    Notes: Summary Proteinase K digestions of bacteriorhodopsin were carried out with the aim of characterizing the membrane-embedded regions of the protein. Products of digestions for two, eight or 24 hours were separated by high-pressure liquid chromotography. A computerized search procedure was used to compare the amino acid analyses of peptide-containing peaks with segments of the bacteriorhodopsin sequence. Molecular weight distributions of the products were determined by sodium dodecylsulfate-urea polyacrylamide gel electrophoresis. The structural integrity of the protein after digestion was monitored through the visible absorption spectrum, by X-ray diffraction of partially dried membranes, and by following release of biosynthetically-incorporated3H leucine from the digested membranes. During mild proteolysis, bacteriorhodopsin was cleaved near the amino and carboxyl termini and at two internal regions previously identified as being accessible to the aqueous medium. Longer digestion resulted in cleavage at new sites. Under conditions where no fragments of bacteriorhodopsin larger than 9000 mol wt were observed, a significant proportion of the digested membranes retained diffraction patterns similar to those of native purple membranes. The harshest digestion conditions led to complete loss of the X-ray diffraction patterns and optical absorption and to release of half the hydrophobic segments of the protein from the membrane in the form of small soluble peptides. Upon cleavage of aqueous loop regions of the protein, isolated transmembrane segments may experience motion in a direction perpendicular to the plane of the membrane, allowing them access to protease.

    Notes: It has been found that the synthesis conditions and subsequent annealing treatments can significantly change the magnetic and transport behavior of the colossal magnetoresistive materials of the general formula La1−zAzMnO3, where A is a divalent ion. In order to clarify the role of vacancies within this structure, resistivity and magnetization measurements have been carried out on a series of samples derived from the parent compound LaMnO3, with La and Mn vacancies introduced by systematically varying the oxygen annealing conditions. Previous studies have shown that for a given carrier concentration, the Curie temperature of the paramagnetic to ferromagnetic transition (Tc) increases as the tolerance factor of the perovskite structure increases and then begins to decrease slowly for even higher tolerance factors. Generally, Tc also increases with the Mn4+ content, consistent with a double exchange mechanism responsible for ferromagnetism. In this study Tc was found to decrease as the vacancy concentration (and therefore the Mn4+ concentration) and tolerance factor both increase. The magnetic and transport data are discussed in the light of structural information obtained from neutron diffraction studies. In the present study it is found that La and Mn vacancies play a significant role in determining the physical properties of these materials: for high vacancy concentrations (x,y≥0.080) the magnetic properties are analogous to those of spin glasses associated with disorder and frustration. Semiconductorlike resistivity was observed for all samples at all temperatures. © 1998 American Institute of Physics.

    Notes: Apparatus for collecting X-ray diffraction data from protein crystals at pressures up to 0.2 GPa is described. The modified techniques required for crystal mounting, determination of crystal orientation in the beryllium pressure cell, and centering in the X-ray beam are described. Special attention is required for intensity absorption corrections due to the marked attenuation (90%) of the beam. Data have been collected from tetragonal hen egg white lysozyme crystals at 1000 atm to 2.0 Å nominal resolution using Cu Kα radiation (1 atm = 1.01325 × 105 Pa). The crystals, crystallized in 0.86 M NaCl, require 1.4 M NaCl in the stabilizing solution to prevent cracking at several hundred atmospheres and higher. The crystals show no measurable hysteresis when cycled through several 1 to 1000 atm transitions. Altered intensities relaxed to their new value within 1 min on pressurization, but required about 90 rain to recover their starting values on pressure release. The unit-cell volume decreases 1.1% at 1000 atm.
