Notes: Abstract We obtain essentially Doppler free spectra of the naturally occuring isotopes of Mg+, which are bound in a Penning trap, by using a frequency stabilized laser to continuously cool the ions, while the scatter rate from a second, frequency swept laser is, monitored. We show that the magnetron motion as well as the cyclotron and axial motion can be minimized. Line position measurements yielding resonance transition energy, isotope and hyperfine shifts are reported.

Notes: Abstract We report on the photorefractive properties of a Cr-doped (K1−x Na x )2A−2 (Sr y Ba1−y )2−A Nb10O30 (x=0.586, y=0.659, A=1.12) single crystal in the near-infrared spectrum. The sample exhibits photorefractivity for wavelengths up to at least 840 nm where the steady-state two-beam coupling gain is found to be larger than 2 cm−1. Photorefractive gain and decay rate are measured as a function of wavelength, grating spacing and intensity. The wavelength dependence of gain fluctuations in two-beam coupling are also measured.

Notes: Summary Handling of small volumes and small amounts of analytes is important in biological science. When the sample amount is limited it is beneficial to be able to investigate the biological variation in expression and processing of cellular material by screening a single cell or a few cells. In this study matrix-assisted laser-desorption/ionization time-of-flight mass spectrometry (MALDI TOF MS) was used to analyse single small mammalian cells (rat adrenal pheochromocytoma PC 12 cells) grown directly on the sample target. The cells were screened for peptide and protein content and analysed before and after stimulation with nerve-growth factor (NGF), a neurotrophic factor. Single-cell analysis of on-target-grown cells was also compared with whole cells placed on top of a matrix layer and with cellular extracts analysed using the conventional sample-matrix mixture technique. The analysis of on-target-grown cells resulted in mass spectra of greater intensity, thus providing more information about cellular peptide and protein content. Stimulation with NGF resulted in the detection of over 14 new peptides and proteins, demonstrating the potential of this technique for the investigation of cellular events, direct protein profiling, and cell fingerprinting. This unique and simple strategy provides the possibility of studying intact cells while keeping the cells in contact with other cells and substrates. Furthermore, the often complicated sample handling in single-cell analysis is also eliminated, making the technique more versatile for new users of biotechnology and cell biology.

Notes: Abstract We have applied experimental techniques that were developed for use in atomic frequency standards andclocks to investigations of local Lorentz invariance, the linearity of quantum mechanics, andanomalous long-range spin-dependent forces. These experiments used a hyperfine transition in9Be+ ions in a Penning trap. Recently, we have studied hyperfine transitions in199Hg+ ions in a linear rf trap. Hg+ ions might be used for similar investigations in the future.
