Notizen: We have developed a Ti:Sa autoscan laser spectrometer and used it to perform a detailed investigation of individual line profiles in the ν1+3ν3 band of 12C2H2. Pressure self-broadening effects were measured for most lines with J″ values ranging from 0 to 22, and for pressures between 10 and 600 Torr. The high-pressure data points (p〉250 Torr) are fitted satisfactorily by a Voigt profile. Line narrowing, characterized by an underestimation of the Lorentz contribution to the Voigt profile, was observed for the low pressures range (p〈150 Torr), indicating the influence of molecular confinement. The two usual limit models (soft and hard collisions) were fitted to the observed line shapes in order to extract more precise information concerning the pressure broadening. The present results confirm the absence of strong vibrational dependence in the self-broadening coefficients. Absolute line intensities are also determined in the fitting procedure. No significant differences were found for the two pressure domains if the appropriate model in each range is used. © 1999 American Institute of Physics.

Notizen: We have recorded the spectrum of the ν1+3ν3 band in 12C2H2, around 12676 cm−1, using an autoscan Ti:Sa spectrometer. Pressure-dependent line positions were determined from an absorption spectrum recorded using a long White-type multiple-pass cell. An optoacoustic cell with a constant, low acetylene pressure allowed reference line position wavenumbers to be measured simultaneously. In different experiments, the multiple-pass cell was filled with increasing acetylene pressures. Self-collisional lineshifts were determined for more than 30 P and R lines in the band. Line wavenumbers for the stronger lines were separately measured from an optoacoustic spectrum calibrated using the results of a previous investigation in the literature [X. Zhan and L. Halonen, J. Mol. Spectrosc. 160, 464 (1993)] and corrected for their pressure dependence using the presently determined self-collisional lineshifts. Lineshift parameters that are significantly different from the overall behavior in the band are unexpectedly observed for the R(17) and P(19) lines. This result is explained in terms of a different intermolecular behavior of the molecule in the upper J=18 rotation level, attributed to an intramolecular Coriolis-type coupling with a nearby state tentatively assigned as containing important excitation in ν5, the cis-bend mode. © 1999 American Institute of Physics.

Notizen: We have performed the rovibrational analysis of the absorption spectrum of 12C2D2 between 5150 and 8000 cm−1, recorded by Fourier transform absorption spectroscopy, and between 12 800 and 16 600 cm−1, recorded by intracavity laser absorption spectroscopy. Respectively 10 and 9 bands are reported for the first time in each range. Improved or new rovibrational parameters were obtained for 34 vibrational levels altogether. The vibrational energies we obtained, together with those reported in the literature, were taken into account to model the vibrational energy pattern in 12C2D2(X˜ 1Σg+). The analysis was performed in successive steps, inferring each time suitable parameters. The 44/55, 11/33, 12/33, and 1/244 quartic order anharmonic resonances were introduced during the procedure. They altogether define vibrational clusters which are characterized by only two dynamical constants of motion, Ns=V1+V2+V3 and k=l4+l5. © 1998 American Institute of Physics.

Notizen: We studied the sequence of several metabolic reactions, representative for oxidative damage and protection, in primary leaves of Phaseolus vulgaris (cv. Limburgse vroege) as a function of root assimilation of a toxic sublethal Cu concentration (630 μM). A transient increase of products of membrane peroxidation was observed in the primary leaves during the period of Cu uptake. This rise was mainly due to the oxidizing properties of copper itself and not to a stimulation of the lipoxygenase (EC 1.13.11.12) activity. In our experimental conditions, membrane lipid peroxidation and K+-leakage were not directly related; during at least three days after Cu application to the roots, when products of lipid peroxidation were already detected in the leaf, permeability of the cytoplasmic membrane for K+ was improved. However, Cu stimulated the capacity of catalase (EC 1.11.1.6) and ascorbate peroxidase (EC 1.11.1.11). These enzymes protect the tissue against oxidative stress since at least the hydrogen peroxide content was significantly reduced. Superoxide dismutase (EC 1.15.1.1) was not involved in this defense mechanism.

Notizen: Abstract  Objective. Comparison of clinically observed neurologic long tract signs in a heterogeneous group of patients with rheumatoid arthritis (RA), with morphologic abnormalities of the cervical spine as depicted on radiographs and magnetic resonance (MR) images. Design. The patients were prospectively assigned to one of three classes on the basis of their neurologic status. Lateral cervical spine radiographs and sagittal T1-weighted and gradient echo images were performed. The qualitative MR features evaluated were erosion of the dens and atlas, brain stem compression, subarachnoid space encroachment, pannus around the dens, appearance of the fat body caudal to the clivus, and the signal intensity of the pannus. The quantitative imaging parameters were the cervicomedullary angle and the distance of the dens to the line of McRae. Patients. Sixty-three consecutive patients with RA and subjective symptoms, especially neck or occipital pain, and/or clinical objective signs consistent with a compromised cervical cord were included in this study. Results and conclusions. Damage documented with radiographs and MR imaging in patients with RA is often severe, even in those without neurologic signs (class 1). None of the abnormalities confined to the atlantoaxial level correlated significantly with neurologic classification. Subarachnoid space encroachment anywhere in the entire cervical spine did correlate significantly with neurologic classification.

