    Notizen: Abstract We have identified two central neurocytomas which contained cells co-expressing glial fibrillary acidic protein and synaptophysin defined by double-label immunostaining. Dual-positive cells were mostly polygonal in shape and with a morphological appearence similar to that of reactive astrocytes. This distinct morphology could be used to distinguish cells expressing glial fibrillary acidic protein from cells with round and clear cytoplasm which did not express glial fibrillary acidic protein and which composed the majority of the tumor. Samples containing polygonal cells were selected for electron microscopy from toluidine blue-stained semithin sections. Ultrastructural findings were similar in both neurocytomas, with both being composed predominantly of round cells with clear cytoplasm corresponding to the clear cells identified by light microscopy. Dense-core vesicles and clear vesicles were frequently observed in the cell processes. Apart from these clear cells, polygonal cells with electron-dense cytoplasm were noted. Paralleling the results of double immunostaining, these polygonal cells contained both dense-core vesicles and intermediate, presumably glial filaments. Microtubules and lipofuscin granules were also observed. These results suggest that cells expressing glial fibrillary acidic protein in central neurocytoma include tumor cells with both neuronal and glial characteristics.

    Notizen: Abstract  We examined the in vivo effect of paraquat on the cell cycle in rat liver and lung tissues and the protective effect of tungsten (a xanthine oxidase inhibitor) on paraquat toxicity. The bromodeoxy- uridine/propidium iodide double-staining method and flow cytometry were used for cell cycle assessment. Wistar rats were fed a standard diet or a tungsten-enriched diet were injected intravenously with 20 mg/kg paraquat, while uninjected rats served as controls. At 1, 3, and 5 days after paraquat injection, the liver and lungs were removed for examination following in vivo labeling with 20 mg/kg bromo- deoxyuridine for 1 h. Liver and lung cells were isolated and incubated with an anti-bromodeoxyuridine antibody and with propidium iodide for DNA staining. Flow cytometry showed that the S-phase cell populations in the liver and lungs of paraquat-injected rats fed a standard diet were increased markedly on days 1 and 3 after injection compared with the control levels. However, on day 5 the liver cells had nearly returned to normal, while the S-phase population remained high in the lungs. In contrast, the S-phase cell populations of liver and lung tissue showed no increase after paraquat injection in rats fed a tungsten-enriched diet. These findings suggest that paraquat-induced cytotoxicity is more prolonged in the lungs than in the liver. In addition, paraquat toxicity appears to be mediated by xanthine oxidase and xanthine oxidase inhibitors may be useful as an antidote.

    Notizen: Summary Antineoplastic effects of interferons (IFNs) on brain tumors have often been reported in the literature, however, so far as we know, there are no reports of the study on the antineoplastic effect of IFNs (α, β, and γ) labelled with fluorescein isothiocyanate (FITC) using flow cytometry (FCM). Three established glioma cell lines and 11 cultured cells of brain tumor from surgical specimens were exposed to IFN-α, β, and γ at the concentrations of 102–105 IU/ml for 24 h, respectively. Using FCM, the viability of the cells was evaluated with fluorescein diacetate stain and the cell cycle was analyzed from the DNA-histogram with propidium iodide stain. Furthermore, FITC-labelled IFN-α, β and γ were also contacted with each cell to calculate respective positive cells. The viability decreased about 60% on day 1 and day 3, indicating the effect of IFN-α and β on U373MG cells and on some cultured glioma cells from surgical materials, whereas, IFN-γ had no effects. Antineoplastic effect of each IFN well correlated with FITC-positive rates, demonstrating S phase block in the cell cycle. IFN-γ had no antineoplastic effects, whereas IFN-α and β showed antineoplastic effects, which fact suggested that IFN-γ receptor be different from those of IFN-α and β. The method of FITC-labelling for IFNs with the aid of FCM has the advantages as follows: 1) Antineoplasticity of IFN can be simply evaluated with FCM; 2) It is easy to analyze the action mechanism of IFN; 3) Information on the receptor is obtainable; and 4) Sensitivity can be evaluated prior to administration of IFN, suggesting possibilities of clinical application of this method.

    Notizen: Summary Flow cytometry (FCM) has been used to evaluate not only the malignancy of tumor cells but also the effects of chemotherapy. Here a new application of FCM for selecting the best antineoplastic agent in the chemotherapy for brain tumors is reported. Through our preliminary study using established brain tumor cell lines, the system for this sensitivity test was developed. Antineoplastic agents were placed in contact with monolayer-cultured cells; then cell viability and changes in the DNA histogram were analyzed by FCM. Cell viabilities were measured with the fluorescein diacetate (FDA) staining method, and the DNA histogram was analyzed by the propidium iodide (PI) staining method. The best antineoplastic agent was determined based on changes in cell viability and cell cycle. In other words, when markedly decreased viability as compared with that of the control, is measured by FCM, then the agents can be considered to have had a cytocydal effect on the tumor cells, and thus the sensitivity of the agents is able to be evaluated. If the viability of the tumor cell is observed to be similar to that of the control, the cytostatic effects of the agents are able to be evaluated only if a marked change is observed in the DNA histogram. After the preliminary study, this system was applied clinically to malignant brain tumor cases, resulting in success in selecting the best antineoplastic agent for each individual case. Our sensitivity test using this FCM established in vitro system has much potential value for clinical use.

    Notizen: Abstract To assess the effect of 1-(4-amino-2-methyl-5-pyrimidil)methyl-3-(2-chloroethyl)3-nitrosourea hydrochloride (ACNU) on human glioma, we performed cell-cycle-specific cell sorting followed by electron microscopic observation. Cells sorted from an accumulated SG2M peak caused by treatment with 10μg/ml ACNU showed a number of cells similar to S-phase cells in the untreated samples, having no microvilli. Cell organelles were intact, and no ultrastructural evidence for cell injury was observed. In contrast, many cells sorted from nonstagnated G0G1 and SG2M peaks in the samples treated with 100μg/ml ACNU showed mitochondrial swelling that suggested cell necrosis.

    Notizen: Abstract The numerous pathology reports on craniopharyngioma describe the classical features of squamous epithelium and keratin. Relatively little attention has been paid to its stroma and the surrounding tissue. We focused our observation on these two areas in a retrospective study of craniopharyngiomas in 36 operative cases and 5 autopsy cases. The tumor interdigitated with the brain and pituitary gland. At the interface, the brain was markedly gliotic with Rosenthal fibers simulating pilocytic astrocytoma. The tumor stroma had reactive changes and commonly showed cyst formation and calcification.
