    Notes: Summary Limonin degradingRhodococcus fascians was immobilized by entrapment in alginate, k-carrageenan, agarose and polyacrylamide gels. Except this latter, gels were used both with and without polyethyleneimine treatment followed by glutaraldehyde crosslinking. Coated derivatives showed lower activity and stability and higher diffusional limitations that uncoated ones. Immobilized cells in k-carrageenan gave, globally, the best results.

    Notes: Summary β-Glucosidase from sweet almond was immobilized onto a nylon support and used to hydrolyze picrocrocin, the glycoside precursor of the saffron essential volatile oil, safranal. The nylon support was derivatized as hydrazide and the enzyme attached through Schiff base to bonds. The coupling efficiency was 46.8%, the immobilization yield 29.5%, and the derivative showed 24.2 and 4.0 U/mg activity for p-nitrophenylglucoside and picrocrocin, respectively, as substrates. Kinetic parameters of the immobilized derivative were determined, with picrocrocin as substrate, showing KM=7.2 mM and Vmax=4,0 U/mg. Glucose behaved as a competitive inhibitor (Ki=95.0 mM). The immobilized derivative was thermally stable up to 45°C; from that temperature onwards thermoinactivation occured. The operational deactivation showed a biphasic pattern, t1/2 being 4.2 days for the first four days of continuous operation, and 20.1 days from that point on. The immobilized enzyme lost a 50% of its initial activity after 30.7 days of storage at 4°C.

    Notes: Abstract Limonin can be effectively degraded byRhodococcus fascians cells. These bacteria can be entraped in κ-carrageenan, and used in a continuous stirred tank reactor to degrade limonin in a continuous process. The effects of temperature limonin concentration, dilution rate, and aeration on the reactor behaviour have been tested, and the results correlated with changes in limonin conversion, substrate degradation rate, and free and immobilized biomass. Results showed that the immobilized cells were able to debitter limonin-containing media and the immobilized biomass was quite stable throughout the operational conditions tested. A population of free biomass was present in the reactor, the quantity of which was dependent on dilution rate. The immobilized bacteria increased its limonin-degrading capability when the substrate concentration was increased. The aeration was not strictly necessary for limonin degradation. Additionally, the immobilized cells were active and stable for more than 2 months of continuous operation, and were able to recover their limonin-degrading capability when used intermittently. Finally, none of the main components of a juice was noticeably altered during limonin degradation, so the reactor response was good enough to consider its application.

    Notes: Summary A sensor based on immobilized tyrosinase and a pH electrode has been constructed. The calibration curve of the sensor is linear up to 0.03–0.24 mg/ml depending on the substrate used. The sensor retains 50% of its sensitivity after 10–11 hours, showing 6–7 hours of stationary activity in continuous working.     Notes: Abstract Grape (V. vinifera L. cv Gamay Fréaux) cells were entrapped in calcium alginate beads. The effect of bead diameter and gel concentration on the viability of the cells was checked. The reduction of 2,3,5-triphenyltetrazolium chloride as well as O2 consumption were used as viability test, and their results compared. The existence of diffusional limitations for O2 introduces an unaccuracy as high as 25% for the O2 consumption method. The reduction assay is more simple, precise and reliable.

The bioreactor combines microporous plates commercially available and industrial pectinases immobilized on nylon membranes in a cross-flow configuration. The kinetic behaviour of the reactor for different recirculation flow rates has been determined. Fresh apricot juice has been continuously clarified in the bioreactor with excellent results.
