    Notizen: Radiofrequency magnetron sputter deposition was used to deposit Ca-P sputter coatings on titanium discs, and these coatings were implanted subcutaneously into the backs of rabbits. Half of the as-sputtered coatings were subjected to additional heat treatment for 2 h at 500°C. X-ray diffraction (XRD) demonstrated that annealing at 500°C changed the amorphous sputtered coating into an amorphous-crystalline apatite structure. Scanning electron microscopic (SEM) examination of the sputtered coatings showed excellent coverage of the substrate surface. Annealing of the 4-μm-thick coatings resulted in the appearence of small cracks. SEM demonstrated that until 4 weeks of implantation, all heat-treated coatings were present and all amorphous coatings were completely or mostly dissolved. Fourier transform infrared spectroscopy showed the formation of carbonate apatite (CO3-AP) on these specimens. Furthermore, XRD analysis showed that these CO3-AP precipitated coatings disappeared after 8 weeks of implantation. On the other hand, SEM inspection of these specimens revealed that the 4-μm heat-treated coating was still partially maintained and that small Ca-P crystals were present on the titanium substrate. On the basis of these results, we conclude that apparently 0.1 μm heat-treated Ca-P sputter coating is of sufficient thicknesses to stimulate carbonate apatite deposition under in vivo conditions. © 1998 John Wiley & Sons, Inc.     Notizen: We investigated the behavior of microgrooved implants in soft tissue using polystyrene implantable disks, either smooth or microgrooved (1-10 μm) on both sides. The implants were placed subcutaneously in a goat for 1, 4, or 12 weeks. Light and transmission electron microscopy showed that fibrous capsule formation around the implants was fairly uniform. After 1 week the implants were covered with a fibrous capsule about 80 μm thick. The collagen matrix was loose, and many inflammatory cells were present. After 4 weeks the matrix was more dense and contained many newly formed blood vessels. At the implant surface a layer of inflammatory cells about 10 μm thick had accumulated. Finally, after 12 weeks the matrix had densified. One cellular layer of inflammatory cells was present at the implant surface. We carried out histomorphometric measurements of capsule thickness, inflammatory layer thickness, and the number of blood vessels. Capsule thickness appeared not to decrease with time. Further, these measurements showed that there were no differences in tissue reaction between smooth and microgrooved implants. On the basis of our observations, we suggest that 1 μm deep and 1-10 μm wide microgrooves do not influence tissue response around polystyrene implants in soft tissue. © 1998 John Wiley & Sons, Inc.     Notizen: The aim of this study was to obtain more information about the bone reaction to titanium and hydroxypatite (HA)-coated titanium implants during the first 3 months after implantation. Therefore, uncoated and coated implants were inserted into the tibia of rabbits for various implantation periods. The histological results demonstrated that although there were no marked differences in bony reaction at the cortical level to the different implant materials, HA-coating appeared to induce more bone formation in the medullary cavity. It was also noted, that 3 months after insertion loss of coating thickness had occurred.

    Notizen: The aim of this study was to investigate bone response to and biostability of three different hydroxyapatite (HA)-coatings. Therefore, coated and uncoated titanium implants were inserted into the tibia of rabbits. Implantation times were at 3 and 12 weeks. The histological evaluation included measurement of the amount of bone apposition to the various implant surfaces. The results demonstrated at 3 and 12 weeks no marked differences in bony reaction at the cortical level to the different implant materials. However, compared with the 3-week specimens, at 12 weeks extensive maturation of the woven bone callus had occurred. In addition, all 12-week implants induced bone formation in the medullary cavity. It was also noted that all HA-coatings showed loss of coating thickness. Quantitative determination of bone contact demonstrated that all 12-week implants showed the same high amount of bone apposition. © 1993 John Wiley & Sons, Inc.

