    Notes: The formation of a nanoicosahedral phase in the diameter range below 20 nm from the melt-spun Zr70Pd70 binary glassy alloy was confirmed, and the kinetics of the precipitation upon isothermal annealing in the supercooled liquid region was examined by differential scanning calorimetry. Based on the kinetic analysis, it is clarified that the precipitation in the supercooled liquid region takes place by a diffusion-controlled growth with increasing nucleation rate. The Arrhenius plot between effective time lag, τ, of nucleation and isothermal annealing temperature yields a single linear relation, in which the activation energy for nucleation is evaluated to be 267 kJ mol−1. The activation energy in the present alloy is much lower than that of the Zr65Al7.5Ni10Cu7.5Pd10 alloy, which is due to the difference in the magnitude as well as the number of atoms for the rearrangements in the nucleation stage. It is concluded that the formation of a nanoscale icosahedral quasicrystalline phase is attributed to the transformation mode of an increase of nucleation rate, which is different from that of Zr65Al7.5Ni10Cu7.5Ag10 quasicrystal arising from an interfacial controlled growth with a steady-state nucleation rate. The formation of a nanoicosahedral phase in the Zr70Pd30 binary glassy alloy implies the existence of the icosahedral short-range order in the glassy state. © 2000 American Institute of Physics.

    Notes: We present a comparative study, down to the single-molecule level, of the linewidth and multiplet structure of the optical transitions of 2.3,8.9-dibenzanthanthrene (DBATT) in shock-frozen solutions of n-decane, n-undecane, and n-tetradecane. The n-alkane matrices show weak structural ordering and distinct incorporations of the DBATT molecules have been recognized. The order does not increase, nor does the ensemble optical linewidth decrease going from bulk to sub-volumes of less than 1 μm3. The relation between lateral position, orientation of the transition moment, and transition frequency has been investigated using single-molecule fluorescence-excitation and polarization spectroscopy. The distribution of the orientation is found not to depend on frequency and vice versa. Structural ordering on a microscopic scale is found not to be decisive as regards the linewidth and multiplet structure. © 2002 American Institute of Physics.

    Notes: An icosahedral quasicrystalline phase was confirmed as a primary precipitation phase in melt-spun Zr70Ni10M20 (M=Pd, Au and Pt) ternary metallic glasses with two-stage crystallization process. The onset temperature of the transformation from amorphous to icosahedral phase are 687 K for Pd, 754 K for Au and 783 K for Pt at a heating rate of 0.67 K s−1. The size of the icosahedral particles is in the range of 5–20 nm. The second crystallization reaction results in the formation of Zr2Ni+Zr2Pd and Zr2Ni+ZrPt phases in the Zr–Ni–Pd and Zr–Ni–Pt alloys, respectively, and Zr3Au phase in the Zr–Ni–Au alloy through a single exothermic reaction. The formation of nanoscale icosahedral phase indicates the possibility that icosahedral short-range order exists in the glassy state. © 2000 American Institute of Physics.

Based on the kinetic analysis, it is clarified that the precipitation in the supercooled liquid region takes place by an interfacial controlled growth with a nearly steady-state nucleation rate. The Arrhenius plot between effective time lag, τ, of nucleation and isothermal annealing temperature yields a single linear relation, in which the activation energy for nucleation is evaluated to be 366 kJ/mol. It is concluded that the transformation of amorphous to quasicrystal proceeds by a homogeneous nucleation mode, which is different from those of Pd–U–Si and Al–Cu–V quasicrystals arising from an inhomogeneous distribution of quenched-in nuclei. © 1999 American Institute of Physics.

