    Notes: Canavalin, the major reserve protein of the jack bean, was obtained in four different crystal forms. From the structure determined by multiple isomorphous replacement in a hexagonal unit cell, the structures of three other crystals were determined by molecular replacement. In two cases, the rhombohedral and cubic crystals, placement was facilitated by coincidence of threefold molecular symmetry with crystallographic operators. In the orthorhombic crystal the canavalin trimer was the asymmetric unit. The rhombohedral, orthorhombic and cubic crystal structures were subsequently refined using a combination of several approaches with resulting R factors of 0.194, 0.185 and 0.211 at resolutions of 2.6, 2.6 and 2.3 Å, respectively. Variation in the conformation of the molecule from crystal to crystal was small with an r.m.s. deviation in Cα positions of 0.89 Å. Packing is quite different among crystal forms but lattice interactions appear to play little role in the conformation of the molecule. Greatest variations in mean position are for those residues that also exhibit the greatest thermal motion. Crystal contacts in all crystals are mediated almost exclusively by hydrophilic side chains, and three to six intermolecular salt bridges per protein subunit are present in each case.

    Notes: Three crystal forms of the sweet-tasting protein thaumatin from the African berry Thaumatococcus daniellii have been grown. These include two naturally occurring isoforms, A and B, that differ by a single amino acid, and a recombinant form of isoform B expressed in yeast. The crystals are of space groups C2 with a = 117.7, b = 44.9, c = 38.0 Å, and β = 94.0°, P212121 with a = 44.3, b = 63.7 and c = 72.7 Å, and a tetragonal form P41212 with a = b = 58.6 and c = 151.8 Å. The structures of all three crystals have been solved by molecular replacement and subsequently refined to R factors of 0.184 for the monoclinic at 2.6 Å, 0.165 for the orthorhombic at 1.75 Å, and 0.181 for the tetragonal, also at 1.75 Å resolution. No solvent was included in the monoclinic crystal while 123 and 105 water molecules were included in the higher resolution orthorhombic and tetragonal structures, respectively. A bound tartrate molecule was also clearly visible in the tetragonal structure. The r.m.s. deviations between molecular structures in the three crystals range from 0.6 to 0.7 Å for Cα atoms, and 1.1 to 1.3 Å for all atoms. This is comparable to the r.m.s. deviation between the three structures and the starting model. Nevertheless, several peptide loops show particularly large variations from the initial model.

These spines, and the outer wall layer are derived from preformed structures (bars) found in the cytoplasm prior to encystment.     Notes: Summary 1. Concentric bodies have been detected in the mycobionts of at least forty-three species of lichens, and their presence in two non-lichenized fungi is reported here for the first time. The structure of the bodies appears to be closely similar in all species. 2. A detailed examination of the concentric bodies in electron micrographs of thin sections of Peltigera aphthosa Willd, indicates that the bodies are basically isodiametric in organization. The designation “concentric bodies”, a translation of the term “konzentrische Körper” first used by Peveling (1969a), is therefore preferred to the original name “ellipsoidal bodies” (Brown and Wilson, 1968). 3. Each concentric body is composed of two zones of osmiophilic material surrounding an electron-transparent core. The inner zone is limited internally by a membrane-like boundary. The outer zone has a variable appearance, but often contains radially arranged stainable structures which appear as lamellae in tangential sections. Concentric bodies may collapse under stress into discs with elimination of the core but without apparent rupture of the surrounding material. It is concluded that the bodies are empty or filled with gas or liquid. 4. The bodies occur singly or in clusters and are frequently surrounded by electron-transparent haloes. Clusters of bodies lie within a distinctive matrix which is almost invariably associated with the cell nucleus. 5. Concentric bodies have been found in all types of hyphae in the lichen thallus but not in the asci or ascospores. 6. Interpretations of previous workers are discussed in relation to the present findings, and hypotheses as to the nature of the concentric bodies are put forward. 7. No information is as yet available concerning the origin, development and functions of the concentric bodies, and the question of their possible significance for the lichenized condition remains open.

    Notes: Abstract The unitary nature of the mitochondrion and the characteristic flattened finger-like morphology of the cristae were demonstrated in the Cryptophyceae. Hemiselmis rufescens contained an unbranched vermi-form mitochondrion in contrast to the variously branched complex, comprising an interconnected peripheral and central reticulum, in Chroomonas sp. and strains of Cryptomonas. The systematic value of the shape and distribution of the mitochondria in the examined genera was suggested.

    Notes: Summary Progressive joint damage characterises rheumatoid arthritis despite treatment with slowacting drugs such as penicillamine. We examined a cohort of 145 RA patients, treated with 250 or 500 mg penicillamine daily for 18 months to study progressive joint damage measured using Larsen's standard radiographs. Overall damage increased significantly over 18 months at both doses of penicillamine. Radiological changes between 6–18 months were studied in detail in 55 cases. They were divided into rapidly progressive (increases in Larsen score of more than 5) or slowly progressive (increases in Larsen score of 5 or less). Overall clinical response, visual analogue pain score, ESR, haemoglobin and platelet count were significantly lower in the slowly progressive patients; articular index and duration of morning stiffness were slightly lower; latex titre, RAHA titre, joint size and white cell count showed no differences between groups. There is an indirect relationship between progressive joint damage and some clinical and laboratory measures. The reasons underlying our failure to control progression in some cases need further definition.

    Notes: Summary Post-meiotic tetraspore mother cells of Corallina officinalis L. have been studied by light and electron microscopy. During the course of post-meiotic cellular reorganisation each nucleus becomes surrounded by a complex of precisely oriented endoplasmic reticulum, termed nuclear endoplasmic reticulum. A distinctive feature of this relationship is an electron dense substance in contact with the nuclear surface and extending as groundplasm between the ER cisternae as far as the outer limits of the complex, where it gives place to the ribosome-containing matrix of the general cytoplasm. There is circumstantial evidence to indicate that the extracisternal electron dense material is a product of nucleo-cytoplasmic interaction, and that it is involved in the assembly of ribosomes. The nuclear endoplasmic reticulum appears to be active in the production of smaller swollen cisternal elements, which form frequently anastomosing reticular tracts in the regions between adjacent nuclei. There is structural evidence of vesicular transport of material from the swollen cisternae to the proximal (“forming”) face of the Golgi apparatus. These events are thought to be of fundamental importance in achieving the cellular reorganisation and transformation which occurs after the second meiotic division.