    Notizen: Plasma-sprayed hydroxylapatite coatings on metals such as titanium have been investigated for many years and have shown a good biocompatibility when implanted in bony tissues. Radiofrequency magnetron sputtering was used as an alternative method to deposit thin films of hydroxylapatite on titanium substrates. X-ray diffraction demonstrated that the sputtered layer was crystalline with a preferred (001) crystallographic orientation with the C-axis perpendicular to the substrate surface. Scanning electron microscopy showed that the deposited films had a uniform and dense structure. The calcium phosphate ratio varied between 1.5 and 2.0, as determined by energy-dispersive X-ray analysis. The in vitro dissolution appeared to be determined by the degree of the coating's crystallinity. © 1994 John Wiley & Sons, Inc.

    Notizen: To evaluate the effect of surface treatment and surface microtexture on cellular behavior, smooth and microtextured silicone substrata were produced. The microtextured substrata possessed parallel surface grooves with a width and spacing of 2.0 (SilD02), 5.0 (SilD05), and 10 μm (SilD10). The groove depth was approximately 0.5 μm. Subsequently, these substrata were either left untreated (NT) or treated by ultraviolet irradiation (UV), radiofrequency glow discharge treatment (RFGD), or both (UVRFGD). After characterization of the substrata, rat dermal fibroblasts (RDF) were cultured on the UV, RFGD, and UVRFGD treated surfaces for 1, 3, 5, and 7 days. Comparison between the NT and UV substrata revealed that UV treatment did not influence the contact angles and surface energies of surfaces with a similar surface topography. However, the contact angles of the RFGD and UVRFGD substrata were significantly smaller than those of the UV and NT substrata. The dimension of the surface microevents did not influence the wettability characteristics. Cell culture experiments revealed that RDF cell growth on UV-treated surfaces was lower than on the RFGD and UVRFGD substrata. SEM examination demonstrated that the parallel surface grooves on the SilD02 and SilD05 substrata were able to induce stronger cell orientation and alignment than the events on SilD10 surfaces. By combining all of our findings, the most important conclusion was that physicochemical parameters such as wettability and surface free energy influence cell growth but play no measurable role in the shape and orientation of cells on microtextured surfaces. © 1995 John Wiley & Sons, Inc.

    Notizen: Various studies already have shown that the occurrence of oral implant failure is higher in the maxilla than in the mandible. To learn whether Ca-P coatings can improve the success rate of oral implants in the maxilla, three different plasma-sprayed, Ca-P-coated, self-tapping Brånemark® implants were inserted in the trabecular bone of the maxilla. Before the insertion of the implants, the two first upper premolars of 16 goats were bilaterally extracted. Four months later, each animal received four types of implants: three different Ca-P-coated types and one uncoated. After an endosseous period of 6 months, the implants were provided with permucosal abutments. Four months later the animals were killed. At the end of the experiment, it appeared that 10 of the 16 installed noncoated implants had failed while of the 48 Ca-P-coated implants, only 6 had failed. All successful implants were retrieved and prepared for histomorphometrical evaluation of the bone and gingiva response. The Ca-P-coated implants showed a significantly greater percentage of bone in contact with the implant surface compared with the uncoated implants. The length of the epithelium was not significantly different for the coated compared to the uncoated implants, but the connective tissue was significantly thicker for the noncoated implants than for the Ca-P-coated implants. Also, measurements revealed that all coatings showed reduction in thickness. On the basis of these findings, we concluded that the application of Ca-P coatings (1) improves the bone-implant reaction, although all coatings reduced in thickness, and (2) is of benefit during the healing period in less mineralized trabecular bone. © 1997 John Wiley & Sons, Inc.

