    Notizen: Histiocytic cells infiltrating the lesions in eosinophilic granuloma of bone as well as in cutaneous histiocytosis X were studied using a murine monoclonal antibody (MA) produced with proliferating cells from an eosinophilic granuloma of bone. This MA reacts with Langerhans cells (LC) of normal human skin or mucous membranes and with proliferating cells of eosinophilic granuloma of bone and skin lesions of Letterer-Siwe disease, as shown by immunohistochemistry and immunogold labelling.As other murine MA's obtained after immunization with human cortical thymocytes, this MA immunoprecipitates the 49-kDa CD la antigen found on human LC and thymic-cell surfaces but not its breakdown product after treatment with trypsin, as demonstrated by analysis of immunoelectron labelling, cytofluorometry and gel electrophoresis. This first production of a CD la MA from an eosinophilic granuloma supports the concept of Langerhans-cell histiocytosis.

    Notizen: The identification of mononuclear cells extracted from cutaneous tumours (basal cell carcinoma, squamous cell carcinoma and superficial spreading melanoma) has been investigated. The relative numbers of T cells and B cells have been determined using the E-rosette test and the EAC-rosette test. The results have been compared to those of delayed hypersensitivity type reactions. Different cell distribution patterns (E/EAC ratio) have been found in the infiltrates according to the type of tumour.An immunocytochemical technique has been developed for the identification in situ of immunoglobulin-producing cells in the inflammatory infiltrates. In each case the class of immunoglobulin (IgM, IgG or IgA) has been identified and the relative frequency of Ig-producing cells has been determined.The results indicate humoral and cellular immune responses with variations attributable to the type of tumour. In weakly malignant tumours, the infiltrate is characterized by an elevated number of T lymphocytes and numerous plasma cells which secrete all classes of Ig; in highly malignant tumours it is characterized by a reduced number of both T lymphocytes (E rosette) and plasma cells which do not secrete all classes of Ig.

    Notizen: HL-A antigens were found with similar frequency in ninety vitiligo patients and in 341 healthy controls. However there was a significant correlation between HL-A 13 and vitiligo with antithyroid antibodies. This supports the concept that vitiligo is a syndrome rather than a disease.

    Notizen: We report the stimulation of elastase activity of human skin fibroblasts by interleukin-1β (IL-1). Elastase activity was determined by the hydrolysis of the synthetic substrate succinoyl trialanine paranitroanilide (Suc(Ala3)NA). Recombinant human IL-1β induced a concentration-dependent increase in the elastase activity of fibroblasts. The supernatants of keratinocyte cultures that contained IL-1/ETAF showed similar stimulatory activity.

    Notizen: We have studied the expression of involucrin in a variety of keratinization disorders, mostly of genetic origin using an avidin-biotin-peroxidase technique. In normal human epidermis 25% of the living epidermis was labelled. The diseases studied fell into two groups. Diseases with greatly increased involucrin staining including collodion baby (38%), Darier's disease (49%), Flegel's disease (56%), erythrokeratoderma variabilis (6o%), epidermal naevus with epidermolytic hyperkeratosis (45%) and congential bullous (58%) and non-bullous (44%) ichythyosiform erythroderma; and diseases with normal or slightly increased staining, including ichthyosis vulgaris (27%), X-linked ichthyosis (25%), confluent and reticulate papillomatosis (27%) and simple epidermal naevus (28%). These results demonstrate that involucrin exression is altered in some keratinization disorders and suggest that in such conditions cellular functions other than keratin metabolism are also affected.

    Notizen: Human keratinocytes from small skin specimens were grown on mouse 3T3 cell feeder layers into epidermal sheets free from Langerhans cells and MHC class II antigen. These were found to be suitable for the permanent coverage of wounds when used as autografts or allografts. We report here the ultrastructural differentiation of this cultured epidermis after grafting onto autologous or allogeneic recipients. The cultured epidermis was a thin but multilayered Malpighian epithelium composed of keratinocytes at different stages of differentiation. The dermo-epidermal basement membrane was newly synthesized during the first few days following transplantation onto de-epidermized wounds. The analysis of keratins and examination of various keratinocyte membrane antigens by immunofluorescence indicated that full terminal epithelial differentiation was only achieved after in vivo transplantation of the cultured epidermis. Langerhans cells, absent in cultures, progressively colonized the grafts, while melanocytes, not detectable in sections of the cultures, were identified among the keratinocytes 2 weeks after grafting.

    Notizen: In many benign and malignant skin disorders, lymphocytes and associated monocytic cells predominate among the various types of cells infiltrating the dermis and/or the epidermis.Immunohistological techniques utilized for the purpose of combining immunological parameters and morphological criteria have been reviewed. The rosetting techniques, enzyme histochemistry and immunohistological techniques using polyclonal and monoclonal antibodies were reported for light microscopy. In the field of immunoelectron microscopy, immunoperoxidase labelling and the immunogold technique were also subject to discussion. These techniques may be supplemented by various methods of cell biology (obtaining enriched cell suspensions of infiltrate cell subsets). The possiblity of quantitative studies was considered.Results obtained in cutaneous pathology using these techniques were reviewed and discussed. Among others, human skin tuberculin lesions and DNCB skin tests represent models of delayed hypersensitivity.Infiltrates in the following conditions were presented: non-tumoral lesions (lichen planus, erythema multiform, erythema fixum, GVH and allograft rejection, cutaneous sarcoidosis and other infiammatory infiltrates); in lymphomas and related conditions; in tumoral lesions (warts and papillomas, peritumoral infiltrates around epidermal neoplasias).Finally Langerhans cells in infiammatory dermatosis, in skin tumours and in histiocytosis X were considered.
