    Notizen: : A unique biomarker, h-caldesmon, was identified and purified from bovine intestine smooth muscle. It was used to develop monoclonal antibodies (MAbs) for use in immunochemical assays to detect prohibited meat and bone meal (MBM) in animal feed. This biomarker with a molecular weight of 150 kDa was demonstrated by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) to be present in MBM samples that were obtained from different manufacturers. The presence of this biomarker in MBM and smooth muscle was also demonstrated by immunostaining with MAb 8B4 in Western blot assay. h-Caldesmon in intestinal smooth muscle was demonstrated to be stable after autoclaving at 130 °C for up to 1 h. Because MAb 8B4 was sensitive in detecting MBM in animal feed at 〉0.05%, it can be used in immunoassays for MBM detection.

    Notizen: : Histamine and other biogenic amines were evaluated in canned anchovies recalled by the U.S. Food and Drug Administration. In addition, bacteria were isolated from the products and identified to species. The recalled products were divided into 2 groups of high and low histamine, depending on the histamine contents as determined by the AOAC method. The high histamine group had the histamine contents 〉200 ppm, and 24 of the 30 cans analyzed belonged to this group. The most prevalent biogenic amine in this group was histamine followed by cadaverine. On the other hand, the low histamine group of 6 cans contained approximately 50 ppm histamine. The most prevalent biogenic amine found in this group of samples was cadaverine at levels 〉200 ppm. Other biogenic amines, such as putrescine, serotonin, and spermidine, were also detected in all the products, although at varied levels. Aerobic and anaerobic bacterial counts, if present in all of the recalled products, were below the detection limit of 102 colony-forming units (CFU)/g. Bacteria were recovered only after enrichment of the test samples. They were mostly halophilic bacteria. Bacillus spp. were most frequently identified, followed by Staphylococcus spp. However, these isolates produced negligible amounts of histamine in culture broth, indicating that they are not the contributors to histamine accumulation in the canned anchovies.

    Notizen: : Testing for Morganella morganii, a prolific histamine former, was carried out in fish and processing plant to determine its origin and contamination source using PCR assay coupled with Southern hybridization. M. morganii was mostly found in mackerel and sardines but rarely in albacore. It was most frequently present in the gill followed by the skin but rarely in the intestine and cavity. No M. morganii was found in the processing plant but was detected on the surfaces of conveyer belts and plastic totes during processing. The compiled results indicated that histamine-forming bacteria are endogenous to fish and pointed out the importance of sanitation in the processing plant to prevent cross-contamination.

    Notizen: : The effects of storage at 0,4,10, and 22°C for 0,1,3,5, and 9 d on the quality of yellowfin tuna fillets as determined by microbiological assessment, development of some biogenic amines, and sensory analysis were studied. Tuna fillets stored at 22 °C for 3 d, 10 °C for 5 d, and 4 °C for 9 d were rated unacceptable for consumption. Those stored at 22 °C for 3 d had total aerobic bacterial count of 〉 8 log10 CFU/g, a histamine-producing bacterial population of 7 log10 CFU/g, and 832 ppm of histamine, 35.8 ppm of putrescine, and 147 ppm of cadaverine. A comparison of the capillary electrophoresis, AOAC fluorometric method, and gas chromatography showed a very good correlation (r2 〉 0.99) among these 3 methods for histamine quantitation in tuna samples. Morganella morganii, Enterobacter agglomerans, Enterobacter intermedium, Pseudomonas fluorescens, Proteins vulgaris, and Serratia liquefaciens were the decarboxylase-positive bacterial species isolated by using the Niven's medium and identified during storage, which were responsible for histamine production in test tuna fillets.

    Notizen: Polyphenoloxidase (PPO) isolated from cuticle of Western Australian lobster (Panulirus cygnus) and Florida spiny lobster (Panulirus argus) catalyzed oxidation of catechol and DL-β-3,4-dihydroxyphenyla!anine (DL-DOPA). The PPO from Florida lobster showed a higher substrate affinity than that from Australian lobster. They both showed higher affinity for catechol than for DL-DOPA, and optimal pH stability at 7.0. The enzymes differed with respect to activation energy and thermal stability. Electrophoretic patterns using SDS-PAGE indicated PPO from Western Australian lobster had two isoforms while Florida spiny PPO had three isoforms. These results suggest variations in enzyme activity may contribute to differences in susceptibility to melanosis between the two species.

    Notizen: The effect of supercritical carbon dioxide on aerobic total plate count (TPC) of fresh-squeezed and quick-frozen single strength orange juice from Valencia oranges was studied. Orange juice was treated with SC CO2 at different temperatures (35° to 60°C) and pressures (8.3 to 33.1 MPa). SC CO2 treatment at 35°C reduced the TPC by 2 log cycles after 1 h treatment at 33 MPa. At 45°C and 33 MPa 2 log cycles reduction required 45 mins, and 15 mins treatment was sufficient at 60°C. D values of temperature control samples decreased as temperature increased, while that of supercritically treated juice decreased as pressure increased. D values at 35, 45 and 60°C of the supercritically treated juice at 33.1 MPa were calculated as 28, 22.6 and 12.7 min, respectively. The calculated values of z were 180°C, 167°C and 72°C at 8.3 MPa, 20.7 MPa and 33.1 MPa, respectively.

    Notizen: Thermolabile (TL) and thermostable (TS) pectinesterases (PE) were extracted from the pulp of fresh Valencia oranges by enzyme hydrolysis using a native pH and high ionic strength buffer system. Each form was separated individually by CM-Sephadex C-50, Phenyl Sepharose CL-4B and CM-Bio-Gel A chromatography. TLPE and TSPE exist as multiple forms which were shown to differ in their stability and effects on cloud loss in single strength orange juice.

    Notizen: This study evaluated seven existing watermelon rind pickle formulations (representing various soaking pretreatments including lime, brine and water) for their chemical, physical, sensory properties, safety, and suitability for industrial production. Titratable acidity (P = 0.0094), pH (P = 0.0042), color (P = 0.0153) and texture (P = 0.0373) were significantly different among various formulations; whereas, viscosity (P = 0.6603), total solids (P = 0.1175), and moisture (P = 0.2519) were not. Sensory data using a mixed gender consumer panel (n = 300, age range 18–65) indicated that overall, consumers preferred lime-soaked samples over brine or water pretreatments. Correlation existed between texture and crunchiness rating (r = 0.80). In addition, older consumers rated overall preference of watermelon rind pickles higher than younger adults (P = 0.0001). Six out of seven formulations used in this study are considered safe for industrial production with current processing times and temperatures (with pH below 4.0); however, one formulation with a final pH nearing 4.6 is considered not safe for current processing conditions.

    Notizen: Two phenoloxidase (PPO) isoforms were purified from Taiwanese black tiger shrimp (Penaeus monodon) purified via ammonium sulfate precipitation and high performance hydrophobic interaction chromatography on phenyl Sepharose CL-4B. Optimal DL-ã-3, 4-dihydroxyphenylalanine (D, L-DOPA) oxidation by these isoforms was observed to occur at pH 6.0, and at 45°C. The isoforms showed both mono- and di-PPO activities, and preferential medabolism of both monoand di-phenolic substrates lacking a carboxyl group either directly attached to the ring, or in the side chain moiety of the phenolic structure.