    Notizen: The aim of this study was to investigate the bone response to calcium phosphate (Ca-P) plasma-spray and radiofrequency magnetron sputter-coated implants with comparable roughness. Therefore, tapered conical screw designed implants were installed in the trabecular bone of the femurs of nine goats. They were provided with two types of coatings, a plasma-spray dual coating of fluorapatite and hydroxyapatite (FA/HA-PS) and a titanium plasma-spray coating, covered with an amorphous Ca-P magnetron sputtercoating (TPS/Ca-P-a). These implants were evaluated histologically and mechanically after 3 months of implantation. A well-controlled method to apply and measure a torsional force to load the screw-type implants to the point of failure was introduced. All implants healed uneventful and were well fixed. No significant difference (Student t test, p 〉 0.05) for the torsional failure force was measured for both type of coatings. Nevertheless, SEM revealed differently situated fracture planes. Light microscopy showed intimate bone-implant contact for both types of coatings; original drill margins were still visible. A lamellar type of bone with some remodeling lacunae was shown. Histomorphometry revealed a higher percentage of bone contact for the FA/HA-PS-coated implants (student t test, p 〈 0.05). Measurement of the amount of bone revealed more bone mass around TPS/Ca-P-a-coated implants (analysis of variance and Tukey multiple comparison, p 〈 0.05). © 1997 John Wiley & Sons, Inc.

    Notizen: In previous studies we developed a RF magnetron sputter technique for the production of thin Ca-P coatings. With this technique coatings can be produced that vary in Ca/P ratio as well as in structural appearance. The aim of this investigation was to obtain more understanding of the biological behavior of these coatings by way of in vitro experiments. The effect of noncoated titanium (Ti) and three different Ca-P-sputtered surfaces on the proliferation and differentiation (morphology and matrix production) of osteoblast-like cells was studied. Proliferation was determined using counting procedures; morphology was studied by scanning electron microscopy (SEM) and transmission electron microscopy (TEM). Fluorescent markers and energy-dispersive X-ray microanalysis (EDX) were used to obtain quantitative and compositional information about the resultant calcified extracellular matrix (ECM). Results demonstrated that proliferation of the osteoblast-like cells was significantly (p 〈 0.05) higher on noncoated than on Ca-P-coated samples. On the other hand, more mineralized ECM was formed on the coated surfaces. In addition, TEM confirmed that the cells on the coated substrates were surrounded by ECM with collagen fibers embedded in crystallized, needle-shaped structures. On the basis of these findings, we concluded that: (1) the investigated Ca-P sputter coatings possess the capacity to activate the differentiation and expression of osteogenic cells, and (2) bone formation proceeds faster on Ca-P surfaces than on Ti substrates. Further, this bone-inductive effect appeared to be dependent on the Ca-P ratio of the deposited coatings. © 1998 John Wiley & Sons, Inc. J Biomed Mater Res, 40, 464-474, 1998.

    Notizen: RF magnetron sputter deposition was used to deposit thin calcium phosphate (hydroxyapatite) layers on titanium alloy substrate material. We determined the precise amount of calcium, phosphorus, oxygen, and hydrogen in these films by combining two ion-beam analysis techniques: Rutherford backscattering spectrometry (RBS) and elastic recoil detection (ERD). Variables were power level, partial oxygen pressure, or additional water vapor. Also examined was the influence of additional bias power to the substrates during deposition and the effect of annealing on the final composition of the deposited layers. Measurements showed that the Ca/P ratio decreased with increasing oxygen pressure or decreasing sputtering power. In addition, the Ca/P ratio increased when a bias was applied to the substrates. The O/P ratio of the films decreased with additional oxygen pressure but increased when additional water vapor was applied during deposition. All as-deposited films showed a higher hydrogen content than stoichiometric HA. The hydrogen content in the films deposited with water vapor was more than 7 times higher than in the films deposited under pure argon conditions. After annealing, the hydrogen content decreased to about 3.5 at % whereas in stoichiometric HA the hydrogen content amounts to 4.5 at %. After annealing, the oxygen concentration in the film also decreased. We assume that hydrogen disappears out of the film as H2O during annealing. For the targets used in these experiments, sputter deposition at 400 W, with additional water vapor and annealing at 500°C, produced films with a stoichiometry closest to that of hydroxyapatite. © 1998 John Wiley & Sons, Inc.
