recordings shows that some mitral cells are excited by nerve inputs close to their glomerular tufts, while they are inhibited by nerve inputs to other parts of the bulb. Finally, recordings from granule and periglomerular cells indicate their potential in mediating components of output cell odor responses. These considerations suggest that the olfactory bulb performs a spatially based analysis on the information coming from the receptor cells. While the spatial organization of the olfactory bulb is probably not faithfully represented in the projections to the olfactory cortex, bulbocortical projections are not random. The fact that spatial factors exist at each of these levels in the olfactory system must be considered in developing models of central olfactory processing. © 1993 Wiley-Liss, Inc.
