    Notizen: A plasma vortex street configuration can evolve when two velocity and one magnetic shear layer interact strongly. A study of the interaction between two- and three-dimensional plasma modes and a mean sheared magnetic field is undertaken using a three-dimensional magnetohydrodynamic spectral Galerkin computation. The initial state is a simple magnetic shear in a plane perpendicular to the plasma velocity shear plane. In a very weak magnetic field, secondary instabilities (three-dimensional modes), expressed by the kinking of vortex tubes, lead to plasma flow along and around the axes of the vortex cores, creating characteristic patterns of kinetic helicity and linkages between vortex filaments. Three-dimensionality leads to the vortex breakdown process. A strong sheared magnetic field inhibits the kinking of vortex tubes, maintaining two-dimensionality. This inhibits vortex breakdown over long dynamical times. There is an anticorrelation in time between linkage indices of the vortex filament (related to kinetic helicity), suggesting that the ellipticity axes of the vortex cores along the street undergo a global inphase evolution. This anticorrelation has a dynamical interpretation. It extends to a relaxing plasma in the Navier–Stokes flow notion that helical regions of opposite helicities interact and screen each other off so that the global helicity remains bounded.

    Notizen: Coarse-grained information from a hybrid simulation show that previously developed quasifluid equations of state capture some aspects of proton cyclotron resonant interactions for parallel propagation in a low β plasma. Direct kinetic information is used as a closure for the information exchanged with the higher-order moment quantities. The coarse-graining procedure involves averaging over many proton-inertial lengths and looking at long time scales associated with the wave envelopes. By use of the coarse-graining prior to statistical analysis, the anticorrelations predicted by the equations of state are very high for both the single resonant wave case and for a broadband spectrum centered on a resonant wave. These anticorrelations are also consistent with what one expects from a single particle orbit analysis. A similar analysis, but done without the prior averaging procedures, shows no relevant correlations, and no simple dynamics emerges from the kinetic data in this case. A comparison is made with a model based on quasilinear theory. These results suggest that, in addition to simply taking velocity moments of particle distributions, the proper averaging of kinetic information over space and time scales lying below the range accessible to a fluid model can be essential to its success. © 1998 American Institute of Physics.

    Notizen: A method for constructing closure relations based on the invariants of the tensors representing nonequilibrium thermodynamic forcing within the plasma is presented. This approach leads to closure relations that describe all higher-order forcing effects contained within the continuum description. Nonlinear convective-momentum transport and nonlinear momentum-exchange operators are constructed as applications of the method. Closure is achieved by relating the pressure tensor to invariants of the rate of strain tensor, and the momentum-exchange operator to invariants of the gradient of magnetic field tensor. These operators lead to positive definite viscous and Joule entropy production and enhance high wave number dissipative couplings over all other dissipative couplings. The nonlinear dissipative action is localized in physical space, where velocity and magnetic gradients are large, while allowing nearly ideal behavior elsewhere. The operators are computationally tested against the standard magnetohydrodynamic (MHD) operators using three-dimensional configurations that lead to vortex street formation and magnetic reconnection. The nonlinear operators allow greater spatial structure and have flatter modal energy spectra than the standard MHD dissipation operators. Closures that describe the plasma response to nonequilibrium thermodynamic forcing of all orders can be constructed using this approach. © 1995 American Institute of Physics.

    Notizen: A laser diode based probing system for the interferometric detection of charge density modulations and temperature changes which accompany electric signals in integrated circuits has been developed. Measurements are performed through the substrate of the device under test. Compared to previously introduced measuring schemes the system offers the advantage of a versatile and inexpensive positioning system of the probing and the reference laser beam, noise reduction due to the use of heterodyn techniques, and the absence of polarizing elements. The temporal resolution is limited by the width of the laser diode pulse to 500 ps. The probe beam can be focused to a diameter of 1.5 μm through a 400-μm-thick silicon substrate which determines the spatial resolution of our measurement system. It should be stressed that probing through the substrate provides the only access to internal measurement nodes for a growing number of highly integrated circuits the front side of which is masked by multilevel wiring, bonding wires, or lead frames.

    Notizen: Abstract: A peptide corresponding to position 32-47 in tyrosine hydroxylase was synthesized (TH-16) and polyclonal antibodies against this peptide were raised in rabbits (anti-TH-16). The effects of anti-TH-16 on modulation of tyrosine hydroxylase activity were investigated. Anti-TH-16 enhanced the enzymatic activity in a concentration-dependent manner, and the antigen TH-16 inhibited the stimulatory activity of the antiserum in a concentration-dependent manner. The activated enzyme had a lower Kmapp for the cofactor 2-amino-4-hydroxy-6-methyl-5,6,7,8-tetrahydropterin and a higher Vmax app than the nonactivated enzyme. Anti-TH-16 was characterized further by its ability to immunoprecipitate the enzyme activity by labeling tyrosine hydroxylase after Western blotting and by immunohistochemical labeling of cate-cholaminergic neurons. Anti-TH-16 did not block activation of tyrosine hydroxylase by phosphorylation catalyzed by cyclic AMP-dependent protein kinase. Exposure of the enzyme to anti-TH-16 and subsequent phosphorylation of the enzyme resulted in a greater activation of the enzyme than the sum of activation produced by these two treatments separately. However, the activation was less than additive when the enzyme was first phosphorylated and subsequently exposed to anti-TH-16. The present study demonstrates the utility of anti-TH-16 in investigating the molecular aspects of the enzyme activation.

