    Notes: A gas chromatographic system for the simultaneous acquisition of hydrocarbon and sulfur chromatograms was developed. Detection of sulfur compounds is achieved using a sulfur chemiluminescence detector (SCD) mounted in series with a flame ionization detector (FID). A constant fraction of the effluent of the FID is transferred to the SCD by means of a fixed restrictor. Unlike previous versions of this approach, the FID is not used to generate the chemiluminescent sulfur species. Rather, the FID is operated under optimum conditions for hydrocarbon analysis and a furnace is used to generate the chemiluminescent sulfur species. The system permits dual acquisition of the hydrocarbon and sulfur signals in a single analysis with a single column, since the detectors are operated in a serial fashion. The application of sulfur simulated distillation using this approach was examined, since this requires simultaneous universal and sulfur selective detection. Precision of absolute response of both the FID and SCD was typically less than 2% RSD for a standard reference material.

    Notes: The capillary gas chromatographic (CGC) analysis of the der-matological drug trans-acitretin (NeotigasonR) and its cis metabolite is described. Separation of the methyl ester derivatives can be achieved on a 90% biscyanopropylsiloxane phase. The importance of using cold on-column injection and short, thin film capillary columns is discussed. For patients treated with the prodrug of acitretin, etretinate (TigasonR), i.e. the ethyl ester of Neotigason, three compounds have to be separated. Selectivity tuning is required for successful CGC separation. An alternative can be found in the selectivity of ion monitoring mass spectroscopy. Analysis of plasma samples involves liquid-liquid extraction, a derivatization step, and HPLC purification.

    Notes: Polynuclear aromatic hydrocarbons (PAHs) are recovered from a soil with a high carbon content (ca. 50%) with supercritical fluid extraction (SFE) as well as with conventional Soxhlet extraction. The influence of temperature and modifier volume on SFE efficiency and the effect of a combined liquid/solid trap for analyte collection are investigated in this study. Such traps, which make analyte collection and clean-up possible in one step, are compared with conventional analyte collection in pure organic solvents. A comparison between reproducibility and efficiency of SFE and Soxhlet extraction is presented.

    Notes: Tetrachlorodiphenoquinones have the same exact mass and elemental composition as the toxic environmental contaminant 2,3,7,8-tetrachlorodibenzo-p-dioxin. However, analysis of 3,3′-5,5′-tetrachlorodiphenoquinone showed a pronounced tendency toward chemical reduction in the mass spectrometer to the quinol compound, producing a molecular ion two mass units higher than 2,3,7,8-tetrachlorodibenzo-p-dioxin. Distinct differences were also apparent between the mass spectral fragmentation patterns of 3,3′,5,5′-tetrachlorodiphenoquinone and 2,3,7,8-tetrachloridibenzo-p-dioxin. The 3,3′,5,5′-tetrachlorodiphenoquinone spectrum shows a successive loss of carbon monoxide, with the most prominent fragment corresponding to loss of two molecules of carbon monoxide plus chlorine. In the mass fragmentation of 2,3,7,8-tetrachlorodibenzo-p-dioxin carbon monoxide loss is suppressed, but loss of one molecule of carbon monoxide plus chlorine is a major fragment ion. During an alumina column clean-up procedure 3,3′,5,5′-tetrachlorodiphenoquinone did not coelute with the fraction containing 2,3,7,8-tetrachlorodibenzo-p-dioxin. This evidence indicates that tetrachlorodiphenoquinones are unlikely to interfere with mass spectrometric determination of 2,3,7,8-tetrachlorodibenzo-p-dioxin in environmental samples.

    Notes: The intact ion yields of phosphotyrosine, phosphothreonine and mono- and diphosphoserine residue-containing peptides have been compared with the non-phosphorylated sequences using plasma desorption mass spectrometry. Equimolar mixtures of the phosphorylated (MP) and non-phosphorylated peptides (M) were also analysed. The positive mass spectra of these mixtures show a higher intensity of the [M + H]+ compared with the [MP + H]+. In the negative mass spectrum, the bias towards the [M - H]- compared with the [MP - H]- was reduced, but the spectra generally did not accurately reflect the stoichiometry.

    Notes: Measurements of DNA dynamics are typically limited to the nanosecond timescale because of the nanosecond decay times of commonly used fluorophores such as ethidium bromide and acridine derivatives. We show that the time-resolved anisotropy measurements can be extended to the submicrosecond timescale using metal-ligand complexes. The ruthenium complex [Ru(2,2′,-bipyridine)2(dipyrido[3,2-a : 2′,3′-c]phenazine)]2+, or [Ru(bpy)2(dppz)]2+, was found to display high anisotropy near 0.2, at -60°C in 100% glycerol, when excited in its long-wavelength absorption bands. The decay time of [Ru(bpy)2(dppz)]2+ when bound to DNA is over 100 ns, allowing intensity to be measured to over 400 ns. Other Ru complexes display still longer lifetimes to 750 ns when bound to DNA. This class of fluorophores should enable measurement of DNA dynamics over a wide range of times (from 1 ns to several μs), which were previously inaccessible using time-resolved fluorescence. © 1995 John Wiley & Sons, Inc.