    Notes: Mannan-binding lectin (MBL), L-ficolin and H-ficolin are pattern recognition molecules of the innate immune system. We investigated the ability of these molecules to bind to different serotypes and noncapsulated variants of Streptococcus pneumonia and Staphylococcus aureus. We found that MBL binds to noncapsulated S. aureus strain (Wood) but not any of the examined S. pneumoniae serotypes. L-ficolin binds to some capsulated S. pneumoniae serotypes (11A, 11D and 11F) as well as some capsulated S. aureus serotypes (Type-1, -8, -9, -11 and -12). H-ficolin does not bind to any of the examined S. pneumoniae and S. aureus serotypes included in this study but did bind to a strain of Aerococcus viridans. When bound to bacteria, MBL and H-ficolin initiated activation of complement factor C4, whereas L-ficolin did not. During this study, quantitative assays for the three proteins were developed and the concentration in 97 plasma samples were determined and the median values were estimated at 0.8 μg of MBL/ml, 3.3 μg of L-ficolin/ml and 18.4 μg of H-ficolin/ml, respectively.

    Notes: Three pathways are recognized in the complement activation cascade. The aim of our study was to elucidate immunohistologically which complement pathway is associated with the activation in IgA glomerulonephritis (GN) and the relation of IgA subclass to the complement activation. Immunohistological staining was performed on biopsied renal specimens from 36 patients with IgA GN, 10 with systemic lupus erythematosus (SLE) and 16 with other glomerulonephritides using polyclonal antibodies of IgG, IgA, IgM, C3c, C4, C1q and monoclonal antibodies of IgA1, IgA2, mannose-binding lectin (MBL) and MBL-associated serine protease-1 (MASP-1). Mesangial deposits of IgA1, IgA2, C3c, C4, MBL and MASP-1 were detected in 19 of the 36 patients with IgA GN, and IgA2 and MBL/MASP-1 were colocalized in the mesangium in these 19 patients. The remaining 17 patients showed mesangial deposition of IgA1 alone. Twelve of these 17 patients presented mesangial deposition of C3c without deposition of C4, MBL and MASP-1. No deposition of C1q was evident in IgA GN patients. Three of the 10 SLE patients showed glomerular deposition of MBL and MASP-1 without deposition of IgA2. No patient with other glomerulonephritides showed glomerular deposition of IgA1, IgA2, MBL and MASP-1. There was no correlation in clinical and pathological indicators between IgA2-positive and IgA2-negative patients with IgA GN. In conclusion, alternative pathway-mediated complement activation is associated in patients with mesangial deposition of IgA1 alone in IgA GN. In those with the deposition of both IgA1 and IgA2, both alternative and lectin pathways are activated, and mesangial deposition of IgA2 is associated with the lectin pathway-mediated complement activation in IgA GN.

    Notes: To elucidate the physiological role of Fyn, we analysed the properties of synaptic transmission and synaptic plasticity in hippocampal slices of mice overexpressing either wild-type Fyn (w-Fyn) or its constitutively active mutant (m-Fyn). These fyn-transgenes were driven by the calcium/calmodulin-dependent protein kinase IIα promoter which turned on in the forebrain neurons including hippocampal pyramidal cells and in late neural development. In the hippocampal slices expressing m-Fyn the paired-pulse facilitation was reduced and the basal synaptic transmission was enhanced. A weak theta-burst stimulation, which was subthreshold for the induction of long-term potentiation (LTP) in control slices, elicited LTP in CA1 region of the slices expressing m-Fyn. When a relatively strong stimulation was applied, the magnitude of LTP in m-Fyn slices was similar to that in control slices. By contrast, the basal synaptic transmission and the threshold for the induction of LTP were not altered in the slices overexpressing wild-type Fyn. To examine the effect of expression of m-Fyn on GABAergic inhibitory system, we applied bicuculline, a GABAA receptor blocker, to the hippocampal slices. The ability of bicuculline to enhance excitatory postsynaptic potentials was attenuated in slices expressing m-Fyn, suggesting that the overexpression of m-Fyn reduced the GABAergic inhibition. The enhancement of synaptic transmission and the reduction of GABAergic inhibition may contribute to the enhanced seizure susceptibility in the mice expressing m-Fyn. Thus, these results suggest that regulation of Fyn tyrosine kinase activity is important for both synaptic transmission and plasticity.