    Notizen: Abstract: De novo l-DOPA biosynthesis was studied in stably transfected AtT-20 cells expressing wild-type- or [Leu40]-recombinant tyrosine hydroxylase (rTH). Basal rates of DOPA accumulation were much higher by cells expressing rTH in which Leu was substituted for Ser40 (S40L-rTH) than by those expressing wild-type rTH (WT-rTH). Treatment of WT-rTH cells with forskolin produced an increase in DOPA accumulation and a concomitant increase in WT-rTH phospho-Ser40 content, whereas DOPA production by cells expressing S40L-rTH was entirely unaffected by forskolin. After forskolin treatment of 32Pi-prelabeled cells, WT-rTH was phosphorylated at Ser8, Ser19, Ser31, and Ser40, whereas 32P incorporation into S40L-rTH was restricted to Ser8, Ser19, and Ser31. Relatively prolonged treatment of AtT-20 cells expressing WT-rTH with either a depolarizing agent (elevated potassium) or a phosphatase inhibitor (okadaic acid) increased DOPA production and increased the phosphorylation state of Ser40; but, unlike forskolin, these treatments also increased DOPA production by cells expressing S40L-rTH. Thus, the present studies demonstrate that Ser40 phosphorylation mediates forskolin-induced increases in DOPA biosynthesis directly but that mechanisms other than Ser40 phosphorylation can mediate the increases in DOPA biosynthesis produced either by depolarization or by protein phosphatase inhibition.

    Notizen: Abstract: A synthetic peptide corresponding to residues 32–47 of rat tyrosine hydroxylase (TH) was phosphorylated by protein kinase A at Ser40 and used to generate antibodies in rabbits. Reactivity of the anti-pTH32–47 antibodies with phospho- and dephospho-Ser40 forms of TH protein and peptide TH32–47 was compared with reactivity of antibodies to nonphosphorylated peptide and to native TH protein. In antibody-capture ELISAs, anti-pTH32–47 was more reactive with the phospho-TH than with the dephospho-TH forms. Conversely, antibodies against the nonphosphorylated peptide reacted preferentially with the dephospho-TH forms. In western blots, labeling of the ∼60-kDa TH band by anti-pTH32–47 was readily detectable in lanes containing protein kinase A-phosphorylated native TH at 10–100 ng/lane. In blots of supernatants prepared from striatal synaptosomes, addition of a phosphatase inhibitor was necessary to discern labeling of the TH band with anti-pTH32–47. Similarly, anti-pTH32–47 failed to immunoprecipitate TH activity from supernatants prepared from untreated tissues, whereas prior treatment with either 8-bromoadenosine 3′,5′-cyclic monophosphate or forskolin enabled removal of TH activity by anti-pTH32–47. Lastly, in immunohistochemical studies, anti-pTH32–47 selectively labeled catecholaminergic cells in tissue sections from perfusion-fixed rat brain.

    Notizen: Abstract: The effects of depolarization by elevated potassium concentrations were studied in PC12 cells and in stably transfected AtT-20 cells expressing wild-type or [Leu19]-recombinant tyrosine hydroxylase (rTH). Changes in the phosphorylation states of Ser19 and Ser40 in tyrosine hydroxylase (TH) were determined immunochemically using antibodies specific for the phosphorylated state of each site and compared with changes in TH activity in PC12 cell lysates and with changes in l-DOPA biosynthesis rates in intact AtT-20 cells. Treatment of either PC12 cells or AtT-20 cells expressing wild-type rTH with elevated potassium produced a transient increase in the phosphorylation state of Ser19 (up to 0.7 mol of phosphate/mol of subunit) in concert with a more gradual and sustained increase in Ser40 phosphorylation. Elevated potassium treatment also increased TH activity in PC12 cell lysates, but these increases paralleled the temporal course of Ser40, as opposed to Ser19, phosphorylation. Similarly, increases in DOPA accumulation produced by elevated potassium in AtT-20 cells expressing wild-type rTH paralleled the increases in the phosphorylation state of Ser40 but not Ser19. Moreover, elevated potassium produced comparable increases in DOPA accumulation in AtT-20 cells expressing rTH in which Ser19 phosphorylation had been eliminated (by substitution of Leu for Ser19). Thus, depolarization-induced increases in the stoichiometry of Ser19 phosphorylation do not appear to influence directly the activity of TH in situ.