Notizen: Abstract  Objective. To analyse to what extent MR imaging based decisions were correct in determining the surgical procedure in patients with osteosarcoma of the distal femur. Design. We compared the findings on MR imaging that determined the surgical strategy with the actual surgical findings or histopathological specimen. We assessed to what extent MR images could be used to determine the surgical procedure in patients with osteosarcoma. Patients. Between 1985 and 1992, 34 patients with an osteosarcoma were treated. Two patients had a low-grade osteosarcoma. Thirty-two patients with a high-grade osteosarcoma of the distal femur (17 stage IIB, 15 stage III) were included in this study. Surgical options consisted of either limb salvage surgery or ablative surgery, which included the Van Nes-Borggreve rotationplasty. Limb salvage surgery consisted of extra-articular or transarticular resection, followed by reconstruction. Surgery was planned depending on the local extent of the tumour as depicted on the MR studies, coupled with data from the biopsy, age, patient compliance and histological grade. Follow-up was available in all patients. Nine of 17 patients graded as IIB were alive with an average follow-up of 68 months (range 46–109 months), of whom one has metastases. No local recurrence was encountered. Results. If no tumour involvement on MR imaging was found and this was used as a determining factor, this proved to be correct at subsequent analysis. We found that in eight of 11 cases, when MR images suggested a close relationship between tumour and nerve, an oncologically safe plane could be achieved during surgery. In three, a free plane could not be accomplished, as confirmed at histopathological examination. Hence, when nerve involvement was equivocal on MR imaging we found it valuable to reassess nerve involvement during surgery and reconsider limb salvage surgery. When the decisive factor was the vascular involvement and tumour extension was read as equivocal, it was possible to obtain a oncologically safe plane in six of 13 cases. When comprehensive tumour involvement of any structure was noted pre-operatively, it proved to be correct at histopathological examination, except for one case of assumed vascular involvement that proved to be free. Conclusions. If no tumour involvement of a structure was found on MR imaging and this was used as a determining factor, this proved to be correct. When nerve involvement was equivocal on MR imaging we found it valuable to reassess nerve involvement during surgery and reconsider limb salvage surgery. Extensive tumour involvement of any structure, as shown by MR imaging, could be used correctly as a decisive argument in planning a surgical procedure.

Notizen: Abstract  Objective. To identify an optimal pulse sequence for in vitro imaging of hyaline cartilage at 0.5 T. Materials and methods. Twelve holes of varying diameter and depth were drilled in cartilage of two pig knees. These were submerged in saline and scanned with a 0.5-T MR system. Sixteen T1-weighted gradient echo (GE), two T2-weighted GE, and 16 fast spin echo sequences were used, by varying repetition time (TR), echo time (TE), flip angle (FA), echo train length, profile order, and by use of fat saturation. Contrast-to-noise ratios (CNR) of cartilage versus saline solution and cartilage versus subchondral bone were measured. Cartilaginous lesions were evaluated separately by three independent observers. Interobserver variability and correlation between the quantitative and qualitative analyses were calculated. Results. The mean CNRs of two specimens of cartilage versus saline solution ranged from 6.3 (±2.1) to 27.7 (±2.5), and those of cartilage versus subchondral bone from 0.3 (±0.2) to 22.5 (±1.4). The highest CNR was obtained with a T1-weighted spoiled 3D-GE technique (TR 65 ms, TE 11.5 ms, FA 45°). The number of lesions observed per sequence varied from 35 to 69. Observer agreement was fair to good. The T1-weighted spoiled GE sequences with a TR of 65 ms, TE of 11.5 ms and FA of 30° and 45° were significantly superior to the other 34 sequences in the qualitative analysis. Conclusion. T1-weighted spoiled 3D-GE sequences with a TR of 65 ms, a TE of 11.5 ms, and a FA of 30–45° were found to be optimal for in vitro imaging of cartilage at 0.5 T.

Notizen: Abstract Thirteen healthy subjects were subjected to tryptophan (TRP) depletion, lysine (LYS) depletion, and a placebo condition in a double blind cross-over study. The aim of the study was to test the specificity of psychological effects induced by TRP depletion. Subjects ingested a 100 g amino acid mixture with or without TRP or LYS. Six hours later, plasma TRP levels had decreased by 77% in the TRP depletion test and LYS levels by 51% in the LYS depletion condition. After 6 h of TRP depletion, subjects reported significantly more tiredness and lowering of mood, compared to subjects in the placebo group, and memory performance declined. After 6 h of LYS depletion, no significant differences in mood and memory compared to placebo were found. We conclude that the effects of TRP depletion on mood and memory are specific for the depletion of TRP and are not caused by the depletion of an amino acid per se. This supports the hypothesis that TRP depletion affects brain serotonin metabolism and not only brain protein metabolism in general.
