    Notizen: Summary The influences of the snake venom taipoxin on the cholinergic synapses of the electric organ ofTorpedo marmorata were investigated. 1. In ultrathin sections presynaptic membrane indentations are noted under the influence of taipoxin/NaCl. 2. The presynaptic membranes often show small interruptions. 3. In taipoxin/horseradish peroxidase incubated organ pieces typical omega-shaped indentations are found, but on the postsynaptic side. 4. The vesicle density decreased under the influence of the taipoxin in contrast to untreated cholinergic synapses. 5. In freeze etched preparations no membrane indentations and interruptions are observed, but only a small vesicle density was seen.

    Notizen: Summary The alkaloid muscarin was isolated from fly agaric. The analyses of crystallized salts (chloride, tetra-chloroaurate,Reinecke's salt) led to C9H20O2N+ as the simplest formula. The presence of aldehyde and tri-methyl-ammonium groups, which had been assumed hitherto, could not be proved. The quaternary nature of the nitrogen atom was shown. Pharmacological tests on frog heart, rabbit gut and cats show the purely peripheral parasympathomimetic effect of the smallest doses.

    Notizen: Summary All synthetic isomers of (+)-muscarine, the racemic forms of epi-muscarine, allo-muscarine, and epiallo-muscarine are at least 100 times less potent on blood pressure of cats and isolated frog hearts than the natural alkaloid. Only muscarones are more potent and exhibit strong nicotinic action on the frog rectus and in blocking ganglionic and neuro-muscular transmission in the cat. They closely resemble acetylcholine in their structure and action. The activity of the isomers depends on the position of the constituents on the tetrahydrofurane ring. Steric hindrance of the hydroxy or carbonylgroup and of etheroxygen plays an important part in the contact with the cholinergic receptor. As nor-muscarine without the quarternary nitrogen is inactive, at least 3 different points of contact have to be considered. This is stressed by the fact that muscarine is highly stereospecific in its action, whereas muscarone is not.
